Stormwater wetlands collect and attenuate runoff-related herbicides, limiting their transport into aquatic ecosystems. Knowledge on wetland bacterial communities with respect to herbicide dissipation is scarce. Previous studies showed that hydrological and hydrochemical conditions, including pesticide removal capacity, may change from spring to summer in stormwater wetlands. We hypothesized that these changes alter bacterial communities, which, in turn, influence pesticide degradation capacities in stormwater wetland. Here, we report on bacterial community changes in a stormwater wetland exposed to pesticide runoff, and the occurrence of trz, atz, puh, and phn genes potentially involved in the biodegradation of simazine, diuron, and glyphosate. Based on T-RFLP analysis of amplified 16S rRNA genes, a response of bacterial communities to pesticide exposure was not detected. Changes in stormwater wetland bacterial community mainly followed seasonal variations in the wetland. Hydrological and hydrochemical fluctuations and vegetation development in the wetland presumably contributed to prevent detection of effects of pesticide exposure on overall bacterial community. End point PCR assays for trz, atz, phn, and puh genes associated with herbicide degradation were positive for several environmental samples, which suggest that microbial degradation contributes to pesticide dissipation. However, a correlation of corresponding genes with herbicide concentrations could not be detected. Overall, this study represents a first step to identify changes in bacterial community associated with the presence of pesticides and their degradation in stormwater wetland.  相似文献

Acrylamide formation under controlled processing conditions was studied in a starch matrix by analyzing volatile compounds in the gas phase using online mass spectrometry. Compounds were identified using mass spectral analysis, authentic standards, and the labeling patterns from isotopically labeled asparagine and sugars. Acrylamide, 3-aminopropanamide, methylpyrazine, 3-oxopropanamide, and aminopropan-2-one were assigned to the ions at m/ z 72, 89, 95, 88, and 74, respectively. Ion m/ z 60 was proposed as the transamination product of glyoxal, but labeling experiments did not support this assignment. Temporal formation of acrylamide and related compounds was studied in 51 samples containing asparagine and selected sugars or carbonyls. Data from the experiments were analyzed to investigate correlations between the amounts of acrylamide, intermediates, and pyrazines formed. A strong correlation between 3-aminopropanamide and acrylamide was found in all samples, whereas other correlations were reactant specific. Preliminary multiway analysis of the data identified temporal similarities in the ion profiles and showed that dynamic monitoring can follow the production and utilization of intermediates leading to acrylamide.  相似文献

Prohexadione-Ca is a structural mimic of 2-oxoglutarate, and according to this property, it is able to inhibit dioxygenase enzymes, which require 2-oxoglutarate as a cosubstrate. Such enzymes are involved in flavonoid biosynthesis; therefore, prohexadione-Ca treatment leads to alterations in the flavonoid metabolism in grapevine tissues. Because of the fact that phenolic compounds often are responsible for enhanced plant resistance, modification of phenylpropanoid metabolism using elicitation can be considered as a new potential strategy in plant protection. The phenolic compounds were analyzed by high-performance liquid chromatography combined with chemical reaction detection. Tissue treatment induced the accumulation of unusual flavonoids, which were identified as derivatives of pentahydroxyflavanone, eriodictyol, and luteoliflavan. Concentrations of constitutive flavonoids were also affected by the bioregulator treatment. The alterations of the flavonoid profiles are discussed with respect to substrate preferences of relevant enzymes.  相似文献

Cover cropping is currently increasing in vineyards as it provides solutions to some of the problems encountered in vine growing. However, its development is still hampered in Mediterranean regions because of fears of severe competition for water. Recent studies have shown that soil resources other than water may also be restrictive, and particularly nitrogen. Over a three-year period, the effect of introducing a cover crop was studied with respect to the temporal and spatial changes to nitrogen dynamics in a Mediterranean vineyard. The experiment compared the impact of three different types of soil cover management on nitrogen dynamics, and particularly on soil nitrogen mineralization which is the principal source of inorganic nitrogen in situations with no application of N fertilizers which are frequent in viticulture.This experiment provided evidence that the presence of an intercrop significantly reduced nitrogen accumulation in aerial parts of grapevine during the year due to competition for soil resources. This reduction varied markedly between years and treatments, and was more pronounced during dry years. The competition for nitrogen was direct as intercrop deprived grapevine of soil nitrogen beneath the inter-row and caused grapevine uptake to be higher beneath the row. In deep soils, a grapevine can adapt its root system in order to access deeper water resources, but it then partially abandons the mineralization zone containing most inorganic nitrogen. Competition for nitrogen was less marked with a temporary cover crop than with a permanent one, because of the shorter period of uptake with the former and the time needed for an annual cover crop to develop its root system each year. Intercrop also competed indirectly for nitrogen with grapevine as it took up soil water and made inorganic nitrogen less mobile and accessible to grapevine. Intercrop markedly decreased soil nitrogen mineralization. Although it did not significantly affect organic matter characteristics or soil temperature, it clearly modified the water regime. Indeed, under either temporary or perennial cover crops, the upper soil layers dried more rapidly than when there was only evaporation from bare soil. Consequently, nitrogen mineralization decreased faster with intercropped treatments and halted prematurely during the summer.The earliness of the reduction in nitrogen accumulation in intercropped vineyards also suggested that a lower level of nitrogen transfer to perennial reserves was involved. Indeed, grass cover grows and competes for nitrogen during the autumn which is a favourable period for nitrogen accumulation in wood reserves. Although better water infiltration was observed in the presence of a cover crop (notably in the autumn), the favourable conditions for nitrogen mineralization were propitious for grass cover growth and uptake. Consequently, intercrop reduced grapevine growth of the year but also the potential growth for the next year by decreasing grapevine nitrogen perennial reserves.  相似文献

OBJECTIVE: To determine plasma and urine concentrations of retinol, retinyl esters, retinol-binding protein (RBP), and Tamm-Horsfall protein (THP) in dogs with chronic renal disease (CRD). ANIMALS: 17 dogs with naturally developing CRD and 21 healthy control dogs. PROCEDURE: A diagnosis of CRD was established on the basis of clinical signs, plasma concentrations of creatinine and urea, and results of urinalysis. Concentrations of retinol and retinyl esters were measured by use of reverse-phase high-performance liquid chromatography. Concentrations of RBP and THP were measured by use of sensitive ELISA systems. RESULTS: Dogs with CRD had higher plasma concentrations of retinol, which were not paralleled by differences in plasma concentrations of RBP. Calculated ratio of urinary total vitamin A (sum of concentrations of retinol and retinyl esters to creatinine concentration) and ratio of the concentration of urinary retinyl esters to creatinine concentration did not differ between groups. However, we detected a significantly higher retinol-to-creatinine ratio in the urine of dogs with CRD, which was paralleled by a higher urinary RBP-to-creatinine ratio. Thus, in dogs with CRD, the estimated fractional clearance of total vitamin A, retinol, and RBP was increased. Furthermore, dogs with CRD had a reduced urinary THP-to-creatinine ratio. CONCLUSIONS AND CLINICAL RELEVANCE: Results of this study documented that CRD affects the concentrations of retinol in plasma and urine of dogs. Analysis of the data indicates that measurement of urinary RBP and urinary THP concentrations provides valuable information that can be helpful in follow-up monitoring of dogs with CRD.  相似文献

Measurement of total urinary proteins in individuals that tested positive by urinary dipstick is a typical method for assessing the presence of potentially serious renal disorders. In the absence of such overt proteinuria, however, measurement of specific urinary proteins may be useful in the diagnosis of nephropathies and may provide greater insight into the pathogenesis. The urine of 28 dogs (16 with renal disease and 12 healthy) was evaluated to determine whether specific low-molecular-weight proteins or the pattern of protein excretion could also be used as a marker of tubular dysfunction in dogs. Specific proteins were assessed by immunological methods, whereas protein profiles were determined by surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (MS). In particular, changes in the excretion of retinol-binding protein (RBP) and Tamm-Horsfall protein (THP) appear to be of clinical relevance in the diagnosis of canine kidney diseases. The pattern of urinary protein and peptides revealed specific changes in abundance in dogs with renal disease at molecular masses (kD) of 11.58, 12.41, 12.60, 14.58, 20.95 (RBP), 27.85, and 65.69 (albumin). In conclusion, comparable proteins as in humans might be used as urinary markers for proximal (RBP) and distal (THP) tubular dysfunction in dogs. Surface-enhanced laser desorption/ionization time-of-flight MS is a promising tool for the study of kidney physiology and pathophysiology and might aid in the discovery of new biomarkers of renal disease.  相似文献

Modeling height–diameter relationships is an important component in estimating and predicting forest development under different forest management scenarios. In this paper, ten widely used candidate height–diameter models were fitted to tree height and diameter at breast height(DBH)data for Populus euphratica Oliv. within a 100 ha permanent plots at Arghan Village in the lower reaches of the Tarim River, Xinjiang Uyghur Autonomous Region of China. Data from 4781 trees were used and split randomly into two sets:75 % of the data were used to estimate model parameters(model calibration), and the remaining data(25 %) were reserved for model validation. All model performances were evaluated and compared by means of multiple model performance criteria such as asymptotic t-statistics of model parameters, standardized residuals against predicted height,root mean square error(RMSE), Akaike’s informationcriterion(AIC), mean prediction error(ME) and mean absolute error(MAE). The estimated parameter a for model(6) was not statistically significant at a level of a = 0.05. RMSE and AIC test result for all models showed that exponential models(1),(2),(3) and(4) performed significantly better than others. All ten models had very small MEs and MAEs. Nearly all models underestimated tree heights except for model(6). Comparing the MEs and MAEs of models, model(1) produced smaller MEs(0.0059) and MAEs(1.3754) than other models. To assess the predictive performance of models, we also calculated MEs by dividing the model validation data set into 10-cm DBH classes. This suggested that all models were likely to create higher mean prediction errors for tree DBH classes[20 cm. However, no clear trend was found among models.Model(6) generated significantly smaller mean prediction errors across all tree DBH classes. Considering all the aforementioned criteria, model(1): TH ? 1:3 t a= e1 t b?eàc?DBHT and model(6): TH ? 1:3 t DBH2= ea t b?DBH t c ? DBH2T are recommended as suitable models for describing the height–diameter relationship of P. euphratica. The limitations of other models showing poor performance in predicting tree height are discussed. We provide explanations for these shortcomings.  相似文献

Ficus insipida Willd. (Moraceae) is a fast growing tree species of early successional stages in the Amazonian nutrient-rich white-water floodplains (várzea). The species is one of the most economically important low-density wood species in the community-based forest management project in the Mamirauá Sustainable Development Reserve (MSDR) in Central Amazonia, where timber species are managed using a polycyclic selection system with a minimum logging diameter (MLD) of 50 cm and a cutting cycle of 25 years. In this study we analyze the floristic composition, stand structure and forest regeneration of a natural 20 year-old stand at an early successional stage and we model tree growth of diameter, height and volume of F. inspida based on tree-ring analysis to define management criteria. The volume growth model indicates that the preferred period for logging should be at a tree age of 17 years when the current annual volume increment peaks. This age corresponds to a diameter of 55 cm, which would be an appropriate MLD.  相似文献

Exotic potato germplasm may contain useful traits for improving nitrogen (N) use efficiency in cultivated potatoes (Solanum tuberosum L.). The objective of this study was to evaluate a “mini” core collection of wild germplasm for biomass production, N uptake, and N use efficiency. A field study was conducted during the 1993 growing season on a loamy sand soil at Becker, Minnesota. Uniform transplants for 39 wild accessions and 3 varieties were grown under greenhouse conditions, and after 40 days, they were transplanted to the field. Plants were subjected to two N treatments, 0 and 225 kg N ha1, replicated three times in a split-plot design. Plant parts were collected separately 111 days after transplanting, dried, weighed, and analyzed for N concentration. Nitrogen rate and potato species had significant effects on total dry weight, N content, and tissue N concentration. The regression coefficient of N concentration on total dry weight was very low (r=0.22, NS), whereas the regression coefficient of total N content on total dry weight was high (r=0.94, P>0.001). At both low and high N environments, Russet Burbank had greater dry weight than all the accessions. Several wild accessions, especiallyS. chacoense, S. commersonii, S. kurtzianum, S. microdontum, andS.phureja, had equal to or greater dry weights than Russet Norkotah or Red Norland. For N uptake efficiency, 2,7, and 20 accessions were ranked better than Russet Burbank, Russet Norkotah, and Red Norland, respectively. Recovery of soil applied N by the varieties ranged from 16 to 36%; the top seven wild accessions recovered between 27 and 49%. Based on plant growth without added N fertilizer and relative response to N fertilizer, the species were categorized into four relative N use efficiency groups: good foragers with good response to N, poor foragers with good response to N, good foragers with poor response to N, and poor foragers with poor response to N.  相似文献
