Compared to other major food crops, progress in potato yield as the result of breeding efforts is very slow. Genetic gains
cannot be fixed in potato due to obligatory out-breeding. Overcoming inbreeding depression using diploid self-compatible clones
should enable to replace the current method of out-breeding and clonal propagation into an F1 hybrid system with true seeds. This idea is not new, but has long been considered unrealistic. Severe inbreeding depression
and self-incompatibility in diploid germplasm have hitherto blocked the development of inbred lines. Back-crossing with a
homozygous progenitor with the Sli gene which inhibits gametophytic self-incompatibility gave self-compatible offspring from elite material from our diploid
breeding programme. We demonstrate that homozygous fixation of donor alleles is possible, with simultaneous improvement of
tuber shape and tuber size grading of the recipient inbred line. These results provide proof of principle for F1 hybrid potato breeding. The technical and economic perspectives are unprecedented as these will enable the development of
new products with combinations of useful traits for all stakeholders in the potato chain. In addition, the hybrid’s seeds
are produced by crossings, rendering the production and voluminous transport of potato seed tubers redundant as it can be

Many oomycete and fungal plant pathogens are obligate biotrophs, which extract nutrients only from living plant tissue and cannot grow apart from their hosts. Although these pathogens cause substantial crop losses, little is known about the molecular basis or evolution of obligate biotrophy. Here, we report the genome sequence of the oomycete Hyaloperonospora arabidopsidis (Hpa), an obligate biotroph and natural pathogen of Arabidopsis thaliana. In comparison with genomes of related, hemibiotrophic Phytophthora species, the Hpa genome exhibits dramatic reductions in genes encoding (i) RXLR effectors and other secreted pathogenicity proteins, (ii) enzymes for assimilation of inorganic nitrogen and sulfur, and (iii) proteins associated with zoospore formation and motility. There is a lack of scientific evidence for objective evaluation of neck and back musculoskeletal sensitivity in horses, although pressure algometry has been described as an objective tool to quantify musculoskeletal responses by mechanical nociceptive threshold (MNT) values. This study aimed to evaluate the use of pressure algometry for objectively quantifying the effect of diagnostic palpation applied by physiotherapists on the musculoskeletal function of the equine neck and back. The inter-examiner repeatability of animal physiotherapists was tested, and their subjective clinical scores for the vertebral column area were objectively compared with MNT values measured at the same locations to investigate the potential clinical implementation of the pressure algometer in daily equine rehabilitation practice. Six adult Dutch Warmblood riding school mares were randomly assigned to an experimental or a control group. The MNT of all horses was measured on 35 predefined sites on the vertebral column in the morning and in the evening of the same day. In the experimental group (n = 3), neck and back surface “temperature”, “pain”, “muscle tone”, and passive “mobility” were scored through palpation by three certified physiotherapists and related to MNT measurements at the same vertebral column locations. Agreement between the physiotherapists was determined from Spearman's rank correlation coefficients (P < .05). These correlation coefficients showed a significant agreement between the scores of individual physiotherapists and with objective MNT measurements. The three physiotherapists agreed best in their subjective gradings of “pain”, but less for “temperature” and “muscle tone”, and least for “mobility”. There was also a significant difference in MNT between individual horses. The physiotherapeutic diagnostic intervention did not significantly alter the MNT of the experimental group compared with the control group. There was a significant difference, however, between morning (7.4 kg/cm2) and evening (6.9 kg/cm2) MNT-measurements within the combined group (n = 6, P < .05). In conclusion, a pressure algometer proved to be a useful tool to objectively monitor the palpation of individual Warmbloods by individual physiotherapists. The correlation of their scores to the objective MNT measurements elucidated that there were differences on which scale (“pain”, “temperature”, “muscle tone”, “mobility”) they merely relied upon in their palpation. Significant effects of physiotherapeutic diagnostic palpation on MNT, however, were not found. The lower MNT of the horses at the second trial in the evening could be a sensitization of the measurement location because of bruising, a learning effect of the horses, or a diurnal fluctuation. The use of pressure algometry has both a potential to quantify clinical neck and back musculoskeletal sensitivity in horses possibly leading to dysfunction, as well as to objectively evaluate treatment results. Repeated measurements on the same day and on the same location along the vertebral column may influence absolute MNT values. The increasing demand for triticale as food, feed, and fuel has resulted in the availability of cultivars with different grain quality characteristics. Analyses of triticale composition can ensure that the most appropriate cultivars are obtained and used for the most suitable applications. Near‐infrared (NIR) spectroscopy is often used for rapid measurements during quality control and has consequently been investigated as a method for the measurement of protein, moisture, and ash contents, as well as kernel hardness (particle size index [PSI]) and sodium dodecyl sulfate (SDS) sedimentation from both whole grain and ground triticale samples. NIR spectroscopy prediction models calculated using ground samples were generally superior to whole grain models. Protein content was the most effectively modeled quality property; the best ground grain calibration had a ratio of the standard error of test set validation to the standard deviation of the reference data of the test set (RPDtest) of 4.81, standard error of prediction (SEP) of 0.52% (w/w), and r2 of 0.95. Whole grain protein calibrations were less accurate, with optimum RPDtest of 3.54, SEP of 0.67% (w/w), and r2 of 0.92. NIR spectroscopy calibrations based on direct chemical reference measurements (protein and moisture contents) were better than those based on indirect measurements (PSI, ash content, and SDS sedimentation). Fourteen United States (U.S.) seed potato certification agencies surveyed all U.S. seed potato growing areas for presence of the potato spindle tuber viroid (PSTVd). The survey included general surveillance, which involved searching for the occurrence of PSTVd in state seed potato certification records from 1990 through 2000, and a field survey, which involved testing selected crops for PSTVd infection by nucleic acid dot blot hybridization during 1999 through 2001. No PSTVd incident was documented in any of the state certification records, nor was PSTVd detected in the field surveys. All U.S. seed-growing areas were determined to be free of PSTVd. It is concluded that PSTVd has been eradicated and freedom from potato spindle tuber viroid has been successfully maintained in all of the seed potato growing areas in the United States. 相似文献

Lamellarins, a family of hexacyclic pyrrole alkaloids originally isolated from marine invertebrates, display promising anti-tumor activity. They induce apoptotic cell death through multi-target mechanisms, including inhibition of topoisomerase I, interaction with DNA and direct effects on mitochondria. We here report that lamellarins inhibit several protein kinases relevant to cancer such as cyclin-dependent kinases, dual-specificity tyrosine phosphorylation activated kinase 1A, casein kinase 1, glycogen synthase kinase-3 and PIM-1. A good correlation is observed between the effects of lamellarins on protein kinases and their action on cell death, suggesting that inhibition of specific kinases may contribute to the cytotoxicity of lamellarins. Structure/activity relationship suggests several paths for the optimization of lamellarins as kinase inhibitors. 相似文献

The occurrence of lameness in dairy cattle is of increasing importance in herd health management and herd productivity. Current practice, involves visual observation by human experts to score cow's locomotion in order to estimate the lameness in the herd. The trackway defined as “hind hoof compared to fore hoof position” is one of the main components to score the locomotion. However, because of the time-consuming observation method, lame cows are undiagnosed until the problem has become severe. Up till now there is no automatic (visual) method for detecting lameness in dairy cattle. The objective of our study was to make an automatic system for continuous on-farm detection and prediction of lameness in the farm by using vision techniques. The current focus was on demonstrating the possibility of capturing cow's hoof locations by vision and strong correlation between automatically calculated hoof trackway and visual locomotion scores. Fifteen selected lactating cows were scored visually by four trained observers at the Gent University Research Farm. Scoring varied from 1 (normal walking) to 5 (extremely lame). Side-view images with resolution of 1024 × 768 pixels were recorded when cows passed an experimental set-up freely. Recorded videos were split into sequences of bitmap images. After background subtraction, binary image operations, calibration and hoof separation, the trackway information containing hoof location in the real world and its related time in the video was calculated. The accuracy of automatically captured results was checked by comparing with the output from manually labeled hoof locations. The mean correlation coefficient of all measurements was 94.8%. Hence, the results suggest that the automatic method by vision analysis is feasible to present the cows’ real locomotion situations. The first result showed a positive linear relationship between cows’ trackways overlap and locomotion scores by human visualization. The Sistan Delta in Iran is located at the end of a closed basin with nearly 100% of the supply coming from Afghanistan. This
supply is supporting irrigated agriculture in the area and is the source for the lake system around the delta. These Hamoun
lakes are ecological very valuable wetlands; a number of them are registered as Ramsar sites. The Iranian government tries
to improve the living conditions of the people in the area, among others by providing infrastructure for irrigated agriculture.
Further development of the irrigated area will mean less water for the Hamouns with resulting lower average water coverage
of the lakes. This will not only endanger the ecosystem that the Hamouns support but also the livelihoods of the people that
depend on the goods and services that the lakes provide. This paper describes a study that has been carried out to support
decision making on potential development schemes in the delta. The non-availability of data from Afghanistan requires the
development of various tools and the use of remote sensing techniques to enable to make estimates for the river flow that
Iran can expect from Afghanistan. An IWRM approach has been used for the balancing of interests involved. Some preliminary
