Handling practices of specimens may affect the sensitivity or specificity of diagnostic tests. In this study, as part of the Voluntary Iowa Bovine Viral Diarrhea Virus Screening Project held in 2006, 2 sample-handling practices were evaluated to determine how they affect the sensitivity and specificity of the antigen-capture enzyme-linked immunosorbent assay (ACE) for bovine viral diarrhea virus (BVDV). The null hypotheses investigated were 1) that maintenance of samples at room temperature would not be associated with decreased sensitivity, and 2) that continued use of a single pair of ear notchers would not be associated with cross-contamination of virus from 1 notch to another and reduce specificity. These hypotheses were tested in 2 studies by collecting known positive and negative samples and giving groups of samples different treatments. The first study used ACE on 4 groups of skin samples, all from a known-positive animal. Each group was subjected to different lengths of time at room temperature, from 24 to 96 hours at 24-hour intervals. No difference in test results was found between specimens subjected to different lengths of time at room temperature. The second study tested the effects of giving 3 different treatments to an ear notcher in between sample collecting (water rinse, Nolvasan solution rinse, or no treatment) on ACE results. No effect on sensitivity or specificity of ACE was observed. No difference in test results was found between the 3 ear-notcher treatment groups. The sample handling practices evaluated appeared to have little impact on test sensitivity or specificity of ACE for BVDV. 相似文献

The aim of this study was to describe animal production on small farms in the Kaski district of Nepal, with the goal of identifying areas where animal health and productivity could be improved. Eighty-five randomly selected farms from four different Village Development Committees were visited. Farmers were interviewed and premises and animals visually inspected on all farms. Feed samples were collected from a subset of farms. The most commonly kept species were water buffalo (used for milk and meat), cattle (used for milk and labor), and goats (used for meat). Average milk production levels were 4.7?kg/day for water buffalo and 1.9?kg/day for cattle. All animals were milked manually, no calves were weaned, and only one farm practiced artificial insemination. A majority of cattle and goats had access to pasture, and a majority of farms fed their working or producing animals concentrates; however, nutritional input was insufficient in terms of energy, protein, and micronutrient content to increase levels of production. Goat-raising was the most profitable endeavor, followed by water buffalo and cattle. We conclude that animals have the potential to contribute significantly to improved livelihoods of farmers in terms of both income generation and non-tangible benefits. However, we found that significant constraints on animal production exist, including insufficient nutritional levels and a lack of preventative care resulting in animal disease. Furthermore, cultural considerations reflecting attitudes toward cattle shape farming in ways that may limit production. Ultrasonography is a sensitive and specific screening method for assessing the adrenal glands. The upper limit of the normal adrenal gland width is used as 7.5 mm. It is not known if adrenal gland width remains consistent with body weight. A reliable criterion of adrenal gland width in small breed dogs should be established. Small breed dogs with body weights of less than 10 kg were divided into two groups: 189 normal dogs and 22 dogs with pituitary-dependent hyperadrenocorticism (PDH). A retrospective study was conducted on dogs seen between January 1, 2006, and February 10, 2008. One hundred eighty-nine dogs of 14 different small breeds were enrolled in the normal adrenal gland group; the median gland width was 4.20 mm. Twenty-two dogs were in the PDH group; the median gland width was 6.30 mm. The cut-off value between normal adrenal glands and PDH was 6.0 mm. This figure gave a sensitivity and specificity of 75 and 94%, respectively, for detecting PDH. The adrenal gland appeared as a peanut shape with homogeneous hypoechoic parenchyma in normal dogs and in most dogs with PDH as well. This study was performed in a large population of small breed dogs and suggests that the normal adrenal gland size in small breed dogs is smaller than previously reported. We believe that a cut-off of 6.0 mm may be used as the criterion for differentiating a normal adrenal gland from adrenal hyperplasia. 相似文献

We sequenced the hemagglutinin (H) genes from four canine distemper virus (CDV) isolates obtained from three dogs and a marten in Korea. These sequences were included in subsequent H gene-focused phylogenetic tree analysis of 89 CDV strains. This analysis revealed eight clades designated as EU1, EU2, EU3, NA1, NA2, Asia 1, Asia 2 and Vaccine. Three of the Korean isolates (97Jindo, 98Marten and 07D111) occurred in the Asia 2 group that also contains many Japanese CDV strains isolated in 1998. The remaining Korean strain (07Q72) fell into the Asia 1 group. The 21 H protein sequences of 25 Asia 1 strains are generally predicted to bear nine potential N-linked glycosylation sites. In contrast, the 9 H protein sequences of 12 Asia 2 strains had eight potential N-linked glycosylation sites. The remaining strains had six (98Marten and 07D111) and seven (97Jindo) potential N-linked glycosylation sites. 相似文献

Recombinant human epidermal growth factor (rhEGF) stimulates the proliferation and migration of epithelial cells in human cell culture systems and animal models of partial-thickness skin wounds. This study investigated the effect of a topical rhEGF ointment on the rate of wound healing and skin re-epithelialization in a rat full thickness wound model, and verified whether or not the rhEGF treatment affected both myofibroblast proliferation and collagen synthesis in the dermis. When rhEGF (10 µg/g ointment) was applied topically twice a day for 14 days, there was significantly enhanced wound closure from the 5th to the 12th day compared with the control (ointment base treatment) group. A histological examination at the postoperative 7th day revealed that the rhEGF treatment increased the number of proliferating nuclear antigen immunoreactive cells in the epidermis layer. In addition, the immunoreactive area of alpha-smooth muscle actin and the expression of prolyl 4-hydroxylase were significantly higher than those of the control group. Overall, a topical treatment of rhEGF ointment promotes wound healing by increasing the rate of epidermal proliferation and accelerating the level of wound contraction related to myofibroblast proliferation and collagen deposition. 相似文献
