Medulloblastoma (MB) is the most common malignant brain tumor of children. To identify the genetic alterations in this tumor type, we searched for copy number alterations using high-density microarrays and sequenced all known protein-coding genes and microRNA genes using Sanger sequencing in a set of 22 MBs. We found that, on average, each tumor had 11 gene alterations, fewer by a factor of 5 to 10 than in the adult solid tumors that have been sequenced to date. In addition to alterations in the Hedgehog and Wnt pathways, our analysis led to the discovery of genes not previously known to be altered in MBs. Most notably, inactivating mutations of the histone-lysine N-methyltransferase genes MLL2 or MLL3 were identified in 16% of MB patients. These results demonstrate key differences between the genetic landscapes of adult and childhood cancers, highlight dysregulation of developmental pathways as an important mechanism underlying MBs, and identify a role for a specific type of histone methylation in human tumorigenesis.  相似文献

In the current study, we describe four novel members of the 90 kDa heat shock protein (HSP90) family expressed in Japanese quail, Coturnix japonica. The coding regions of the genes, CjHSP90AA1, CjHSP90AB1, CjHSP90B1 and CjTRAP1, exhibited more than 94% similarity to their related genes in chicken. The putative proteins encoded by these quail genes contained motifs considered essential for HSP90 gene function. In addition, the predicted proteins were more similar to HSP90AA1, HSP90AB1, HSP90B1 and TRAP1 proteins expressed in vertebrates than they were to other members of the HSP90 family. Exon numbers of CjHSP90AA1 (11), CjHSP90AB1 (12) or CjTRAP1 (18) are the same as the chicken and mammalian orthologs. Furthermore, gene order in the regions surrounding CjHSP90AB1 and CjTRAP1 has been preserved, providing evidence that the genomic regions were orthologous to HSP90‐containing regions in the chicken genome. The promoter regions of the genes also contained conserved motifs identified in related genes of chicken. However, the nucleotide sequences of the 5′‐flanking region of these genes were highly polymorphic. We also found that CjHSP90AA1 exhibited a robust response to heat shock treatment. Taken together, the data suggest that CjHSP90AA1, CjHSP90AB1, CjHSP90B1 and CjTRAP1 encode orthologs of HSP90AA1, HSP90AB1, HSP90B1 and TRAP1, respectively.  相似文献

ABSTRACT:   The dynamics of vitamin A (VA) compounds in live food during enrichment were examined under different light conditions. Rotifers Brachionus plicatilis and Artemia nauplii ( Artemia ) were enriched with or without 10 mg VA palmitate (VAp) in 1 L of culture medium for 24 h under either bright (2000 lx), or dark (<1 lx) conditions. VAp, retinol (ROH), retinal (RAL) and retinoic acid (RA) contents were analyzed at 0 h (before enrichment) and at 3, 6, 12, 18 and 24 h after the onset of enrichment. Retinoid content in rotifers enriched in darkness was always higher than that enriched under light. VAp content showed two peaks at 3 and 18 h in rotifers enriched in darkness, but it showed one peak at 3 h in rotifers enriched under light. ROH and RA contents increased over the 24-h period in rotifers enriched in darkness, whereas they decreased 12 h onward in rotifers enriched under light. In Artemia , VAp and ROH contents were always higher than when enriched under the bright condition, but their dynamics showed a similar pattern in Artemia enriched under dark and bright conditions.  相似文献

Previously we showed that the relaxation modulusEt of water-saturated wood during temperature reduction maintained its initial value despite the decrease in temperature, although during temperature elevationEt showed a marked decrease. In the present study, to clarify the mechanism of relaxation during temperature elevation and reduction, Young's modulus was measured in stress relaxation experiments with changes in temperature, and relaxation behavior was simulated using a Maxwell model consisting of five elements. Furthermore, the dynamic Young's modulus and dynamic loss modulus were measured during both temperature elevation and reduction. The results obtained suggested that the unique relaxation behavior during temperature reduction was caused by decreases in Young's modulus and coefficient of viscosity (i.e., an increase in fluidity) compared with those during elevation of temperature. The decrease in Young's modulus and increase in fluidity were considered to be due to an unstable structure in wood that occurred during temperature reduction. This unstable structure probably develops in the nonequilibrium state of temperature toward a true equilibrium state. Wood should be more unstable during temperature reduction than during temperature elevation because of the decrease in molecular motion when the temperature is lowered.Part of this report was presented at the 49th annual meeting of the Japan Wood Research Society, Tokyo, April 1999  相似文献

The effects of twisting and type (single- or double-lumen) of aspiration needle on the efficiency of transvaginal ultrasound-guided ovum pick-up (US-guided OPU) were investigated in cattle. The first study using slaughterhouse ovaries revealed that twisting of the needle during follicle aspiration improved the oocyte recovery rate without deleterious effects on the attachment of cumulus layers. Vacuum pressure affected the oocyte recovery and cumulus attachment, regardless of the needle type. The needle type did not affect the oocyte recovery or cumulus attachment with an optimized vacuum pressure. In the second study, US-guided OPU was performed in live cows using two types of needles with a vacuum pressure of 75 mmHg. The needle type did not affect the oocyte recovery or cumulus attachment of the recovered oocytes. The results revealed that twisting of the needle is effective in follicle aspiration, and suggested that a single-lumen needle is as useful as a double-lumen needle for US-guided OPU in cattle.  相似文献

To assess the effects of elevated CO(2) concentration ([CO(2)]) on the photosynthetic properties around spring budbreak, we monitored the total leaf sugar and starch content, and chlorophyll fluorescence in 1-year-old needles of Sakhalin spruce (Picea glehnii Masters) seedlings in relation to the timing of budbreak, grown in a phytotron under natural daylight at two [CO(2)] levels (ambient: 360?μmol mol(-1) and elevated: 720?μmol mol(-1)). Budbreak was accelerated by elevated [CO(2)] accompanied with earlier temporal declines in the quantum yield of PSII electron transport (Φ(PSII)) and photochemical quenching (q(L)). Plants grown under elevated [CO(2)] showed pre-budbreak leaf starch content twice as high with no significant difference in Φ(PSII) from ambient-CO(2)-grown plants when compared at the same measurement [CO(2)], i.e., 360 or 720?μmol mol(-1), suggesting that the enhanced pre-budbreak leaf starch accumulation might not cause down-regulation of photosynthesis in pre-existing needles under elevated [CO(2)]. Conversely, lower excitation pressure adjusted for the efficiency of PSII photochemistry ((1?-?q(P)) F(v)'/F(m)') was observed in plants grown under elevated [CO(2)] around budbreak when compared at their growth [CO(2)] (i.e., comparing (1?-?q(P)) F(v)'/F(m)' measured at 720?μmol mol(-1) in elevated-CO(2)-grown plants with that at 360?μmol mol(-1) in ambient-CO(2)-grown plants), which suggests lower rate of photoinactivation of PSII in the elevated-CO(2)-grown plants around spring budbreak. The degree of photoinhibition, as indicated by the overnight-dark-adapted F(v)/F(m), however, showed no difference between CO(2) treatments, thereby suggesting that photoprotection during the daytime or the repair of PSII at night was sufficient to alleviate differences in the rate of photoinactivation.  相似文献

Changes in the modulus of elasticity (MOE), modulus of rupture (MOR), and stress relaxation in the radial direction of wood (hinoki:Chamaecyparis obtusa) moisture-conditioned by the adsorption process from a dry state and by the desorption process from a moisture content slightly below the fiber saturation point were investigated. The MOE and MOR of wood conditioned by the adsorption process showed significant increases during the later stages of conditioning when the moisture content scarcely changed. However, with the desorption process they did not increase as much during later stages of conditioning, though they increased during early stages of conditioning when the moisture content greatly decreased. The stress relaxation of wood decreased with an increase in the conditioning period with both the adsorption and desorption processes. These results suggest that wood in an unstable state, caused by the existing state of moisture differed from that in a true equilibrium state shows lower elasticity and strength and higher fluidity than wood in a true equilibrium state. Furthermore, the present study demonstrates that the unstable states of wood induced during the course of drying, desorption, and possibly adsorption of moisture are slowly modified as wood approaches a true equilibrium state.  相似文献

The aim of this study was to identify factors that regulate ruminal epithelial insulin‐like growth factor‐binding protein (IGFBP) expression and determine its role in rumen epithelial cell proliferation. Primary bovine rumen epithelial cells (BREC) were incubated with short‐chain fatty acids (SCFAs) at pH 7.4 or 5.6, lactate, lipopolysaccharide (LPS), insulin‐like growth factor‐I (IGF‐I), ‐II (IGF‐II), or recombinant bovine IGFBP2 (rbIGFBP2). The mRNA expression levels of IGFBP in BREC were analyzed using quantitative real‐time polymerase chain reaction (qRT‐PCR). The proliferation rate of BREC was analyzed using a WST‐1 assay. IGFBP2 gene expression tended to be lower with SCFA treatment (p < .1), and IGFBP6 gene expression was significantly lower with SCFA treatment (p < .05). IGFBP3 and IGFBP6 gene expression tended to be higher with d ‐Lactate treatment (p < .1). IGFBP3 gene expression was significantly higher (p < .05) with LPS treatment. BREC treated with IGF‐I grew more rapidly than vehicle control‐treated cells (p < .01); however, recombinant bovine rbIGFBP2 inhibited IGF‐I‐induced proliferation. IGF‐II and/or rbIGFBP2 did not affect BREC proliferation. Taken together, SCFA treatment decreased IGFBP2 and IGFBP6 expression in rumen epithelial cells, and lower expression of these IGFBP might promote rumen epithelial cell proliferation by facilitating IGF‐I.  相似文献

The modulus of elasticity and the modulus of rupture during static bending in the radial direction, and the viscoelastic properties in the radial direction in the temperature range 20°–100°C of hinoki (Chamaecyparis obtusa) swollen in organic liquids with two or more functional groups in a molecule were compared with those of wood swollen by moisture. The wood swollen in organic liquids in or near the swelling equilibrium, but not that swollen in organic liquids distant from the swelling equilibrium, showed higher moduli of elasticity and rupture than the wood swollen to a similar degree by moisture. This suggests that wood exists in an unstable state as it approaches the swelling equilibrium, rendering it highly flexible and weak. During the first viscoelastic measurements for wood swollen in various organic liquids, thermal softening was observed in 40°–60°C range and above 80°C, though this softening disappeared during the second measurement. The softening observed in the 40°–60°C range and above 80°C was thought to have been caused by the redistribution of liquid toward the equilibrium state at a higher temperature and the swelling accompanying an elevated temperature, respectively.Part of this report was presented at the 49th Annual Meeting of the Japan Wood Research Society, Tokyo, April 1999  相似文献
