Horses may be exposed to aflatoxin B(1) (AFB(1)) via inhalation of mouldy dust, leading to high exposure of olfactory and respiratory tissues. In the present study the metabolic activation of AFB(1) was examined in olfactory and respiratory tissues in horse. The results showed covalent binding of AFB(1)-metabolites in sustentacular cells and cells of Bowman's glands in the olfactory mucosa, in some cells of the surface epithelium of nasal respiratory, tracheal, bronchial and bronchiolar mucosa and in some glands in these areas. Immunohistochemistry revealed that cells expressing proteins reacting with CYP 3A4- and CYP 2A6/2B6-antibodies had a similar distribution as those having capacity to activate AFB(1). Our data indicate that the cell-specific activation of AFB(1) correlates with presence of some CYP-enzymes in olfactory and respiratory tissues in horse. 相似文献

This study aimed to validate the use of an automated chemiluminescent immunoassay analyser for salivary testosterone measurements in growing pigs and study how circadian pattern during daytime and stress can influence its values. The test method had intra- and inter-assay coefficient of variation lower than 10%. The method showed good linearity and recovery, and detection limits were low enough to detect salivary testosterone levels. No significant differences were observed in testosterone concentrations at different sampling time, and age and gender did not influence circadian pattern. In addition, this assay was used to quantify testosterone in two models of acute stress and, in both cases, significant increases (P < 0.01) in salivary testosterone were detected. Therefore, the automated assay system tested for porcine testosterone determinations would be suitable for its use in saliva samples and, furthermore, salivary testosterone levels could be used as a possible marker of acute stress in pigs. 相似文献

Delayed acute capture myopathy is the term used to describe the clinical syndrome observed in three roe deer captured by drive-nets and transported to an enclosure for scientific purposes. The animals died 48 h, 60 h and 8 days after being captured. The simultaneous deaths coincided with a previous episode of deliberate human disturbance. The histopathological findings were indicative of acute myopathy and myoglobinaemic nephrosis. These could be related to an ataxic myoglobinuric syndrome brought on by capture and transport operations. The lack of clinical signs and negative prognosis indicators in the period between capture and just before death. the absence of gross muscular lesions in the animal that died after 8 days post-capture, the simultaneous deaths of animals captured at different times and the evidence of deliberate human disturbance in the enclosure are suggestive of death triggered by a second stress episode. 相似文献

To evaluate the effect of Spanish summer commercial journeys on the stress response of young bulls born and reared under extensive conditions, 2 replicates of a transport from an assembly centre to a growing-finishing farm were studied. Journeys lasted 27 h, involving a total of 62 young bulls. Variables under study included haematocrit, red blood cell count (RBC), total white blood cell count (WBC), differential WBC counts, serum haptoglobin (Hp), cortisol, glucose, creatine phosphokinase (CPK), lactate dehydrogenase (LDH), total protein, and albumin at loading, at the end of an intermediate market stop, and at the unloading. Before the beginning of the journey elevated WBC and neutrophil counts, and high Hp values were detected, reflecting high stress levels probably as a consequence of previous procedures associated with the grouping at the assembly centre. Some stress was also detected at the end of the market stop, with cortisol increasing from 6.5 to 12.6 ± 2.0 ng/mL (P < 0.001), although a change in Hp concentration was not observed. Neither CPK and LDH activities, related to muscular tissue damage, nor haematocrit and RBC count, related to dehydration revealed a significant effect of this first stage of the journey on the physical stress of the young bulls. Subsequent 13 h transport to the growing-finishing farm induced an increase in Hp levels from 0.48 to 0.78 ± 0.16 mg/mL (P < 0.001), reflecting an onset of the acute stress response, although cortisol levels immediately after the unloading were similar to those found before loading at the market, suggesting that calves got accustomed to transport. At the end of the journey some dehydration and physical stress were also detected. Overall, our study provides new information to the discussion of the effect of temperatures during cattle transport. Although an improvement in pre-transport conditions is essential if the welfare of assembled and transported cattle is to be improved, the stress-related alteration of cattle physiology under Spanish summer commercial transport conditions is similar to that observed under colder conditions. 相似文献

Gene and protein expression as well as catalytic activity of cytochrome P450 (CYP) 3A were studied in the nasal olfactory and respiratory mucosa and the tracheal mucosa of the horse. We also examined the activity of NADPH cytochrome P450 reductase (NADPH P450 reductase), the amount of cytochrome b(5) and the total CYP content in these tissues. Comparative values for the above were obtained using liver as a control. The CYP3A related catalytic activity in the tissues of the upper airways was considerably higher than in the liver. The CYP3A gene and protein expression, on the other hand, was higher in the liver than in the upper airway tissues. Thus, the pattern of CYP3A metabolic activity does not correlate with the CYP3A gene and protein expression. Our results showed that the activity of NADPH P450 reductase and the level of cytochrome b(5) in the relation to the gene and protein expression of CYP3A were higher in the tissues of the upper airways than in the liver. It is concluded that CYP3A related metabolism in horse is not solely dependent on the expression of the enzyme but also on adequate levels of NADPH P450 reductase and cytochrome b(5). 相似文献

Serum electrophoresis is a useful technique to diagnose some diseases characterized by quantitative and qualitative changes in the serum proteins, such as the acute phase proteins. An increased synthesis of acute phase proteins has been associated with acute infection and inflammation. We wanted to study whether gastrointestinal diseases and disorders of the locomotor system in horses produce enough inflammatory reaction so the acute phase proteins can be detected by electrophoresis in serum. Serum total proteins were calculated, and the serum electrophoresis from 63 horses was performed. The horses were classified in 3 groups: Group I (19 healthy horses), Group II (20 horses suffering from inflammatory disorders of the gastrointestinal tract), and Group III (24 horses with inflammatory disorders of the locomotor system). Results indicated an increase of the acute phase proteins only in patent inflammatory disorders that showed a rapid progression. Trypanosoma evansi is the most widely spread of the pathogenic African trypanosomes of animals. The disease (surra) was first diagnosed in the Canary Islands in a dromedary camel in 1997; thus, a control plan was implemented achieving the eventual eradication of T. evansi from most of the infected areas in the Archipelago. However, a little area remains still infected despite the use of the same control measures. To evaluate possible reservoirs in the area a representative sample of domestic ruminants was examined by serological, parasitological and molecular tests. Of a total of 1228 ruminants assessed, 61 (5%) were serologically positive (7 cattle, 21 goats, 33 sheep), but T. evansi could be demonstrated in none of them. According to FreeCalc assessment, cattle and goat populations would be free from disease; however, the results from sheep are not adequate to conclude that the population would be free from disease. As a conclusion, surveillance must be exercised on ruminant farms in the surroundings of the infected area in order to evaluate the possible extension of the disease and their potential role as reservoirs of T. evansi. 相似文献
