Photosynthetic responses to a series of 1-min lightflecks (1,000μmol m−2 s−1) superimposed on a background with different duration (1, 5, and 10 min) and intensity (25 and 50μmol m−2 s−1) of low background photosynthetic photon flux density (PPFD) were measured in the leaves ofFagus crenata grown in a gap and understory of aFagus crenata forest in the Naeba Mountains. The two background PPFD intensities most frequently occurred in understory and gap sites respectively.
The maximum net photosynthetic rate (P
Nmax) and maximum stomatal conductance (g
smax) were higher in the gap seedlings than in the understory seedlings. However, when the background PPFD was 25μmol m−2s−1, the net photosynthetic rate (P

s) that showed the influence of lightflecks on carbon gain were higher in the understory seedlings than in the gap seedlings
when the background PPFD was 25μmol m−2 s−1. This means that understory seedling are capable of utilizing fluctuating light more efficiently under low light conditions
than the gap seedlings although the net carbon gain of single lightflecks (CG
s) in the understory seedlings was not higher than that in the gap seedlings. There were no significant differences inIS andLUE

fluctuating light at the same efficiency in both the gap and understory seedlings under natural light environments.

We transferred the linear part of a load-deflection diagram from an impact bending test of wood into a load-time diagram. In addition, we proposed a method to obtain the proportional limit from the load-time diagram. Japanese cedar (Cryptomeria japonica D. Don), hondo spruce (Picea yezoensis Carr. var.hondoensis Rehd.), hiba arbor-vitae (Thujopsis dolabrata Sieb. et Zucc.), Japanese red pine (Pinus densiflora Sieb. et Zucc.), paulownia (Paulownia tomentosa Steud.), Manchurian ash (Fraxinus mandshurica Rupr.), and Japanese evergreen oak (Quercus acuta Thunb.) were used in this study. The dimensions of each specimen were 115mm (L) * 7mm (R) * 7mm (T). The results were as follows: (1) The linear region of the load-deflection diagram in the impact bending test could be transferred with sufficient accuracy to a load-time diagram using a sine function. Artepillin C (AC), an active ingredient of Brazilian propolis, permeates intact across Caco-2 cells by transcellular passive diffusion. The permeation of AC across Caco-2 cells is as efficient as that of phenolic acids and the microbial metabolites of poorly absorbed polyphenols, which are actively absorbed by the monocarboxylic acid transporter (MCT) (Biochim. Biophys. Acta 2005, 1713, 138-144). Here, the absorption of orally administered AC in rats has been studied to evaluate its pharmacokinetics and bioavailability in vivo in comparison with those of p-coumaric acid (CA), a substrate of MCT. Rats were given 100 micromol/kg of body weight of AC or CA, and blood was subsequently collected from the portal vein and abdominal artery. AC, CA, and their metabolites were quantified by coulometric detection using HPLC-ECD. The serum concentration of intact AC and CA in the portal vein peaked at 5-10 min after administration, with a C(max) of 19.7 micromol/L for AC and 74.8 micromol/L for CA. The area under the curve (AUC) for intact AC and CA in the portal vein was calculated from the serum concentration as 182.6 and 3057.3 micromol.min.L(-1), respectively. The absorption efficiency of CA was about 17-fold higher than that of AC. Furthermore, the bioavailability of CA was about 278-fold higher than that of AC, and the ratio of AUC in the abdominal artery to AUC in the portal vein was 0.04 and 0.70, for AC and CA, respectively. Thus, AC is likely to be more susceptible to hepatic elimination than is CA. The bioactive compound of AC in vivo should be investigated further. 相似文献

The load-deflection curve for static bending and the force-time curve for impact bending of heat-treated wood were examined in detail. The effect of oxygen in air was also investigated. Sitka spruce (Picea sitchensis Carr.) was heated for 0.5–16.0h at a temperature of 160°C in nitrogen gas or air. The dynamic Young's modulus was measured by the free-free flexural vibration test, the static Young's modulus and work needed for rupture by the static bending test, and the absorbed energy in impact bending by the impact bending test. The results obtained were as follows: (1) The static Young's modulus increased at the initial stage of the heat treatment and decreased later. It decreased more in air than in nitrogen. (2) The bending strength increased at the initial stage of the heat treatment and decreased later. It decreased more in air than in nitrogen. (3) The work needed for rupture decreased steadily as the heating time increased. It decreased more in nitrogen than in air. It is thought that heat-treated wood was more brittle than untreated wood in the static bending test because W12 was reduced by the heat treatment. This means that the main factors contributing to the reduction of the work needed for rupture were viscosity and plasticity, not elasticity. (4) The absorbed energy in impact bending increased at the initial stage of the heat treatment and decreased later. It decreased more in air than in nitrogen. Anthocyanin synthesis began in the skin of ‘Olympia’ (Vitis labruscana Bailey) grape when the berry growth reached a late stage in the second sigmoid growth-curve. During berry development, shikimate NADP oxidoreductase (SORase, EC.1.1.1.25), phenylalanine ammonia-lyase (PAL, EC.4.3.1.5), and chalcone flavanone isomerase (CHFI, EC.5.5.1.6) were investigated in the skin. These enzyme activities were high during early development of the young berry, but rapidly decreased thereafter, and were activated again when rapid anthocyanin synthesis subsequently occurred. When PAL from the young berry was analyzed by isoelectricfocusing polyacrylamide gel electrophoresis (IEF-PAGE), it showed a single activity band with a pI value of approximately 5.4, whereas PAL from the ripe berry showed two activity bands with pIs of 5.4 and 6.5. Thus, SORase, PAL, and CHFI seem to be involved in anthocyanin accumulation simultaneously, and newly synthesized isozyme of PAL may play a key role in pigment formation in ‘Olympia’ skin. Recently, we introduced an organoid-based chemical carcinogenesis model using mouse normal tissue-derived organoids. In the present review article, the histopathological and immunohistochemical characteristics of mouse normal tissue-derived organoids and tumors derived from these organoids after their in vitro treatment with genotoxic carcinogens and injection into nude mouse are reviewed. In organoids treated in vitro with genotoxic carcinogens, we confirmed macroscopic tumorigenicity and histopathological findings, including neoplastic characteristics, such as multilayered epithelia and/or invasion of epithelia into the surrounding interstitium. In contrast glandular/cystic structures with monolayered epithelia were clearly demarcated from the surrounding Matrigel/interstitium in the untreated control groups. In addition to macroscopic tumorigenicity, these microscopic epithelial changes, which are characteristic of the early stages of carcinogenesis, are included in the requirements for carcinogenicity-positive judgement of the organoid-based carcinogenesis model. Immunohistochemistry of cytokeratins (CKs), used to determine the origin of epithelia and distribution of extraductal invasive lesions, or oncogenic kinases, which reflect molecular activation in epithelia following chemical treatment, is helpful for accurate diagnosis and molecular evaluation in the early stages of carcinogenesis. We examined various methods to enhance the accessibility of intracytoplasmic sperm injection (ICSI) technology to more users by making the technique easier, more efficient, and practical. First, the methods for artificially removing the mouse sperm tail were evaluated. Trypsin treatment was found to efficiently remove the sperm tails. The resultant sperm cells had a lower oocyte activation capacity; however, the use of activated oocytes resulted in the same fecundity as that of fresh, untreated sperm. Pre-activated oocytes were more resistant to physical damage, showed higher survival rates, and required less time per injection. Testing this method in rats yielded similar results, although the oocyte activation method was different. Remarkably, this method resulted in higher birth rates of rat progeny than with conventional methods of rat ICSI. Our method thereby streamlines mouse and rat ICSI, making it more accessible to laboratories across many disciplines. 相似文献

Two kinds of FeIFN-alpha consisting of 166 amino acids (aa) and 171 aa were expressed in Escherichia coli, and the purified proteins were tested for antiviral activity on homologous and heterologous animal cells. Crude FeIFN induced in feline cells revealed antiviral activity on both homologous and heterologous animal cells. In contrast, both types of recombinant FeIFN-alpha revealed antiviral activity only on the feline cells. All of the FeIFN-alpha subtypes showed high activity to vesicular stomatitis virus, and the three species of feline viruses belonging to different families. 相似文献
