Knowledge about the extent of genetic diversity/relatedness in mango germplasm is vital for developing coherent strategies for future gains in productivity. The genetic diversity/relatedness among mango cultivars/genotypes developed in Pakistan has not been investigated previously. We have assessed the genetic diversity among 25 mango genotypes/cultivars using randomly amplified polymorphic DNA (RAPD). Sixty random ten-mer primers were surveyed, out of which 45 yielded amplicons in all the genotypes. Genetic similarity between genotypes/cultivars was in the range of 64–89% with an average of 74%. Similarly, the genetic relatedness among all variants derived from a mango cultivar Chaunsa was in the range of 81.18–88.63%. These coefficients were utilized to construct a dendrogram using the unweighted pair group of arithmetic means (UPGMA). The genotypes were grouped into three (A, B, C) clusters. Generally, genotypes originating from Pakistan were grouped in cluster ‘A’ while cluster ‘B’ primarily composed of southern India as well as Florida cultivars. Kensington Pride was the most distantly related genotype which grouped with Maya and Yakta, forming a distinct cluster ‘C’.  相似文献

Effect of different nitrogen and carbon sources and their concentrations in liquid media on the mycelial growth of six different ectomycorrhizal (ECM) fungal species was studied. Differences were found in the utilization of the different nitrogen and carbon sources between the fungal species. All the species showed better mycelial growth in the medium containing ammonium as nitrogen source. Growth was low in all species in medium in which nitrogen was supplied in nitrate form. All the ECM isolates investigated showed reduced growth in the medium containing maleic acid as the carbon source. The effect of glucose and di-ammonium hydrogen orthophosphate concentrations on mycelia growth of all the six fungal species was studied with ranges for glucose of 2–40 g/l, and for di-ammonium hydrogen orthophosphate of 2–20 g/l. Cortinarius fulvoconicus and Cortinarius flexipes showed maximal mycelial growth at a glucose concentration greater than 20 g/l. Suillus luteus, Scleroderma citrinum, Laccaria laccata, and Tricholoma aurantium showed maximal growth at a glucose concentration of 20 g/l. All six species showed maximal mycelial growth at 5–10 g/l of di-ammonium hydrogen orthophosphate concentration and with increased concentration mycelial growth in all species decreased.  相似文献

In the present study, a total of 380 samples of 5 rice varieties were investigated for the presence of aflatoxins (AFs) and tocopherols (Vitamin E) using HPLC, equipped with a fluorescence detector. The data revealed that super basmati rice has the highest mean level of total AFs (12.45 μg/kg) followed by KS-282 (11.2 μg/kg), basmati PK-385 (9.8 μg/kg), Irri-6 (9.7 μg/kg) and the lowest mean level was found in Irri-9 (8.9 μg/kg). Furthermore, the results have shown that the variety with lowest level of AFs has the highest level of tocopherol content; i.e. Irri-9 (53.2 mg/100 g), basmati PK-385 (45.9 mg/100 g), Irri-6 (45.3 mg/100 g), KS-282 (40.4 mg/100 g) and super basmati rice (40.2 mg/100 g). The data has shown correlation (r = −0.62, p < 0.05) between vitamin E content and AFs concentration in different rice varieties. The results are interesting and need further study to investigate the mechanistic background of vitamin E content and its effect on aflatoxins contamination.  相似文献

The effect of inoculation of Rhizobium or Pseudomonas, or both, in the presence and absence of phosphorus pentoxide (P2O5) fertilizer on wheat yield was tested. The experiment was conducted outdoors in potted soil during two consecutive years under natural conditions. Rhizobial strain (Thal 8-chickpea nodulating bacteria) and Pseudomonas strain (54RB), both indigenous P solubilizers were applied in broth culture at seedling stage. Fertilizer in the form of P2O5 was an additional treatment applied at sowing time. Results revealed that inoculation of Pseudomonas strain in presence of phosphorus (P) fertilizer increased yield up to 41.8% when it was compared with only P fertilizer applied. Co-inoculation of Rhizobium and Pseudomonas strains with the P2O5 treatment however resulted in increased grains yield by 10% over the P2O5 treatment alone. It is inferred that inoculation with Rhizobium and Pseudomonas combined with P2O5 is not only environmentally beneficial but also economically sound and productive, with wheat yield increases of 10% to 42%.  相似文献

Normalized difference vegetation index (NDVI), which is a measure of leaf greenness (chlorophyll content), is considered to be correlated with crop productivity. This study was conducted to examine genotypic variations for NDVI at different growth stages and its relationship to yield in winter wheat under terminal heat stress. Thirty winter wheat genotypes were evaluated at two locations in 2009–2010 and 2010–2011 in Uzbekistan. The NDVI was recorded at booting, heading, milk and dough stages. The wheat genotypes differed significantly for NDVI at each stage. Grain yield ranged from 3.9 to 6.1 t/ha. Wheat genotypes differed in per cent decline in NDVI from booting to dough stage. However, several high‐yielding genotypes maintained higher NDVI than low‐yielding genotypes when heat stress was evident. The findings suggest change in NDVI during heat stress could be a measure of tolerance. The positive correlation of NDVI with grain yield suggests that it could be used as an indirect selection criterion for identifying physiologically superior, high‐yielding wheat lines under terminal heat stress.  相似文献

Genetic diversity among 20 wheat genotypes/cultivars from diverse locations of Pakistan was studied using random amplified polymorphic DNA (RAPD) analysis. A total of 445 DNA fragments were amplified with50 random decamer primers 64.38% of which were polymorphic. Genetic similarity matrix based on Nei & Li's (1979) index detected coefficients ranging from 75.60% to 92.74%. These coefficients were used to construct a dendrogram using unweighted pair group of arithmetic means (UPGMA). The wheat genotypes were clustered into one major group (A) and two small groups (B and C). The most distant genotype in the dendrogram was PARC-1 that was 75.60% to 84.94% genetically similar with the other genotypes and clustered with PARC-3 which formed a group distantly related with the other clusters. Moreover, most of the wheat genotypes developed from the same breeding centre clustered in one group. It has been clearly shown that most of the cultivars except PARC-1possessed narrow genetic background. The information would be helpful for future genome mapping programs as well as for the application of intellectual breeder rights in the country. The study will also work as indicator for wheat breeders to evolve varieties with diverse genetic background to achieve sustainability in wheat production in the country. Summary Leptochloa fusca (L.) Kunth (kallar grass) has previously been found to exhibit high rates of nitrogen fixation. A series of experiments to determine the level of biological nitrogen fixation using 15N isotopic dilution were carried out in nutrient solution and saline soil. In the nutrient solution, E. coli inoculated plants were taken as non-nitrogen-fixing control. It was observed that nearly 60%–80% of the plant N was derived from atmospheric fixation. Estimations based on the N difference method gave much lower values (18%–35%). In experiments with saline soil which was initially sterilized with chloroform fumigation, a mixed culture of N2-fixing rhizospheric isolates from kallar grass roots was inoculated and planted to kallar grass. Uninoculated treatments were regarded as controls. The soil was previously labelled with 15N by adding cellulose and (15NH4)2SO4. The results of these studies showed fixation values of 6%–32% when estimated by 15N dilution, whereas by the N difference method 54% of the plant N was estimated to be derived from fixation. This discrepancy is due to the increase in root proliferation due to inoculation, which results in greater uptake of soil N. The distribution of 15N in different fractions of the soil-N indicted isotopic dilution due to bacterial fixation of atmospheric N2.  相似文献

Natural forests offer numerous benefits to indigenous communities and society at large. Incomes from forest sources play an important role in rural households. In addition to this, environmental sources in the forests contribute significantly to rural households’ livelihoods and economic well-being. This paper examines the contributions of forests to the livelihoods of the Chakma tribe in Bangladesh. Using the data from 60 randomly sampled households from three villages, it measured forest-resource use with a monetary yardstick. As revealed through analyses, natural forest-sourced income occupies the second-largest share in total average household income next to shifting cultivation income in the study area. Forest products represent an important component in the local livelihoods, with a direct forest income of 11,256?tk?year?1?household?1 (tk = taka, the national currency of Bangladesh; US$1?=?tk70), or 11% of the total income, in addition to the households receiving monetary benefits of 18,951?tk?year?1?household?1, or 21% of the total income, through the consumption of forest products. The remaining income came from shifting cultivation practice. It was also observed that larger families with more people gathering forest products realized more forest income. This study will be relevant to forest and environmental policy-makers as well as indigenous community development practitioners.  相似文献

Aetiology of mango malformation (MM) has intrigued the scientists since its inception. The objective of the study was to glean an insight into association of the fungus, Fusarium mangiferae, with different malformed regions, viz. panicle-shoot juncture, apical buds, primary and secondary peduncles, in five exotic mango cultivars. Tissue assays revealed an infection of 88.5, 84.75 and 82.5% in cvs Zill, Sensation and Tommy Atkins, respectively. Least infection of 69.75% was found in cv. Keitt. No exotic cultivar was found to be free of fungal infection. Apical buds proved to be the potential infection site of the fungus amongst the four malformed regions, hosting maximum within-tissue infection of 86.2%. Determination of F. mangiferae at proximal and distant sites of the malformed panicles exhibited maximum recovery of 82.0% at 0 cm and only 3% at >30 cm distance beneath the panicle. In the case of non-malformed panicles, an infection of 14.0% was recorded at 0 cm distance while no detection could be made from non-malformed branches. Moreover, examination of ultra-thin bud sections under Transmission Electron Microscopy (TEM) revealed inter- and intra-cellular ramification of fungal hyphae, indicating fungal ingress in malformed bud tissues of local cv. Malda. The present studies explored the sites hosting the causal fungus in mango and provide convincing evidence that F. mangiferae is responsible for turning healthy tissues into the malformed condition. These findings suggest that inoculum specific management strategies are needed in future to curb malformation disease in mango orchards.  相似文献
