The presence of genetically modified organisms (GMOs) in food and feed products is subject to regulation in the European Union (EU) and elsewhere. As part of the EU authorization procedure for GMOs intended for food and feed use, reference materials must be produced for the quality control of measurements to quantify the GMOs. Certified reference materials (CRMs) are available for a range of herbicide- and insect-resistant genetically modified crops such as corn, soybean, and cotton. Here the development of the first CRM for a GMO that differs from its non-GMO counterpart in a major compositional constituent, that is, starch, is described. It is shown that the modification of the starch composition of potato (Solanum tuberosum L.) tubers, together with other characteristics of the delivered materials, have important consequences for the certification strategy. Moreover, the processing and characterization of the EH92-527-1 potato material required both new and modified procedures, different from those used routinely for CRMs produced from genetically modified seeds. 相似文献

The objective of the study is to map citizen attitudes towards pig meat production systems, and to investigate whether these attitudes associate with pork and pork product consumption. A conjoint experiment was carried out with empirical data collected from 1931 individuals in four European countries with higher-than-average per capita meat consumption (Belgium, Denmark, Poland and Germany). The experiment was based on the following pig farming characteristics: herd size, housing and floor type, efforts to reduce the impact of the production system on the environment, animal feed designed for producing pork with specific fat content, and finally the preferred quality characteristics of the pork end-product. The results of the conjoint analysis were used for a subsequent cluster analysis in order to identify European citizen clusters. Respondents' socio-demographic profile, attitudes towards issues that are expected to influence the way people evaluate pig meat production systems, and consumption frequency of various pork products are used as background information to identify segment profiles. Pork appears to play a substantial role as a part of the diet for most respondents in the four countries of this study. The results of the conjoint experiment at the sample level show that people assigned most importance to animal and environmental well-being as criteria to discriminate between “good” and “bad” pig production practices, despite the fact that their attitudes towards environmental protection, animal welfare and industrial food production were only moderately strong. Moreover, the results of the cluster analysis allowed small-sized, clear-cut clusters of citizens to appear, which pay attention to specific pig farming attributes. In conclusion, attitudes towards environment and nature, animal welfare and the need for an environment-friendly food production were indeed related to citizens' specific attitudes towards pig farming at the cluster level. However, the relationship between citizenship and consumption behaviour was found to be weak. What people think in their role as citizens related to pig production did not appear to significantly influence their pork consumption choices. 相似文献

Protein dynamics are essential for protein function, and yet it has been challenging to access the underlying atomic motions in solution on nanosecond-to-microsecond time scales. We present a structural ensemble of ubiquitin, refined against residual dipolar couplings (RDCs), comprising solution dynamics up to microseconds. The ensemble covers the complete structural heterogeneity observed in 46 ubiquitin crystal structures, most of which are complexes with other proteins. Conformational selection, rather than induced-fit motion, thus suffices to explain the molecular recognition dynamics of ubiquitin. Marked correlations are seen between the flexibility of the ensemble and contacts formed in ubiquitin complexes. A large part of the solution dynamics is concentrated in one concerted mode, which accounts for most of ubiquitin's molecular recognition heterogeneity and ensures a low entropic complex formation cost. 相似文献

Purpose
Many agricultural and brownfield soils are polluted and more have become marginalised due to the introduction of new, risk-based legislation. The European Environment Agency estimates that there are at least 250,000 polluted sites in the member states that require urgent remedial action. There is also significant volumes of wastewaters and dredged polluted sediments. Phytotechnologies potentially offer a cost-effective in situ alternative to conventional technologies for remediation of low to medium-contaminated matrices, e.g. soils, sediments, tailings, solid wastes and waters. For further development, social and commercial acceptance, there is a clear requirement for up-to-date information on successes and failures of these technologies based on evidence from the field. Introduction
Given the complex interactions of re-suspension processes and bioavailability of sediment-bound pollutants such as dioxin-like chemicals, there is need for a better integrative understanding of the cause–effect relationship of these pollutants. Currently, the majority of studies investigating potential risks of these chemicals only focus on characterizing sediment extracts via in vitro bioassays, thereby disregarding bioavailability, uptake, metabolism, and elimination rates of these compounds in vivo. To determine to which extent mechanism-specific effects in vitro reflect possible adverse effects in vivo, the research project dioRAMA, involving partnership between the Institute for Environmental Research of RWTH Aachen University and the Department Biochemistry/Ecotoxicology of the German Federal Institute of Hydrology, was established.Methods
Animals from an elevated trophic level—common roach (Rutilus rutilus)—will be exposed to sediments from two major German rivers. Exposure will be performed in a system that enables a concurrent monitoring of environmental parameters. In parallel, in vitro studies will be conducted to determine dioxin-like potentials of sediment and fish extracts from the in vivo exposure experiments using different cell lines with varying endpoints. Moreover, extract fractionation procedures, using the strategy of effect-directed analysis, will enable the detection of specific contaminant groups responsible for the biological activity observed.Conclusion
A closer interconnection between applied ecotoxicological science and regulatory needs will facilitate the improved assessment of dioxin-like compounds in sediment and biota. BACKGROUND: Spray volume can influence the amount of free water on the leaf surface and subsequently the ability of entomopathogenic nematodes (EPNs) to move. In this study, an investigation was made of the effect of spray volume (548, 730 and 1095 L ha−1) on the deposition, viability and infectivity of EPNs against Galleria mellonella on savoy cabbage, cauliflower and leek. RESULTS: Increasing spray volume decreased nematode deposition on 7.1 cm2 leek leaf discs at a 15° angle with the spray nozzle. Although the number of living nematodes observed on leek after 240 min of exposure was not significantly different between the low-volume application (548 L ha−1) and the high-volume application (1095 L ha−1), a greater infectivity was obtained in the latter application. The higher number of droplets deposited on the leek discs in the high-volume application may have stimulated nematode movement. No significant effect of spray volume was observed on the relative deposition of Steinernema carpocapsae on the bottom side of cauliflower and savoy cabbage leaf discs. In spite of the low S. carpocapsae deposition on the bottom side of the savoy cabbage discs, high infectivity was obtained against G. mellonella. Using the lowest spray volume on savoy cabbage, infectivity decreased with increasing exposure time, while infectivity was not affected by exposure time when a spray volume of 730 L ha−1 or more was used. CONCLUSION: Spray volume is an important application parameter, as it affects nematode infectivity. The purpose of this study was to assess the feasibility of cone beam computed tomography (CBCT) in imaging dogs and cats for diagnostic dental veterinary applications. CBCT scans of heads of six dogs and two cats were made. Dental panoramic and multi-planar reformatted (MPR) para-sagittal reconstructions were created using specialized software. Image quality and visibility of anatomical landmarks were subjectively assessed by two observers. Good image quality was obtained for the MPR para-sagittal reconstructions through multiple teeth. The image quality of the panoramic reconstructions of dogs was moderate while the panoramic reconstructions of cats were poor since the images were associated with an increased noise level. Segmental panoramic reconstructions of the mouth seem to be useful for studying the dental anatomy especially in dogs. The results of this study using human dental CBCT technology demonstrate the potential of this scanning technology in veterinary medicine. Unfortunately, the moderate image quality obtained with the CBCT technique reported here seems to be inferior to the diagnostic image quality obtained from 2-dimensional dental radiographs. Further research is required to optimize scanning and reconstruction protocols for veterinary applications. 相似文献
