ABSTRACT:   In order to find out the role of sodium bicarbonate (NaHCO3) on the initiation of sperm motility in the Japanese eel Anguilla japonica , interactions were investigated between NaHCO3 and various reagents (K+ channel blocker 4-aminopyridine [4-AP], ammonium chloride [NH4Cl], sodium acetate and calcium chloride [CaCl2]) that could regulate internal factors (intracellular K+, intracellular pH [[pH]i] and intracellular Ca2+) in sperm motility. Contradictory effects of NaHCO3 were observed (i.e. an inhibitory effect when 4-AP was absent and a promoting effect when 4-AP was present). Sodium bicarbonate inhibited the initiation of sperm motility in the Japanese eel. However, NaHCO3 restored the motility of immotile sperm that 4-AP inhibited. The inhibitory effect of NaHCO3 disappeared with the addition of NH4Cl, which raised [pH]i, but the promoting effect was not affected by [pH]i. Although NaHCO3 recovered motility in the presence of 4-AP, this recovery was also observed with the addition of CaCl2 instead of NaHCO3. In the initiation of sperm motility in the Japanese eel, two roles for NaHCO3 are suggested: an inhibitory role relating to the regulation of [pH]i and a promoting role relating to the uptake of another initiation factor, which could be Ca2+.  相似文献

The authors developed a computer program of transient predictive calculation method on underground ventilation network. The program approximated a field data set of seasonal cyclic changes of air Temperature and humidity at the inlet to the sine curves, and included sensible heat conduction from rock to air current. The temperature of rock around and airway was calculated by finite difference method. The prediction method can be used to calculate the variation of flow rates, temperatures and humidities of air flow in the airways along underground network.  相似文献

Mammalian tooth crowns have precise functional requirements but cannot be substantially remodeled after eruption. In developing teeth, epithelial signaling centers, the enamel knots, form at future cusp positions and are the first signs of cusp patterns that distinguish species. We report that ectodin, a secreted bone morphogenetic protein (BMP) inhibitor, is expressed as a "negative" image of mouse enamel knots. Furthermore, we show that ectodin-deficient mice have enlarged enamel knots, highly altered cusp patterns, and extra teeth. Unlike in normal teeth, excess BMP accelerates patterning in ectodin-deficient teeth. We propose that ectodin is critical for robust spatial delineation of enamel knots and cusps.  相似文献

By using an ionic liquid of 1-butyl-3-methylimidazolium bis(trifluoromethanesulfonyl)imide, we uniformly dispersed single-walled carbon nanotubes (SWNTs) as chemically stable dopants in a vinylidene fluoride-hexafluoropropylene copolymer matrix to form a composite film. We found that the SWNT content can be increased up to 20 weight percent without reducing the mechanical flexibility or softness of the copolymer. The SWNT composite film was coated with dimethyl-siloxane-based rubber, which exhibited a conductivity of 57 siemens per centimeter and a stretchability of 134%. Further, the elastic conductor was integrated with printed organic transistors to fabricate a rubberlike active matrix with an effective area of 20 by 20 square centimeters. The active matrix sheet can be uniaxially and biaxially stretched by 70% without mechanical or electrical damage. The elastic conductor allows for the construction of electronic integrated circuits, which can be mounted anywhere, including arbitrary curved surfaces and movable parts, such as the joints of a robot's arm.  相似文献

Methanol extracts of 11 kinds of commonly available vegetables were examined for hydroxyl radical scavenging potency using the bleomycin-Fe method. In this method, the iron ion and bleomycin in water form hydroxyl radicals, and the scavenging activity is monitored by the modified thiobarbituric acid method. All extracts showed scavenging capacity, even though the activity of some of them was lower than that of l-ascorbic acid. Those vegetables were classified into three groups according to their activity, groups showing strong activity, moderate activity, and weak activity, as compared to the activity of l-ascorbic acid at the same concentration. Among them, the methanol extract of radish sprout (Japanese name "kaiware-daikon") exhibited the highest potency (1.8 times as l-ascorbic acid). Then, we investigated the constituents of the methanol extract of radish sprout and the contribution to the overall activity of each compound by examining their activity. As the result, several kinds of sinapinic acid esters and flavonoids were isolated with high radical scavenging potency, which must contribute substantially to the activity.  相似文献

A novel flavonol glycoside, 7-O-methylmearnsitrin (7,4'-O-dimethylmyricetin 3-O-alpha-L-rhamnopyranoside), and myricetrin, kaempferol 3-O-alpha-L-rhamnopyranoside, europetin 3-O-alpha-L-rhamnoside, and 7-O-methyl quercetin 3-O-alpha-L-rhamnopyranoside were isolated from the leaves of Sageretia theezans, and their chemical structures were identified by spectroscopic analyses including two-dimensional NMR (HSQC, HMBC). Whereas myricetrin, europetin 3-O-alpha-L-rhamnoside, and 7-O-methylquercetin 3-O-alpha-L-rhamnopyranoside showed stronger activities than ascorbic acid and alpha-tocopherol, 7-O-methylmearnsitrin showed very weak antioxidant activities by ESR and LDL oxidation inhibition tests.  相似文献

The developmental potential of nuclei from a bovine mammary epithelial cell line (BMEC) in nuclear transfer was investigated. For nuclear transfer donors, BMEC cells (passage 15) were cultured for 4–5 days after seeding at cell densities of 1.0 × 105 cells/cm2 (high‐density group) or 0.8 × 104 cells/cm2 (low‐density group). First, the effective electric stimulation for fusion of enucleated oocytes with BMEC cells was examined. Fusion rates reached maximum with two DC pulses of 30 V/150 µm for 20 µs in the high‐density group and with two DC pulses of 25 V/150 µm for 10 µs in the low‐density group. The fusion rate (37.5%) in the high‐density group was significantly (P < 0.005) lower than in the low‐density group (71.4%). Second, the in vitro developmental potential of nuclear transfer embryos derived from BMEC cells was examined. In the high‐density and low‐density groups, 18.8% and 24.1% of fused oocytes, respectively, developed to blastocyst stage. The results of this study indicate that nuclei from BMEC cells support the development of nuclear transfer embryos to the blastocyst stage and that the efficiency of oocyte–cell fusion is affected by the culture conditions of the donor BEMC cells before nuclear transfer.  相似文献

Sarcoplasmic protein content decreased significantly and myosin heavy chain was degraded gradually during spawning migration of chum salmon (Oncorhynchus keta). Acid and neutral proteinase activities increased significantly during spawning migration. These proteinase activities were higher in females than in males. High levels of acid proteinase were not caused by the injury of lysosomal membranes. Considering the physiological states of chum salmon, neutral proteinase activity might be related to the degradation of muscle protein. As the changes in serum sex steroids were similar to those in the protease activities during spawning migration, it was considered that high levels of protease activity during spawning migration were closely related to the serum levels of androgens.  相似文献

The larvicidal activity of a series of N-2,6-difluoro- and N-2,6-dichlorobenzoyl-N′-(4-substituted phenyl)ureas against nondiapause larvae of the rice stem borer, Chilo suppressalis Walker, was measured by topical application and oral administration methods under conditions with and without piperonyl butoxide as an inhibitor of oxidative metabolism. The effects of substituents at the anilide moiety on the larvicidal activity were analyzed quantitatively using physicochemical substituent parameters and regression analysis. The results indicate that the oxidative metabolism in the larval body which is favored by electron-donating substituents is significant in determining the activity. The activity, when the metabolic factor is eliminated, is enhanced by electron-with-drawing and hydrophobic substituents and lowered by bulky groups. The inhibitory activity against new cuticle formation of the same series of compounds was also measured using cultured integument of the rice stem borer diapause larva. The comparison of the quantitative analyses between larvicidal and integument-level activities shows that inhibition of cuticular development is the most important factor governing larvicidal activity.  相似文献
