佛山岭南天地旧改项目中开放空间的特色分析

近年来,国内出现了不少基于城市老街区改造为商业街区的项目。通过对佛山岭南天地项目中有代表性的广场、街道、建筑等节点进行调研,选取开放空间的3个特色要素——空间形态和尺度,传统装饰元素,植物和水景布置,探析"新天地"模式中开放空间营造的特色手法,包括保持街巷形态和宜人尺度,增强街区公共属性;保护传统建筑构件,利用传统材料、技术;保留古树,并结合植物和水景布置,形成多层次的景观系统等。     

EZH2-mediated regulation of NF-κB target gene expression in gastric cancer

AIM: To explore the mechanism by which over-expression of enhancer of zeste homolog 2 (EZH2) in a panel of gastric cancer cell lines is involved in tumorigenesis of gastric cancer. METHODS: Real-time PCR and Western blot were employed to examine the mRNA and protein levels of EZH2, respectively. MTS assay, cell migration and soft agar assay were performed to investigate the role of EZH2 in the regulation of stomach cancer behaviors. The effect of EZH2 on NF-κB target gene expression was determined by Luciferase reporter and real-time PCR. Co-immunoprecipitation was used to analyze the interaction of EZH2 and p65 in HEK293T cells. RESULTS: The expression levels of EZH2 were significantly increased in the gastric cancer cells compared with normal gastric epithelial cells. Pharmacological inhibition by DZNep or knockdown of EZH2 significantly compromised AGS and SNU-16 cell activity, cell migration and anchorage-independent cell growth. Moreover, siRNA knockdown of EZH2 impaired NF-κB downstream targets, such as IL-8, CXCL5 and CCL20. In addition, the interaction of EZH2 and p65 was detected. CONCLUSION: EZH2 mediates the growth of gastric cancer cells through the regulation of NF-κB downstream gene expression.     

Impact of fluxapyroxad on the microbial community structure and functional diversity in the silty-loam soil

The aim of this work was to assess the effect of applying three different doses of fluxapyroxad on microbial activity, community structure and functional diversity as measured by respiration, microbial biomass C, phospholipid fatty acid(PLFA) and community-level physiological profiles(CLPPs). Our results demonstrated that substrate-induced respiration(on day 15) and microbial biomass C(on days 7 and 15) were inhibited by fluxapyroxad, but stimulation was observed thereafter. In contrast, fluxapyroxad addition increased the basal respiration and metabolic quotients(q CO2) and respiratory quotients(QR). Analysis of the PLFA profiles revealed that the total and bacterial biomass(both Gram-positive bacteria(GP) and Gram-negative bacteria(GN)) were decreased within the initial 15 days, whereas those as well as the GN/GP ratio were increased at days 30 and 60. F luxapyroxad input decreased the fungi biomass but increased the bacteria/fungi ratio at all incubation time. Moreover, high fluxapyroxad input(75 mg fluxapyroxad kg–1 soil dry weight) increased the microbial stress level. A principal component analysis(PCA) of the PLFAs revealed that fluxapyroxad treatment s ignificantly shifted the microbial community structure, but all of the observed effects were transient. Biolog results showed that average well color development(AWCD) and functional diversity index(H′) were increased only on day 60. I n addition, the dissipation of fluxapyroxad was slow in soil, and the degradation half-lives varied from 158 to 385 days depending on the concentration tested.     

Cloning and Sequence Analysis of PTTG1 Gene in Luchuan Pig

This experiment was aimed to clone PTTG1 gene CDS sequence of Luchuan pig,and was analyzed by bioinformatics methods.A pair of special primers was designed according to predicted sequence of porcine PTTG1 in GenBank.The coding sequence of PTTG1 in Luchuan pig was amplified by RT-PCR,its gene sequence characteristics and protein structure was systemically analyzed by bioinformatics techniques.The results showed that the cloned PTTG1 fragment included a 609 bp CDS (coding 202 amino acids).The sequence multi-aligned results showed that Luchuan pig shared 90.15%,87.85%,87.52%,87.03%,76.03%,74.38%,55.74% and 44.48% of similar nucleotide sequence with that of Bos,Pan troglodytes,Homos,Macaca,Rattus,Mus,Gallus and Danio retio,respectively.The protein structure analysis results showed that the protein attributed hydrophilic protein without signal peptide,localized in cell cytoplasm and had 16 phosphorylation sites.The phylogentic tree of amino acid indicated that PTTG1 was highly conserved in the process of evolution of different species.The cloning and analysis of PTTG1 gene provided an important foundation for further study biological function of porcine PTTG1 during early embryonic development.     

白羊草R2R3-MYB转录因子的挖掘及对干旱胁迫反应的研究

本文以白羊草（Bothriochloa ischaemum）为材料，利用其目前已有的转录组序列，通过搜索、比对、功能域多态性分析、进化树构建和基因表达分析等手段，对白羊草R2R3-MYB型转录因子进行挖掘和干旱胁迫下的表达模式分析。结果从白羊草挖掘出43个R2R3-MYB转录因子，它们具有高度保守的[W]-X（19）-[W]-X（19）-[W]-X（n）-[W]-X（18）-[W]结构，主要参与生长发育、次生代谢和逆境响应等生物过程。通过进一步的分析，本研究筛选出8个可能参与干旱胁迫响应的R2R3-MYB转录因子。这为白羊草R2R3-MYB基因的功能研究和开发利用奠定了基础。     

肾上腺皮质肿瘤良恶性鉴别方法探究

肾上腺皮质肿瘤（Adrenocortical Tumor,ACT）是一种常见的肿瘤,存在于人体的内分泌系统中,并且是引发肾上腺皮质癌（Adrcnocortical Carcinoma,ACC）进而导致死亡的主要原因.经典的＂Weiss系统＂在诊断明确性上存在种种无法解决的问题,但随着科技水平的发展,人们开始探索新的途径来鉴别ACT的良恶性,于是开始有了影像学和分子生物学技术等特异性的检查方法,从而进一步增强了诊断的明确性.现通过对比的方式,探究肾上腺皮质肿瘤（Adrenocortical Tumor,ACT）影像学检查和分子生物学技术检查的结果,从而能更准确地鉴别其良恶性.     

Spectrum-Effect Relationship Between High Performance Liquid Chromatography Fingerprints and Anticoccidial Activities of a Compound Chinese Medicine

Quality control and screening of active substances in traditional Chinese medicines have been performed using fingerprint analysis. The spectrum-effect relationship between chromatography fingerprints and efficacy of herbal drugs is considered as a potentially useful method for determining active ingredients in complex mixtures. The study was designed to develop a method for determining the bioactive components of a compound Chinese medicine called Tiefeng based on spectrum-effect relationships between high-performance liquid chromatography （HPLC） fingerprints and anticoccidial activities. Four peaks of the established HPLC fingerprint indicate the main bioactive components of this medicine. In addition, pharrnacodynamic atlas was defined and used to assess the anticoccidial activity of Tiefeng from different sources for the first time. We found that the level of anticoccidial activity of Tiefeng was consistent with the degree of similarity between the pharmacodynamic atlas and chromatogram of any sample. Furthermore, effect of this medicine was related with the main active constituents, along with the origin and the harvesting time.     

考虑地震载荷作用的水电机组转动部件强度分析

近年地震多发,震中地区的水电机组均受到不同程度的损坏。为研究地震后机组运行的可靠性,根据恢复运行后其运行条件和方式较灾前变化情况,把承受转动部件的上机架纳入分析对象,对应不同工况在上机架上采用应变片测试来验证计算分析的正确性。为了准确计算转动部件承受的载荷,采用CFD对水轮机内部流场进行三维数值模拟,将流场的计算结果作为结构场的初始条件。以地震力和地震加速度两种方式加载地震载荷,采用单向流固耦合方法对各部件进行地震载荷下的仿真计算与工程实测数据对比发现二者吻合较好。结果表明,文章采用的方法能正确评估水轮机结构强度,对机组灾后运行安全性分析有一定指导作用。
