To evaluate the effect of thermal and microbial stress on the immune response of common carp (Cyprinus carpio L.), relative mRNA expression level of pro-inflammatory cytokines [tumor necrosis factor alpha (TNF-α) and interleukin (IL)-1β] and other genes related to immune or stress response [inducible nitric oxide synthase (iNOS), heat shock protein 70 (Hsp70), superoxide dismutase one (SOD1), and glucocorticoid receptor (GR)] was measured by quantitative PCR (qPCR). In addition, total protein and total immunoglobulin level in blood plasma of experimental common carp was also assayed. All the above parameters were estimated 24 h post-challenge with Gram-negative bacterium, Aeromonas hydrophila. Common carp (54.89?±?6.90 g) were initially exposed to 20 °C (control group) and 30 °C (thermal stress group) water temperature for 30 days, followed by experimental challenge with 2.29?×?108 colony forming unit/mL (CFU/mL; LD50 dose) of A. hydrophila. Exposure of fish to thermal stress and subsequently challenge with A. hydrophila significantly (P?<?0.05) increases the IL-1β mRNA expression in head kidney and spleen of common carp by ~?39.94 and ~?4.11-fold, respectively. However, TNF-α mRNA expression in spleen decreased ~?5.63-fold in control fish challenged with A. hydrophila. Thermal stress and challenge with bacterium suppresses the iNOS and GR mRNA expression in spleen of common carp. Moreover, significant (P?<?0.05) increase in total protein content of blood plasma (~?43 mg/g) was evident in fish exposed to thermal stress and challenged with A. hydrophila. A whole life‐cycle bioenergetic model based on the dynamic energy budget (DEB) theory was proposed for the three‐spined stickleback (Gasterosteus aculeatus). To develop this model, experiments on growth and reproduction were performed: adult and juvenile growth, size at first reproduction and amount of eggs spawned by females were monitored under different feeding levels and temperatures. The DEB parameters were estimated, using Bayesian statistics, based on the data produced during these experiments and on other data found in the literature. The model fitted accurately the different data used for the calibration process and, in addition, predicted accurately the data sets used to assess its predictability. Our bioenergetic model of the whole life cycle of the three‐spined stickleback accounting for environmental variations could contribute in many ways to improved ecological assessment: supporting change of scale from individual to populations; developing new biomarkers of exposure and effect; analysing ecotoxicity tests with biology‐based models. 相似文献

assay using liver membranes from pregnant rabbits and bovine GH as standard and labelled hormone. The enrichment of the preparation
was checked with electrophoresis (SDS-PAGE). Extraction and further steps were carried out using low molarity alkaline buffer
(pH 8–10, M = 100 mM). Three chromatography steps were performed (Concanavalin-A sepharose, Bio-gel P60, DEAE). Ion exchange
chromatography was performed under isocratic conditions (using a 50 cm column). Two isoforms (sGH1 and sGH2) were isolated.
The purification yield is 0.7% compared to lyophilized pituitaries. The molecule is homogeneous in SDS-PAGE. Contamination
by prolactin, gonadotrophin and corticotrophin is negligible (< 0.5%). It could be demonstrated that the biological activity

The present study was undertaken to investigate whether sequential exposure to aerosols of parainfluenza-3 virus followed by Pasteurella haemolytica, or P. haemolytica followed by parainfluenza-3 virus, could lead to the production of pulmonary lesions in conventionally-raised calves. Twenty male calves with low serum antibody titres to both organisms were placed in five equal groups. Synergism of parainfluenza-3 virus and P. haemolytica was not demonstrated in any of the sequentially infected groups and pulmonary lesions were mild in all challenged calves. Clinical signs of disease were not present after exposure to parainfluenza-3 virus although the virus was repeatedly isolated from nasal secretions of all inoculated calves. Exposure to P. haemolytica produced a transient response which consisted of increased rectal temperatures and respiratory rates, with a mild neutrophilic leukocytosis and a mild left shift present six hours postinoculation and returning to normal within 24 hours. Results from this study suggest, although do not confirm, that reduced pulmonary clearance of inhaled P. haemolytica in parainfluenza-3 virus infected calves does not necessarily lead to production of severe pulmonary lesions and that previous exposure to aerosols of P. haemolytica may not enhance secondary parainfluenza-3 virus infection. 相似文献

To establish the sublethal concentrations domain, acute and chronic oral tests were conducted on caged honeybee workers (Apis mellifera L) using imidacloprid and a metabolite, 5-OH-imidacloprid, under laboratory conditions. The latter showed a 48-h oral LD50 value (153 ng per bee) five times higher than that of imidacloprid (30 ng per bee). Chronic feeding tests indicated that the lowest observed effect concentrations (LOEC) of imidacloprid and of 5-OH-imidacloprid on mortality of winter bees were 24 and 120 microg kg(-1) respectively. Behavioural effects of imidacloprid and 5-OH-imidacloprid were studied using the olfactory conditioning of proboscis extension response at two periods of the year. Winter bees surviving chronic treatment with imidacloprid and 5-OH-imidacloprid had reduced learning performances. The LOEC of imidacloprid was lower in summer bees (12 microg kg(-1)) than in winter bees (48 microg kg(-1)), which points to a greater sensitivity of honeybees behaviour in summer bees, compared to winter bees. 相似文献

Riparian forests are classified as endangered ecosystems in general,particularly in sahelian countries like Burkina Faso because of human-induced alterations and civil engineering works.The modification of this important habitat is continuing,with little attention being paid to the ecological or human consequences of these changes.The objective of this study is to describe the variation of woody species diversity and dynamic in riparian forests on different type of watercourse banks along phytogeographical gradient in Burkina Faso.All woody species were systematically measured in 90 sample plots with sides of 50 m × 20 m.Density,dominance,frequency and species and family importance values were computed to characterize the species composition.Different diver-sity indices were calculated to examine the heterogeneity of riparian forests.A total of 196 species representing 139 genera and 51 families were recorded in the overall riparian forests.The species richness of individuals with dbh ≥ 5cm increased significantly from the North to the South along the phytogeographical gradient and varied significantly between the different types of riparian forests.Similarity in tree species composition between riparian forests was low,which indicates high beta diversity and reflects differences in habitat conditions and topography.The structural characteristics varied significantly along the phyto-geographical gradient and between the different types of riparian forests.The diameter class distribution of trees in all riparian forests showed a reverse "J" shaped curve except riparian forest of stream indicating vegetation dominated by juvenile individuals.Considering the ecological importance of riparian forest,there is a need to delineate and classify them along watercourses throughout the country. 相似文献
