Chickpea (Cicer arietinum L.) is cultivated mostly in the arid and semi‐arid regions of the world. Climate change will bring new production scenarios as the entire growing area in Indo–Pak subcontinent, major producing area of chickpea, is expected to undergo ecological change, warranting strategic planning for crop breeding and husbandry. Conventional breeding has produced several high‐yielding chickpea genotypes without exploiting its potential yield owing to a number of constraints. Among these, abiotic stresses include drought, salinity, water logging, high temperature and chilling frequently limit growth and productivity of chickpea. The genetic complexity of these abiotic stresses and lack of proper screening techniques and phenotyping techniques and genotype‐by‐environment interaction have further jeopardized the breeding programme of chickpea. Therefore, considering all dispiriting aspects of abiotic stresses, the scientists have to understand the knowledge gap involving the physiological, biochemical and molecular complex network of abiotic stresses mechanism. Above all emerging ‘omics’ approaches will lead the breeders to mine the ‘treasuring genes’ from wild donors and tailor a genotype harbouring ‘climate resilient’ genes to mitigate the challenges in chickpea production.  相似文献

Soaking—drying treatment of stored tomato seeds greatly reduced their deterioration under accelerated and natural ageing conditions. Hydration by moisture equilibration at 100% RH for 24 hours followed by drying-back was also very effective. Treatment of harvest-fresh seed was ineffective. The beneficial effects of the soaking—drying treatments were reflected in the productivity of the crop. Treatment with dilute solutions of sodium chloride (10?3 M) or sodium phosphate (dibasic, 10?4 M) proved better than water for field performance and productivity.  相似文献

Background

To identify plant genes involved in various key traits, QTL mapping is a powerful approach. This approach is based on the use of mapped molecular markers to identify genomic regions controlling quantitative traits followed by a fine mapping and eventually positional cloning of candidate genes. Mapping technologies using SNP markers are still rather expensive and not feasible in every laboratory. In contrast, microsatellite (also called SSR for Simple Sequence Repeat) markers are technologically less demanding and less costly for any laboratory interested in genetic mapping.

Results

In this study, we present the development and the characterization of a panel of 96 highly polymorphic SSR markers along the Arabidopsis thaliana genome allowing QTL mapping among accessions of the Versailles 24 core collection that covers a high percentage of the A. thaliana genetic diversity. These markers can be used for any QTL mapping analysis involving any of these accessions. We optimized the use of these markers in order to reveal polymorphism using standard PCR conditions and agarose gel electrophoresis. In addition, we showed that the use of only three of these markers allows differentiating all 24 accessions which makes this set of markers a powerful tool to control accession identity or any cross between any of these accessions.

Conclusion

The set of SSR markers developed in this study provides a simple and efficient tool for any laboratory focusing on QTL mapping in A. thaliana and a simple means to control seed stock or crosses between accessions.  相似文献

The initial ratios of neodymium-143 to neodymium-144 in kimberlites ranging in age between 90 x 10(6) to 1300 x 10(6) years from South Africa, India, and the United States are different from the corresponding ratios in the minerals of peridotite inclusions in the kimberlites but are identical to the ratios in the basaltic achondrite Juvinas at the times of emplacement of the respective kimberlite pipes. This correlation between the kimberlites and Juvinas, which represents the bulk chondritic earth in rare-earth elements, strongly indicates that the kimberlite's source in the mantle is chondritic in rare-earth elements and relatively primeval in composition.  相似文献

Previous studies have demonstrated that fish inhabiting polluted waterways often have an impaired stress response at the organismal level. Given the possible link between the organismal (i.e. cortisol) and cellular (i.e. heat shock proteins; hsp) stress responses, we conducted this study to examine the ability of rainbow trout to respond to a 2 h, +14 °C heat stress (HS) challenge following a 28 d, sub-chronic exposure to increased concentrations of cortisol (5 mg kg−1 b.w.), β-napthoflavone (bnf; 50 mg kg−1 b.w.), and a combination of both (mixture), through the diet (1.5% b.w. every 48 h). While control fish responded to the HS by significantly increasing components of their organismal (cortisol, glucose, and lactate) and cellular (hepatic hsp70 protein) stress responses 6 and 24 h post HS, cortisol-, bnf-, and mixture-fed fish had impaired stress responses at both levels of organization. Additionally, hepatic hsp70 levels were significantly reduced 6 h post HS in cortisol-fed fish. While bnf-fed fish had significantly higher EROD activity, cortisol potentiated EROD activity in the mixture-fed fish. Similarly, plasma cortisol concentrations in the mixture-fed fish were significantly lower relative to cortisol-fed fish. These data are the first to indicate that sub-chronically stressed fish can have impaired stress responses at both the organismal and cellular levels. These findings raise questions regarding: (a) the universal and simple applicability of biological indicators of stress in fish; (b) the possible functional relationship between these two levels of stress responses; and (c) the importance of hsps in the generalized stress response of the whole organism. Multiple chondritic meteorite fragments have been found in two sedimentary rock samples from an end-Permian bed at Graphite Peak in Antarctica. The Ni/Fe, Co/Ni, and P/Fe ratios in metal grains; the Fe/Mg and Mn/Fe ratios in olivine and pyroxene; and the chemistry of Fe-, Ni-, P-, and S-bearing oxide in the meteorite fragments are typical of CM-type chondritic meteorites. In one sample, the meteoritic fragments are accompanied by more abundant discrete metal grains, which are also found in an end-Permian bed at Meishan, southern China. We discuss the implications of this finding for a suggested global impact event at the Permian-Triassic boundary.  相似文献

Methotrexate coupled to maleylated bovine serum albumin was taken up efficiently through the "scavenger" receptors present on macrophages and led to selective killing of intracellular Leishmania mexicana amazonensis amastigotes in cultured hamster peritoneal macrophages. The drug conjugate was nearly 100 times as effective as free methotrexate in eliminating the intracellular parasites. Furthermore, in a model of experimental cutaneous leishmaniasis in hamsters, the drug conjugate brought about more than 90% reduction in the size of footpad lesions within 11 days. In contrast, the free drug at a similar concentration did not significantly affect lesion size. These studies demonstrate the potential of receptor-mediated drug delivery in the therapy of macrophage-associated diseases.  相似文献

An olivine nephelinite from the lower part of a thick alkalic ultrabasic and mafic sequence of volcanic rocks of the northeastern part of the Siberian flood basalt province (SFBP) yielded a (40)Ar/(39)Ar plateau age of 253.3 +/- 2.6 million years, distinctly older than the main tholeiitic pulse of the SFBP at 250.0 million years. Olivine phenocrysts of this rock showed (3)He/(4)He ratios up to 12.7 times the atmospheric ratio; these values suggest a lower mantle plume origin. The neodymium and strontium isotopes, rare earth element concentration patterns, and cerium/lead ratios of the associated rocks were also consistent with their derivation from a near-chondritic, primitive plume. Geochemical data from the 250-million-year-old volcanic rocks higher up in the sequence indicate interaction of this high-(3)He SFBP plume with a suboceanic-type upper mantle beneath Siberia.  相似文献
