Metabolomics is a recently developed technology for comprehensive analysis of metabolites in organisms. Plant metabolites that are produced for the growth, development, and chemical defense of plants against climatic alterations or natural predators are also useful to us as nutrients or medicines; hence, it is important to comprehend the amounts and varieties of plant metabolites. Besides providing an understanding of the metabolic state in plants under various circumstances, metabolomic techniques are applicable to the clarification of the functions of unknown genes by using natural variants or mutants of the target plants. Furthermore, a metabolomic approach might be useful in the breeding of crops, since valuable plant traits such as taste and yield are closely related to metabolic conditions. Here, we describe the methodology of metabolomics including sample extraction and preparation, metabolite detection, and data processing and analysis, and introduce the application of metabolomic studies to rice.  相似文献

The present study aimed to determine estrogen feedback action sites to mediate prepubertal restraint of gonadotropin-releasing hormone (GnRH)/luteinizing hormone (LH) release in female rats. Wistar-Imamichi strain rats were ovariectomized (OVX) and received a local estradiol-17β (estradiol) or cholesterol microimplant in several brain areas, such as the medial preoptic area (mPOA), paraventricular nucleus, ventromedial nucleus and arcuate nucleus (ARC), at 20 or 35 days of age. Six days after receiving the estradiol microimplant, animals were bled to detect LH pulses at 26 or 41 days of age, representing the pre- or postpubertal period, respectively. Estradiol microimplants in the mPOA or ARC, but not in other brain regions, suppressed LH pulses in prepubertal OVX rats. Apparent LH pulses were found in the postpubertal period in all animals bearing estradiol or cholesterol implants. It is unlikely that pubertal changes in responsiveness to estrogen are due to a change in estrogen receptor (ER) expression, because the number of ERα-immunoreactive cells and mRNA levels of Esr1, Esr2 and Gpr30 in the mPOA and ARC were comparable between the pre- and postpubertal periods. In addition, kisspeptin or GnRH injection overrode estradiol-dependent prepubertal LH suppression, suggesting that estrogen inhibits the kisspeptin-GnRH cascade during the prepubertal period. Thus, estrogen-responsive neurons located in the mPOA and ARC may play key roles in estrogen-dependent prepubertal restraint of GnRH/LH secretion in female rats.  相似文献

The quality of habitat at and around a spawning site, and the availability of movement between spawning and nonspawning habitats are likely to be important determinants for population persistence in a degraded and fragmented landscape. We assessed the influence of habitat connectivity, habitat quality and invasive species for distributions and local abundance of eggs and larvae of crucian carp (Crassius auratus complex, which is listed as “data deficient” on the Japanese Red List) in agricultural landscapes surrounding Lake Mikata, Japan, where drainage ditches and paddy fields are extensively utilised for spawning (lake or river shores are also used). We investigated the presence and abundance of eggs and larvae of crucian carp and habitat components at 146 sites across a range of presumed spawning habitats. Egg presence was affected strongly by connectivity to the lake (watercourse distance from the lake), and egg abundance was significantly influenced by both connectivity and habitat quality. In contrast, larval presence was primarily related to habitat quality. Larval abundance was influenced by connectivity and habitat quality, but the effect of connectivity was relatively low. Furthermore, larval abundance was negatively related to the presence of the invasive species red swamp crayfish (Procambarus craki) and bullfrog (Rana catesbeiana). Our findings indicate that connectivity, habitat quality and the presences of invasive species are crucial in determining suitable spawning and nursery habitats, but their relative importance may vary depending on egg and larval life stages. We suggest that restoring connectivity, improving habitat quality and removal of invasive species could be effective conservation strategies for the declining populations of crucian carp in agricultural landscapes.  相似文献

Recent discovery of a phase transition from perovskite to post-perovskite suggests that the physical properties of Earth's lowermost mantle, called the D' layer, may be different from those of the overlying mantle. We report that the electrical conductivity of (Mg0.9Fe0.1)SiO3 post-perovskite is >10(2) siemens per meter and does not vary greatly with temperature at the conditions of the D' layer. A post-perovskite layer above the core-mantle boundary would, by electromagnetic coupling, enhance the exchange of angular momentum between the fluid core and the solid mantle, which can explain the observed changes in the length of a day on decadal time scales. Heterogeneity in the conductivity of the lowermost mantle is likely to depend on changes in chemistry of the boundary region, not fluctuations in temperature.  相似文献
