To assess the effects of three insecticides (aldicarb, chlorpyrifos, deltamethrin) and two fungicides (tebuconazole and metalaxyl + mancozeb) on the PCR-DGGE fingerprints of culturable soil bacterial communities (CSBC), a greenhouse experiment was carried out with soil samples from an Integrated System for Agroecological Production (ISAP), a Conventional Potato Production Area (CPPA) and a Secondary Forest Area (SFA) close to the CPPA. Samples were obtained at 15 day intervals starting at 32 until 77 days after sowing (DAS) to perform the PCR-DGGE analysis of the CSBC cultured on media amended with soil suspension. Analysis of variance from PCR-DGGE data indicated significant differences among treatments. Regardless the type of pesticide applied, CSBC was disturbed and similarity values varied from 5% to 90% in comparison to the control. Significant shifts on CSBC were only detected among treatments in the first two harvests, while CSBC tended to be more akin to each other at the last two harvest dates. The most significant responses observed were due to different soil sample origins, where values of 5% of similarity to the control were observed on CPPA soil. The use of analysis of variance on PCR-DGGE data was useful to a better understanding of the changes on CSBC induced by pesticides applications. 相似文献

CoFe2O4/Zn2SnO4 composite was synthesized using a simple two-step process and applied as a novel-efficient photocatalyst for the rhodamine B degradation from aqueous solution. Characterization techniques such as X-ray diffraction (XRD), N2 adsorption-desorption isotherms, scanning electron microscopy (SEM), EDS analysis, and diffuse reflectance spectroscopy were employed in order to investigate the physical and chemical properties of composite. Higher values of the specific surface area, pore volume and diameter, and a smaller band-gap energy promoted a greater catalytic activity of CoFe2O4/Zn2SnO4 composite when compared to Zn2SnO4. A rapid decolorization of dye solution was observed at 40 min of reaction using the CoFe2O4/Zn2SnO4 catalyst, being 2.5 times faster than the Zn2SnO4 alone. Therefore, the CoFe2O4/Zn2SnO4 composite shows extraordinarily high photocatalytic activity toward the degradation of rhodamine B dye from aqueous solution. 相似文献

Although hardly any polyunsaturated fatty acids (PUFAs) are present in the endproduct, the ingredients used for the production of beer contain a high concentration of PUFAs, such as linolic and linolenic acid. These compounds are readily oxidized, resulting in the formation of lipid-derived products that reduce the taste and quality of beer enormously. During mashing relatively high amounts of PUFAs are exposed to atmospheric oxygen at a relatively high temperature. This makes mashing a critical step in the brewing process with regard to the formation of lipid-derived off-taste products. F1 phytoprostane (PPF1) changes in antioxidant capacity and monohydroxy fatty acids (OH-FAs) were used as markers for the detection of oxidative damage to fatty acids during mashing. The pattern of OH-FA formation indicates that enzymatic oxidation of PUFAs is more important than nonenzymatic oxidation during the mashing process. Nevertheless, substantial nonenzymatic radical formation is evident from the increase of specific OH-FAs and PPF1s. It was found that a low oxygen tension reduces oxidative damage and gives a high antioxidant capacity of the mashing mixture. This study was carried out to establish the changes in the free amino acid contents of floral honeys, honeydew honeys, and blend honeys during storage at room temperature and to test the capacity of the amino acids to distinguish the origin of the honeys after storage. For this purpose, 54 artisanal honeys (39 floral, 5 honeydew, and 10 blend) were studied. Samples were taken from recently collected honeys and at 3, 6, 9, 12, 16, 20, and 24 months after harvesting. The contents of most of the free amino acids were found to decrease with storage time, with the greatest reduction observed in the first 9 months. The contents of the amino acids aspartic acid, beta-alanine, and proline increased in the first few months after storage, reaching maximum values at 6 months, suggesting the possible existence of enzymatic activities. The application of stepwise discriminant analysis to the free amino acid content data demonstrated that the contents of the amino acids valine, beta-alanine, gamma-aminobutyric acid, serine, isoleucine, alpha-alanine, ornithine, and glutamine correctly assigned 87% of honeys to their group of origin: floral, honeydew, or blend. 相似文献

We assessed the sensitivity of mixed exchange resin (MER) and Mehlich-3 (M3) to calcium-bound phosphate (Ca-P) in Oxisols. These methods were compared with Mehlich-1 (M1) and anion exchange resin (AER), which have high and low sensitivity, respectively, to this soil P form. We used maize (Zea mays L.) as a test plant. Three samples from two Oxisols were treated with five ratios of Triple Superphosphate—TS and Bayóvar Phosphate Rock—BPR (0 + 100; 25 + 75, 50 + 50, 75 + 25 and 100 + 0% TS + BPR). The MER extracted more P than AER and M3 at the highest BPR:TS ratios (high Ca-P) and similar quantities at the higher TS:BPR ratios (low Ca-P) for two soil samples. Unlike M1-extracted P, M3-P, AER-P, and MER-P positively correlated with maize dry matter production and P uptake. Mehlich-3 and MER were suitable to assess P availability in high Ca-P Oxisols. 相似文献

The purpose of this study was to evaluate the physico-chemical characteristics and the post-harvest loss of mass of curly lettuce (Lactuca sativa L.) cv. Verônica that was grown in a protective environment with applications of biopesticides. The curly lettuce was treated by spraying with neem oil, cassava flour wastewater (manipueira), biofertiliser or the control treatment of deltamethrin. We determined the weight, circumference, dry matter, ash, crude protein, total phenols and minerals profile for the heads of lettuce. After harvesting, the lettuce was packed into opened plastic bags and kept at 5 °C for 5 days to evaluate its weight loss. No differences were observed among the treatments for the physico-chemical parameters. The lettuce that was sprayed with manipueira or biofertiliser had the greatest weight loss during storage. The achievement of similar yields with cheaper products suggests a benefit of using organic bioinsecticides. 相似文献

The sorption of molybdate by goethite, hematite, bayerite and α-Al2O3 was studied as a function of molybdate concentration and pH. The sorption isotherms exhibited double sorption plateaus in which the amount of Mo sorbed in the second plateau was double that in the first. This was attributed to the polymerization of molybdates as the concentration increases. Furthermore Mo sorption was found to be associated with a cosorption of Na cations, probably present to maintain electroneutrality in the surface zone. The sorption showed high sensitivity towards the pH, attaining a maximum at pH 4. For the iron oxides the decrease in sorption was much less pronounced on the acid side of the pH maximum and occured at lower pH values than that of the Al oxides. In this study, factors influencing the disruption and aggregation of casein micelles during high-pressure (HP) treatment at 250 MPa for 40 min were studied in situ in serum protein-free casein micelle suspensions. In control milk, light transmission increased with treatment time for approximately 15 min, after which a progressive partial reversal of the HP-induced increase in light transmission occurred, indicating initial HP-induced disruption of casein micelles, followed by reformation of casein aggregates from micellar fragments. The extent of HP-induced micellar disruption was negatively correlated with the concentration of casein micelles, milk pH, and levels of added ethanol, calcium chloride, or sodium chloride and positively correlated with the level of added sodium phosphate. The reformation of casein aggregates during prolonged HP treatment did not occur when HP-induced disruption of casein micelles was limited (<60%) or very extensive (>95%) and was promoted by a low initial milk pH or added sodium phosphate, sodium chloride, or ethanol. On the basis of these findings, a mechanism for HP-induced disruption of casein micelles and subsequent aggregation of micellar fragments is proposed, in which the main element appears to be HP-induced solubilization of micellar calcium phosphate. 相似文献

A pot experiment was conducted to study the response of nodulated bean (Phaseolus vulgaris L.), vars. Ceca and Montalbán, to inoculation with the arbuscular‐mycorrhizae (AM) Glomus spurcum strain IES‐3 and G. mosseae strain IES‐8 in two non‐sterilized non‐amended soil substrates (S‐l and S‐2) of contrasting fertility. S‐l was an acid soil (pH 5.0) with 77% exchangeable aluminum (Al), low available phosphorus (P) (3.5 ug g‐1 dry soil) and 1.8% organic matter. S‐2 was a soil with pH 7.0, 7.9 μg P g‐1 dry soil, 9.6% organic matter and no exchangeable Al. Plants growing in S‐1 (S‐1 plants) and S‐2 (S‐2 plants) soils were sampled at the beginning of the flowering stage. S‐1 and S‐2 plants were nodulated by the rhizobial populations native of each soil. S‐1 plants of both varieties did not respond to AM inoculation in term of shoot and root mass, P content, relative abundance of ureides (RAU) and seed yields. Only the total chlorophyll, chlorophyll‐a, chlrophyll‐a/b ratio and nodule mass were significantly enhanced in AM‐inoculated plants. Increased total reducing sugars concentration was detected in roots of S‐1 plants inoculated with G. mosseae. S‐2 plants displayed significantly higher shoot and nodule mass as well as increased total chlorophyll, chlorophyll‐a, chlorophyll‐a/b ratio and P content than S‐1 plants, regardless of the variety and AM inoculation. S‐2 plants of the var Ceca showed a two fold increase in seed yields but similar RAU values (>60%) when compared to S‐1 plants of the same variety. In contrast, the significant increase in RAU detected in S‐2 plants of the var Montalban was not translated into higher seed yields. In S‐2, the productivity of plants of the var Ceca doubled that of the var Montalban. For both bean varieties the highest significant P content and seed yield were observed exclusively in S‐2 plants inoculated with G. mosseae. This Glomelean strain enhanced the sink‐source ratio of the S‐2 plants as evidenced by the higher total reducing sugar concentration in the root mass. Arbuscular‐mycorrhizae inoculation significantly decreased the acid phosphatase activity in the rhizosphere of S‐1 and S‐2 plants, respectively, pointing toward a negative effect of foreign AM on the native microbial biomass. The effectiveness of the rhizobial populations native of each soil type and the weak response elicited by G. mosseae in S‐2 plants do not justify, at present, the inclusion of foreign inocula in the bean crops carried out at S‐1 and S‐2 soils of the Sucre State of Venezuela. Results also indicated the higher adaptability of var Ceca to conditions prevailing in S‐1 and S‐2. 相似文献
