Skeletal muscle genes are potential candidates for production and meat quality. Screening a subtracted cDNA library constructed with mRNA obtained from longissimus dorsi muscles of F1 hybrids Landrace × Yorkshire and their female parents Yorkshire, we isolated two partial sequences coding for the H3-K4-specific methyltransferase (KIAA1717) and skeletal muscle myosin regulatory light chain (HUMMLC2B) genes. Database search revealed KIAA1717 and HUMMLC2B encoded proteins with SET domain and EF-hand calcium binding motif, respectively. In the present work we identified their partial polymorphisms and two SNPs, one (C1354T) at the 3′ untranslated region (UTR) of KIAA1717 and one (A345G) at the SINE (PRE-1) element of HUMMLC2B, both created/disrupted a restriction site for endonuclease Msp I. The selected pigs were genotyped at the KIAA1717 C1354T and HUMMLC2B A345G sites by means of a PCR-RFLP protocol. Significant associations were observed for the KIAA1717 C1354T polymorphic site with meat marbling (longissimus doris (p < 0.05), biceps femoris (p < 0.01)) and intramuscular fat (p < 0.01). HUMMLC2B A345G were significantly associated with meat pH (longissimus doris (p < 0.05), biceps femoris (p < 0.01)), drip loss (p < 0.01), water holding capacity (p < 0.01) and meat color value (longissimus doris (p < 0.01), biceps femoris (p < 0.05)). The objective of this study was to determine the effect of different dietary levels of sodium in diets with and without sodium chloride on mineral metabolism, including blood electrolyte levels and tibia mineralization parameters, in young turkeys (to six weeks of age). The influence of diets with a low (L), medium (M) and high (H) sodium content, at 0.34, 1.34 and 2.82 g/kg respectively, was compared. The content of chloride and potassium in turkey diets (1.7 - 5.9 and 11 g/kg, respectively) was above the recommended levels. The sodium-deficient diet L decreased the serum concentrations of sodium, chloride and phosphorus, and it increased the serum levels of calcium and magnesium in turkeys, compared with groups M and H. Relative to group L, different dietary sodium levels in groups M and H contributed to a similar increase in the body weights of birds (1.06 vs. 1.46 and 1.44 kg, p < 0.001) and in the absolute (4.60 vs. 6.83 and 6.62 g, p < 0.001) and relative tibia weight (0.42 vs. 0.46 and 0.46% body weight, p = 0.031). No significant differences were found between groups with respect to the content of ash, calcium and phosphorus in tibia dry matter. Supplemental sodium increased the bone density index (from 50.6 to 68.4 and 66.3 mg/mm in groups L, M and H, respectively, p < 0.001), the maximum bending moment (from 5.27 to 7.40 and 7.33 N/mm, p = 0.002) and the minimum breaking strength of tibia (from 0.136 to 0.191 and 0.189, p = 0.002). In conclusion, our study indicates that the applied dietary treatment with a moderate sodium level (1.34 g/kg) resulted in a rate of bird growth and tibia mineralization similar to those observed with the treatment with much higher Na content (2.82 g/kg). 相似文献

Two experiments were conducted to determine whether the decreased proportion of fast muscle fibers seen previously in 2-wk-old lambs from ewes that were dietary restricted from d 30 to 70 of gestation are still evident in older lambs and what the consequences may be in terms of growth rates and carcass composition. Throughout both experiments, ewes were fed on an individual basis according to the recommended dietary allowance throughout pregnancy relative to metabolic BW (BW(0.73)). Control groups were fed as above, and the treatment groups had their nutrient supply reduced to 50% of this recommended allowance from d 30 to 70 (Exp. 1) or d 30 to 85 (Exp. 2) of gestation, after which they were returned to the same level of nutrition as the control group. All twin lambs were kept with their dams, and at 2 wk were given access to creep. After weaning, lambs were individually housed and fed ad libitum to 24 or 17 wk of age for Exp. 1 and 2, respectively. Although not significant (P = 0.18), growth to 24 wk (Exp. 1) resulted in a small decrease in the protein content and therefore an increase in the fat:lean ratio in the carcass of lambs subjected to maternal dietary restriction. This was not apparent when animals were slaughtered at 17 wk (Exp. 2; P > 0.68). Few significant effects of maternal dietary restriction on the fiber type composition of muscles were observed. In Exp. 1 the number of fast fibers increased (P < 0.008) with no effect on slow fiber number in LM. In Exp. 2 an increase in the total number of fibers in male lambs and an increase in type II (A and B) fibers in female lambs were observed in the LM, and an increase in IIB fiber number was observed in semitendinosus (ST) muscle from male lambs. Prenatal maternal dietary restriction during the time of muscle differentiation demonstrated an increase in type IIB muscle fibers and increase in intramuscular fat; although significant, effects on subsequent carcass quality of lambs were relatively small. These data suggest that the lambs adapted to changes in muscle fiber composition previously observed at 2 wk. However, lambs in this study were well fed during postnatal growth. Whether offspring would still have been able to compensate if they had received poor nutrition postnatally and whether that failure to compensate would have influenced carcass composition remain to be determined. 相似文献
1. The aim of the study was to investigate the effects of feeding fast growing turkeys with differentiated dietary calcium (Ca) content, and the partial replacement of vitamin D3 in the feed with 25-hydroxycholecalciferol (25(OH)D3), on skeletal properties.
2. One-day-old Big-6 male turkeys (n?=?1008) were randomly divided into 4 groups, and two subgroups were created within each group. The groups were differentiated with 4 levels of Ca provision in the feed, namely 85% of the National Research Council (NRC) recommendation (Group Ca1); 95% as above (Group Ca2); 105% as above (Group Ca3); and 115% as above (Group Ca4). The first subgroup received the recommended dosage of cholecalciferol (vitamin D3 subgroup) in the feed, while in the second subgroup (Hy-D subgroup), half of the dosage of cholecalciferol was replaced with 25(OH)D3. At the ages of 4, 8, 12 and 20 weeks, 7 turkeys from each subgroup were randomly selected and killed to obtain the right tibia for densitometric, geometric and mechanical analyses.
3. This study showed advantageous effects of increased calcium supply in the diet on skeletal system properties, that were increased and produced the most desirable traits in turkeys receiving 95%, 105% and 115% of the NRC calcium recommendation. Benefits resulting from administration of 25(OH)D3 in the diet were also obtained in the skeletal formation of turkeys, and the most advantageous effects were present in the group receiving 105% of recommended dietary Ca.
4. Effects on the metabolic response of the skeleton of turkeys to manipulation of dietary calcium content and vitamin D3 source were the most evident in the groups between 4 and 12 weeks of life, and demonstrated a limited ability to induce a positive influence on bone properties at advanced stages of the production cycle by alteration of these dietary factors. 相似文献
Mature weight and rate of maturing were estimated in 177 Angus, Brahman, and reciprocal-cross cows grazing bermudagrass or endophyte-infected tall fescue over a 4-yr period to evaluate genotype x environment interactions. Data were collected every 28 d until cows were approximately 18 mo of age and then at prebreeding, postcalving, and weaning of calf. All cows with weight data to at least 42 mo of age were included in the analysis. Mature weight and rate of maturing were estimated using the three-parameter growth curve model described by Brody (1945). Data were pooled over year and analyzed by the general linear model procedure of SAS. Included in the models for mature weight and rate of maturing were the independent variables of genotype, environment, and genotype x environment interaction. There was a genotype x environment interaction (P < 0.01) for mature body weight (BW) but not for rate of maturing. Angus cows grazing fescue pastures had greater (P < 0.01) mean mature BW than Angus x Brahman cows grazing bermudagrass (611 +/- 17 vs 546 +/- 16 kg). Angus x Brahman cows grazing bermudagrass had lower (P < 0.05) mean mature BW than Brahman x Angus cows grazing bermudagrass or endophyte-infected fescue and Brahman cows grazing bermudagrass (546 +/- 16 vs 624 +/- 19, 614 +/- 22 and 598 +/- 20 kg, respectively). Brahman cows grazing endophyte-infected fescue had smaller (P < 0.05) mean mature BW than all genotype x forage combinations except for Angus x Brahman cows grazing bermudagrass. Angus cows had a smaller (P < 0.05) mean rate of maturing than Angus x Brahman and Brahman x Angus cows (0.039 +/- 0.002 vs 0.054 +/- 0.002 and 0.049 +/- 0.002%/mo, respectively), respectively, and Angus x Brahman cows had a larger (P < 0.05) mean rate of maturing than Brahman x Angus and Brahman cows (0.054 +/- 0.002 vs 0.049 +/- 0.002 and 0.041 +/- 0.002 %/mo, respectively). There was a direct breed x forage interaction (P < 0.05) for mature BW. These data suggest that the choice of breed type is important in maintaining a crossbreeding program, in that mature BW and rate of maturing are critical to the matching of animal requirements to available production resources. 相似文献
SummaryA Leptospira interrogans serogroup australis serovar lora infection in a stud farm is reported. During three successive years (1984–1986) clinical leptospirosis with a severe often rapid, fatal course was seen in 12 foals.Clinical examination revealed severe respiratory distress, depression and pyrexia. Other symptoms were diarrhea (2), jaundice (1), and an unsteady gait (1). Morphological characteristics of the disease were massive pulmonary haemorrhage and haemorrhagicthrombotic or extracapillary glomerulonephritis with tubulonephrosis and interstitial oedema. In most foals high or increasing MAT titres to serovar bratislava were found; from one foal Leptospira interrogans serovar lora was isolated.Serological examination of all 56 mares at the farm (August 1986) revealed antibodies to serovar bratislava in 64 per cent of the animals. These findings support the idea that Leptospira interrogans serovar bratislava and closely related strains (in this study serovar lora) may be adapted to and maintained by the horse population.相似文献
