ABSTRACT DNA markers tightly linked to resistance (R) genes provide a very powerful tool for both marker-assisted selection in plant breeding and positional cloning of R genes. In the present study, a linkage of random amplified polymorphic DNA (RAPD) markers to the single dominant gene (Cr2) for resistance to white pine blister rust fungus (Cronartium ribicola) was investigated in western white pine (Pinus monticola). A mapping population of 128 individual megagametophytes was generated from seeds of a heterozygous resistant tree (Cr2/cr2), and the corresponding seedlings of each megagametophyte were subjected to the test of phenotype segregation by inoculation with C. ribicola. Bulked segregant analysis and haploid segregation analysis identified eight robust RAPD markers linked to Cr2. This constitutes the first Cr2 genetic linkage map spanning 84.7 cM with four markers only 3.2 cM from Cr2. One sequence (U256-1385) of these linked markers was significantly similar to the Ty3/gypsy-like long terminal direct repeats retrotransposons. Another marker, U570-843, had no significant similarity to any entry in either GenBank or the loblolly genomics data bank. As presumed that the average physical distance per centimorgan is about 10 Mb in P. monticola, it is probably unrealistic to use these DNA markers for positional cloning of the Cr2 gene. 相似文献

ABSTRACT Host plant resistance is an economical and environmentally sound method of control of leaf rust caused by the fungus Puccinia triticina, which is one of the most serious diseases of wheat (Triticum aestivum) worldwide. Wild relatives of wheat, including the tetraploid T. timopheevii subsp. armeniacum, represent an important source of genes for resistance to leaf rust. The objectives of this study were to (i) evaluate the performance of leaf rust resistance genes previously transferred to wheat from three accessions of T. timopheevii subsp. armeniacum, (ii) determine inheritance and allelic relationship of the new leaf rust resistance genes, and (iii) determine the genetic map location of one of the T. timopheevii subsp. armeniacum-derived genes using microsatellite markers. The leaf rust resistance gene transferred to hexaploid wheat from accession TA 28 of T. timopheevii subsp. armeniacum exhibited slightly different infection types (ITs) to diverse races of leaf rust in inoculated tests of seedlings compared with the gene transferred from TA 870 and TA 874. High ITs were exhibited when seedlings of all the germ plasm lines were inoculated with P. triticina races MBRL and PNMQ. However, low ITs were observed on adult plants of all lines having the T. timopheevii subsp. armeniacum-derived genes for resistance in the field at locations in Kansas and Texas. Analysis of crosses between resistant germ plasm lines showed that accessions TA 870 and TA 874 donated the same gene for resistance to leaf rust and TA 28 donated an independent resistance gene. The gene donated to germ plasm line KS96WGRC36 from TA 870 of T. timopheevii subsp. armeniacum was linked to microsatellite markers Xgwm382 (6.7 cM) and Xgdm87 (9.4 cM) on wheat chromosome arm 2B long. This new leaf rust resistance gene is designated Lr50. It is the first named gene for leaf rust resistance transferred from wild timopheevi wheat and is the only Lr gene located on the long arm of wheat homoeologous group 2 chromosomes. 相似文献

OBJECTIVE: To determine and correlate subchondral bone mineral density and overlying cartilage structure and tensile integrity in mature healthy equine stifle (low magnitude loading) and metacarpophalangeal (high magnitude loading) joints. ANIMALS: 8 healthy horses, 2 to 3 years of age. PROCEDURE: Osteochondral samples were acquired from the medial femoral condyle (FC) and medial trochlear ridge (TR) of the stifle joint and from the dorsal (MC3D) and palmar (MC3P) aspects of the distal medial third metacarpal condyles of the metacarpophalangeal joint. Articular cartilage surface fibrillation (evaluated via India ink staining) and tensile biomechanical properties were determined. The volumetric bone mineral density (vBMD) of the underlying subchondral plate was assessed via dual-energy x-ray absorptiometry. RESULTS: Cartilage staining (fibrillation), tensile moduli, tensile strength, and vBMD were greater in the MC3D and MC3P locations, compared with the FC and TR locations, whereas tensile strain at failure was less in MC3D and MC3P locations than FC and TR locations. Cartilage tensile moduli correlated positively with vBMD, whereas cartilage staining and tensile strain at failure correlated negatively with vBMD. CONCLUSIONS AND CLINICAL RELEVANCE: In areas of high joint loading, the subchondral bone had high vBMD and the articular cartilage surface layer had high tensile stiffness but signs of structural wear (fibrillation and low failure strain). The site-dependent variations and relationships in this study support the concept that articular cartilage and subchondral bone normally adapt to physiologic loading in a coordinated way. 相似文献

OBJECTIVE: To evaluate changes over time in echocardiographic measurements in young Standardbred racehorses undergoing training and racing and determine whether there was any relationship between cardiac dimensions and racing performance. DESIGN: Longitudinal observational study. ANIMALS: 103 horses. PROCEDURE: 2-dimensional and M-mode echocardiography was performed 4 times at 6-month intervals. RESULTS: Significant cardiac enlargement took place during the study period as indicated by increases in left ventricular internal diameter in diastole (LVIDd), estimated left ventricular muscle mass (LV mass), and mean wall thickness attributable to eccentric left ventricular hypertrophy. Estimated body weight was positively correlated with left ventricular size, and males had significantly larger LVIDd and LV mass than did females. Horses that were racing regularly had larger LVIDd and LV mass than did unraced horses. A significant relationship between left ventricular size and racing performance was observed. The relationship was strongest at the time of the fourth examination. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that age must be taken into account when interpreting results of echocardiography in young Standardbred racehorses because significant cardiac enlargement takes place with age and training. A larger heart was found in horses that were racing, and size of the heart was correlated with athletic performance of the horse. 相似文献
