Antarctic snowfall exhibits substantial variability over a range of time scales, with consequent impacts on global sea level and the mass balance of the ice sheets. To assess how snowfall has affected the thickness of the ice sheets in Antarctica and to provide an extended perspective, we derived a 50-year time series of snowfall accumulation over the continent by combining model simulations and observations primarily from ice cores. There has been no statistically significant change in snowfall since the 1950s, indicating that Antarctic precipitation is not mitigating global sea level rise as expected, despite recent winter warming of the overlying atmosphere.  相似文献

Lichens form the dominant plant cover in extreme environments and participate in mineral weathering, fine-earth stabilization and primary accumulation of soil organic matter. However, biochemical role of lichens in soil processes has never been investigated. Recently, laccases and tyrosinases have been discovered in representatives of the order Peltigerales (Laufer et al., 2006a, b; Zavarzina and Zavarzin, 2006). Laccases from most species had unusually large molecular weights (Laufer et al., 2009). Together with oligomeric laccases, we have found monomeric enzymes in Solorina crocea and Peltigera aphthosa (Lisov et al., 2007). In the present work we have purified homodimeric (large) and monomeric (small) laccases of the soil-stabilizing lichen S. crocea, determined their physico-chemical and catalytic properties and studied their reactions with soil humic acids. Our results suggest that oligomeric nature of lichen laccases can be artifactual, because homodimeric laccase was transformed into the monomeric form following hydrophobic interaction chromatography. We hypothesize that large laccase consists of two monomeric enzymes, each of which is bound with additional hydrophobic component(s). Small laccase is similar in its properties to the laccases of basidiomycetes. It is more resistant to elevated temperature and storage than the large form, showed a higher oxidation potential, had different pH-optima in oxidizing substrates and was less inhibited by humic acids. Despite these differences, both laccases depolymerized and decolorized humic acids from soils at comparable rates, with small laccase being slightly more effective. This finding suggests that lichens have a potential to participate in transformation of soil organic matter.  相似文献

Using pitfall traps, we evaluated the spatial and seasonal variance in arthropod abundance, species richness, higher taxonomic and species composition, and guild structure within the ground litter of seven sites in a relatively undisturbed rainforest in Panama. We examined each of these five arthropod-dependent variables at two spatial scales (a few meters and a few hundred meters) and one temporal scale (a few months encompassing dry and wet periods), against environmental variables including local illumination and plant composition. Trap catches (9458 arthropods collected during 630 trap-days) were high compared to similar studies in temperate forests. We observed spatial and seasonal differences in abundance, species richness and composition of litter-dwelling arthropods. Often these differences appeared weakly related to geographical coordinates. They reflected forest structure (basal area) and local plant composition, and less so illumination patterns or seasonal changes in radiation. Seasonal variance was high and may relate to surrogate variables accounting for seasonal changes in litter moisture. The composition of higher taxa and species was often predicted by different independent variables at the three scales studied. Guild structure was difficult to predict. Our study lead us to expect that litter-dwelling arthropods may be more seasonal than soil microarthropods in tropical rainforests; and that tropical litter-dwelling arthropods may also be more spatially variable and seasonal than their temperate counterparts. We also recommend that conservation studies using pitfall traps in tropical rainforests should focus on: (1) taxonomic resolution to understand the functional complexity of soil organisms; (2) spatial replication to address subtle changes in plant composition throughout the study area; and (3) seasonal replicates to be commensurate with seasonal changes in litter moisture.  相似文献

Generally, dry deposition processes are very important for atmospheric chemistry of pollutants providing up to 30–80 % of the removal for certain compounds from the atmosphere. The model for calculating of dry deposition fluxes for a large territory seems unsophisticated in spite of the dependence on surface characteristics, pollutant properties and atmospheric conditions. The approach of combining monthly average concentrations measured at the Integrated Background Monitoring Network (IBMoN) and EMEP stations and linear dry deposition velocity was used to calculate total sulphur and nitrogen fluxes for the whole of the former Soviet Union (FSU) taking into account large-scale geographical variability in climate and lands. Most values of all SO2 and SO4 2? concentrations were below 2.9 and 3.1 mgS/m3, and NO2 concentration were 1.5 mgN/m3 over European part and 0.6 mgN/m3 in Western Siberia. The long-term trends of oxidised sulphur and nitrogen compounds in the atmosphere were examined for 1982–1998 in certain FSU regions. Annual dry deposition of sulphur was estimated as 3.64 Mt S (in sulphate form) and 2.76 Mt S (in SO2 form) for the whole area of FSU. Annual removal of NO2 by dry deposition was calculated at 1.27 Mt N. These values constituted between 44 and 50% of total oxidised sulphur and nitrogen deposition.  相似文献

We have identified a small-molecule inhibitor of tumor necrosis factor alpha (TNF-alpha) that promotes subunit disassembly of this trimeric cytokine family member. The compound inhibits TNF-alpha activity in biochemical and cell-based assays with median inhibitory concentrations of 22 and 4.6 micromolar, respectively. Formation of an intermediate complex between the compound and the intact trimer results in a 600-fold accelerated subunit dissociation rate that leads to trimer dissociation. A structure solved by x-ray crystallography reveals that a single compound molecule displaces a subunit of the trimer to form a complex with a dimer of TNF-alpha subunits.  相似文献

The application of single molecules as templates for nanodevices is a promising direction for nanotechnology. We used a pair of suspended DNA molecules as templates for superconducting two-nanowire devices. Because the resulting wires are very thin, comparable to the DNA molecules themselves, they are susceptible to thermal fluctuations typical for one-dimensional superconductors and exhibit a nonzero resistance over a broad temperature range. We observed resistance oscillations in these two-nanowire structures that are different from the usual Little-Parks oscillations. Here, we provide a quantitative explanation for the observed quantum interference phenomenon, which takes into account strong phase gradients created in the leads by the applied magnetic field.  相似文献

Triple-negative breast cancer (TNBC) represents the deadliest form of gynecological tumors currently lacking targeted therapies. The ethanol extract of the North Pacific brittle star Ophiura sarsii presented promising anti-TNBC activities. After elimination of the inert material, the active extract was submitted to a bioguided isolation approach using high-resolution semipreparative HPLC-UV, resulting in one-step isolation of an unusual porphyrin derivative possessing strong cytotoxic activity. HRMS and 2D NMR resulted in the structure elucidation of the compound as (3S,4S)-14-Ethyl-9-(hydroxymethyl)-4,8,13,18-tetramethyl-20-oxo-3-phorbinepropanoic acid. Never identified before in Ophiuroidea, porphyrins have found broad applications as photosensitizers in the anticancer photodynamic therapy. The simple isolation of a cytotoxic porphyrin from an abundant brittle star species we describe here may pave the way for novel natural-based developments of targeted anti-cancer therapies.  相似文献

Among the brain tumors, glioma is the most common. In general, different biochemical mechanisms, involving nicotinic acetylcholine receptors (nAChRs) and the arachidonic acid cascade are involved in oncogenesis. Although the engagement of the latter in survival and proliferation of rat C6 glioma has been shown, there are practically no data about the presence and the role of nAChRs in C6 cells. In this work we studied the effects of nAChR antagonists, marine snail α-conotoxins and snake α-cobratoxin, on the survival and proliferation of C6 glioma cells. The effects of the lipoxygenase and cyclooxygenase inhibitors either alone or together with α-conotoxins and α-cobratoxin were studied in parallel. It was found that α-conotoxins and α-cobratoxin promoted the proliferation of C6 glioma cells, while nicotine had practically no effect at concentrations below 1 µL/mL. Nordihydroguaiaretic acid, a nonspecific lipoxygenase inhibitor, and baicalein, a 12-lipoxygenase inhibitor, exerted antiproliferative and cytotoxic effects on C6 cells. nAChR inhibitors weaken this effect after 24 h cultivation but produced no effects at longer times. Quantitative real-time polymerase chain reaction showed that mRNA for α4, α7, β2 and β4 subunits of nAChR were expressed in C6 glioma cells. This is the first indication for involvement of nAChRs in mechanisms of glioma cell proliferation.  相似文献

Anionic polysaccharides fucosylated chondroitin sulfates (FCS) from holothurian species were shown to affect various biological processes, such as metastasis, angiogenesis, clot formation, thrombosis, inflammation, and some others. To understand the mechanism of FCSs action, knowledge about their spatial arrangement is required. We have started the systematic synthesis, conformational analysis, and study of biological activity of the oligosaccharides related to various fragments of these types of natural polysaccharides. In this communication, five molecules representing distinct structural fragments of chondroitin sulfate have been studied by means of molecular modeling and NMR. These are three disaccharides and two trisaccharides containing fucose and glucuronic acid residues with one sulfate group per each fucose residue or without it. Long-range C–H coupling constants were used for the verification of the theoretical models. The presence of two conformers for both linkage types was revealed. For the Fuc–GlA linkage, the dominant conformer was the same as described previously in a literature as the molecular dynamics (MD) average in a dodechasaccharide FCS fragment representing the backbone chain of the polysaccharide including GalNAc residues. This shows that the studied oligosaccharides, in addition to larger ones, may be considered as reliable models for Quantitative Structure-Activity Relationship (QSAR) studies to reveal pharmacophore fragments of FCS.  相似文献
