综述了解淀粉芽孢杆菌在果蔬采后病害防治上的应用及研究进展,归纳了解淀粉芽孢杆菌的筛选途径和研究现状,总结了解淀粉芽孢杆菌的控病作用机制和提高解淀粉芽孢杆菌作用效果的方法;指出解淀粉芽孢杆菌用于果蔬保鲜存在的诸如防效不稳定和商业化困难等问题,并提出与低量化学药剂混用和运用分子生物学手段研究抑菌机理等建议;最后,认为以解淀粉芽孢杆菌为主要成分的生物农药具有广阔的应用前景。 相似文献

旨在克隆山羊SRSF10基因序列,明确其生物学特性,并通过过表达和干扰手段阐明SRSF10对山羊肌内脂肪细胞分化的影响。本研究以简州大耳羊(Capra hircus)为试验对象,利用RT-PCR技术克隆山羊SRSF10基因序列,并进行生物信息学分析; 利用实时荧光定量PCR(real-time quantitative PCR,qPCR)技术研究其在成脂诱导分化不同阶段细胞中的表达水平; 转染pcDNA3.1-SRSF10过表达载体和SRSF10-siRNA至山羊肌内前体脂肪细胞并诱导分化,通过油红O和Bodipy染色法从形态学上明确其对脂滴积聚的影响; 通过qPCR方法检测脂肪细胞分化标志基因表达的变化。结果显示,获得山羊SRSF10基因序列为1 026 bp,其中CDS区552 bp,共编码183个氨基酸; SRSF10在诱导分化96 h的肌内脂肪细胞中表达量最高; 过表达和干扰山羊SRSF10分别促进和抑制了肌内脂肪细胞中的脂滴积聚; 过表达后基因SREBP1、PPARγ和C/EBPα的相对表达量极显著上调(P < 0.01),干扰后分化标志基因SREBP1、PPARγ和C/EBPα相对表达水平极显著降低(P < 0.01)。结果表明,山羊SRSF10是肌内脂肪细胞分化的正调控作用因子,并且这种调控作用可能主要通过调节SREBP1、PPARγ和C/EBPα的表达来实现。 相似文献

富硒茶硒的浸出率及其化学性质的研究 总被引:14,自引:1,他引:14
研究了高硒土壤茶园、低硒土壤茶园、传统制茶工艺、制茶新工艺生产的富硒茶硒的浸出率、化学性质。结果表明,茶叶含硒量对富硒茶硒的浸出率无显著影响,制茶工艺对茶叶硒的浸出率影响显著,传统工艺生产的富硒茶硒的浸出率为12.40%~19.58%,红茶高于绿茶。笔者采用新工艺生产的富硒茶硒的浸出率为19.41%~29.38%,硒的浸出率提高50%。不同来源的富硒茶中近80%以上的硒为有机硒,并主要呈蛋白质结合态。富硒茶粗蛋白质中胱氨酸、甲硫氨酸的含量随茶叶含硒量提高而增加。不同来源的富硒茶含硒蛋白SDS-电泳带分析表明,富硒茶的化学性质相似,低硒土壤茶园通过施用硒肥,经茶树生物转化生产富硒茶与天然富硒茶无显著差异。 相似文献

Analysis by GC/MS/MS showed that a worldwide collection of 58 wheat accessions differed significantly in the production of seven phenolic acids in the roots of 17-day-old wheat seedlings. The allelochemical contents among wheat accessions ranged from 24.5 to 94.5, 19.9 to 91.7, 3.7 to 15.4, 2.2 to 38.6, 1.0 to 42.2, 19.3 to 183.6, and 11.7 to 187.6 mg/kg of root dry weight for p-hydroxybenzoic, vanillic, cis-p-coumaric, syringic, cis-ferulic, trans-p-coumaric, and trans-ferulic acids, respectively. trans-Ferulic acid was identified as the most predominant phenolic acid in the roots. Phenolic acids, with the exception of syringic acid, were more concentrated in roots than in shoots. Significant correlation was found between the roots and the shoots in the contents of vanillic, cis-p-coumaric, syringic, trans-p-coumaric, and trans-ferulic acids, and in the content of each structural group of phenolic acids. Wheat accessions with high levels of total identified phenolic acids in the roots were generally strongly allelopathic to the growth of annual ryegrass. 相似文献

Alkylpyrazines are a very important class of Maillard flavor compounds, but their mechanism of formation is complex and consists of different pathways. The model reaction of 20 different amino acids with 1,3-dihydroxyacetone, as a precursor of 2-oxopropanal, was studied by means of SPME-GC-MS to investigate the involvement of the amino acid side chain in the substitution pattern of the resulting pyrazines. 2,5-Dimethylpyrazine was quantitatively the most important pyrazine formed from all of the amino acids. The amino acid side chain is not involved in its formation. The substituents of other less abundant pyrazines resulted mainly from the incorporation of the Strecker aldehyde or aldol condensation products in the intermediate dihydropyrazine. The importance of different reaction mechanisms was evaluated, taking into account the pattern of pyrazines identified. In the solvent extracts of aqueous model reactions of 2-oxopropanal with amino acids, the main reaction product was not a pyrazine but a novel pyrrole. This pyrrole was identified as 2,5-diacetyl-3-methyl-1 H-pyrrole by means of spectral analysis, secured by chemical synthesis. A reaction mechanism for its formation was proposed and evaluated. The influence of various reaction conditions on the formation of 2,5-diacetyl-3-methyl-1 H-pyrrole and 2,5-dimethylpyrazine in the model reaction of alanine with 2-oxopropanal was studied. The aim of the present study was to establish the technology of intracytoplasmic sperm injection (ICSI) in rabbit by using the sperm frozen without cryoprotectants. Observation under an electron microscope revealed that the rabbit spermatozoa frozen without cryoprotectants had severe damage especially in the plasma membrane and junction between head and tail. However, after being injected into the oocytes, the sperm frozen without cryoprotectants retained the capability of supporting the cleavage and development of the ICSI oocytes, with no significant difference from that of fresh sperm, although the development of ICSI embryos derived from either frozen sperm or fresh sperm is much lower than that of in vivo‐fertilized zygotes. When additional artificial activation was applied following ICSI, the rates of cleavage and blastocyst formation of ICSI oocytes were significantly increased when compared with the oocytes without additional activation. Yet, the cell numbers in blastocysts were not significantly different between the activation and non‐activation group. After embryo transfer, four offspring were obtained from the oocytes microinjected with the sperm frozen without cryoprotectants. The technology established by this study may facilitate exploring the ICSI‐based transgenic method in rabbit and broaden the application of ICSI technique in related field. 相似文献
