On four occasions, four horses with heaves and four horses with small airway inflammatory diseases inhaled 0.9% saline based aerosol mixtures with or without lipopolysaccharides (LPS). Prior to the first saline and LPS inhalation, horses were untreated, while three and a half days prior to the third and forth inhalation horses had received 0.8 μ g/kg clenbuterol intravenously twice daily. The messenger RNA (mRNA) expression of tumour necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-4, IL-6, IL-8, IL-10 and interferon- γ (IFN- γ) was investigated by RT-PCR, all of which were expressed in the white blood cells of samples collected. Inhalation of LPS only changed the cytokine expression profile of IL-10, IL-4 and TNF-α mRNA which were higher after challenge with LPS. However in those horses that were treated with clenbuterol the LPS-induced IL-10 mRNA expression was shown to be suppressed. Further changes in IL-4 and TNF-α were not significant. Thus the results of this study indicated that clenbuterol can modulate the expression of IL-10 mRNA in peripheral white blood cells in those horses with small airway diseases that have been exposed to LPS. van den Hoven, R., Duvigneau, J.C., Hartl, R.T. and Gemeiner, M., 2006. A number of parameters linked to storage of potatoes were evaluated with regard to their potential to influence the acrylamide formation in French fries. Acrylamide, which is a potential human carcinogen, is reported to be formed during the frying of potatoes as a result of the reactions between asparagine and reducing sugars. This study was conducted using three potato varieties (Bintje, Ramos, and Saturna) typically used in Belgium, The Netherlands, and the northern part of France for French fry and crisp production. Saturna, mainly used in crisp production, appeared to be the least susceptible for acrylamide formation during frying. Especially storage at low temperatures (4 degrees C) compared to storage at 8 degrees C seemed to enhance acrylamide formation due to a strong increase in reducing sugars caused by low-temperature storage. Because of the reversible nature of this physiological reaction, it was possible to achieve a significant reduction of the reducing sugars after a reconditioning of the cold-stored potatoes for 3 weeks at 15 degrees C. All changes in acrylamide concentrations could mainly be explained by the reducing sugar content of the potato (R2 = 0.84, n = 160). This means that, by ensuring a low reducing sugar content of the potato tuber, the risk for acrylamide formation will largely be reduced. Finally the use of a sprout inhibitor did not influence the composition of the potato, and thus acrylamide formation was not susceptible to this treatment.  相似文献

The question was examined under which conditions the water extraction rate of plant roots in the field can be limited by water transport to the roots. For this purpose we used a numerical solution of the single root model. Scenario calculations were carried out in order to investigate the general model behaviour. A sensitivity analysis showed that initial volumetric water content and root length density are of greater importance than root diameter in determining the maximum water transport rate to the roots. Data from a field experiment were taken, describing root length density, volumetric water content and water uptake rates under oats (Avena saliva L.) and faba beans (Vicia faba L.) as model input parameters. With this data the model calculated the water content difference between the bulk soil and the root surface which is necessary to induce a water flow to the roots matching the observed water uptake rate. Root length densities below the grain legume crop faba beans are one order of magnitude lower compared to that of the cereal crop oats. The therefore higher specific water influx rates of faba beans roots resulted in a higher decrease in water content near the root surface. However, water uptake by faba beans was controlled by the water flow towards the roots probably only in deeper soil layers with very low root length density. For the given conditions no transport limitation of water uptake was calculated, when rooting densities were higher than about 0.1 cm.cm?3.  相似文献

As repeatedly reported, soil flooding improves the availability of P to rice. This is in contrast with an increased P sorption in paddy soils. The effects of soil flooding on the transformation of Fe oxides and the adsorption/desorption of P of two paddy soils of Zhejiang Province in Southeast‐China were studied in anaerobic incubation experiments (submerging with water in N2 atmosphere). Soil flooding significantly increased oxalate‐extractable Fe (Feox), mainly at the expense of dithionite‐soluble Fe (FeDCB), as well as oxalate‐extractable P (Pox), but decreased the ratio of Pox/Feox. Flooding largely increased both, P adsorption and the maximum P adsorption capacity. The majority of newly sorbed P in the soils was Pox, but also more newly retained P was found to be not extractable by oxalate. Flooding also changed the characteristics of P desorption in the soils. Due to a decrease of the saturation index of the P sorption capacity, P adsorbed by flooded soils was much less desorbable than that from non‐flooded soils. There are obviously significant differences in the nature of both, the Feox and Pox fractions under non‐flooded and flooded conditions. The degree of the changes in Feox, Pox, P adsorption and P desorption by flooding depended on the contents of amorphous and total Fe oxides in non‐flooded soils. Our results confirm that the adsorption and desorption behavior of P in paddy soils is largely controlled by the transformation of the Fe oxides. The reasons of the often‐reported improved P availability to rice induced by flooding, in spite of the unfavorable effect on P desorbability, are discussed.  相似文献

The first transgenic livestock were produced in 1985 by microinjection of foreign DNA into zygotic pronuclei. This was the method of choice for more than 20 years, but more efficient protocols are now available, including somatic cell nuclear transfer and lentiviral transgenesis. Typical applications include carcass composition, lactational performance and wool production, as well as enhanced disease resistance and reduced environmental impact. Transgenic farm animal production for biomedical applications has found broad acceptance. In 2006 the European Medicines Agency (EMA) approved commercialization of the first recombinant pharmaceutical protein, antithrombin, produced in the mammary gland of transgenic goats. As the genome sequencing projects for various farm animal species are completed, it has become feasible to perform precise genetic modifications employing the emerging tools of lentiviral vectors, small interfering ribonucleic acids, meganucleases, zinc finger nucleases and transposons. We anticipate that genetic modification of farm animals will be instrumental in meeting global challenges in agricultural production and will open new horizons in biomedicine.  相似文献

The biologically active form of vitamine D(3) [1alpha,25(OH)(2)D(3)] has recently been described not only to influence bone metabolism but also to exert immunomodulating activities, which may have an impact on bone formation/resorption as well. In this study, we analysed the effects of 1alpha,25(OH)(2)D(3) on the cytokine pattern of porcine bone marrow-derived cells from piglets aged 1-3 weeks. After culture for 1 week, the number of osteoclasts was determined, with tartrate-resistant acid phosphatase (TRAP)-positive, multinucleated cells being considered osteoclasts. Cultured bone marrow cell-derived mRNA was subjected to semiquantitative RT-PCR specific for a panel of porcine cytokines (IL-1alpha, IL-6, IL-8, IL-10, and TNF-alpha). In addition, an immunofluorescence analysis using anti-porcine mAbs specific for IL-1beta, IL-2, IL-4, IL-6, IL-12, TNF-alpha, and IFN-gamma was performed. In order to prove the existence of a porcine homologue of the receptor activator of NF-kappaB ligand (RANKL) bone marrow cell- as well as porcine white blood cell-derived mRNA was investigated by RT-PCR using primer pairs specific for murine RANKL. Cell culture supernatant was analysed for soluble RANKL by means of an ELISA designed for quantification of human RANKL. By means of RT-PCR, expression of IL-1alpha, IL-6, IL-8, IL-10 and TNF-alpha mRNA could be found in cells cultured with and without 1alpha,25(OH)(2)D(3). Immunofluorescence analysis revealed that IL-1, IL-6, and TNF-alpha were produced by both stromal cells and osteoclasts. Besides its known osteoclastogenic effects, 1alpha,25(OH)(2)D(3) tended to downregulate the respective cytokines, but significantly upregulated RANKL expression. The homology between the porcine RANKL-specific sequence and the corresponding human RANKL sequence was 79%. The data found support the idea that porcine bone marrow cell cultures may provide a suitable alternative to murine systems in human osteological research.  相似文献

The secretion of O2 by rice roots results in aerobic conditions in the rhizoshere compared to the bulk flooded soil. The effect of this phenomenon on the adsorption/desorption behavior and on the availability of phosphorus (P) in a flooded soil was investigated in a model experiment. An experimental set‐up was developed that imitates both O2 release and P uptake by the rice root. The results showed that O2 secretion significantly reduced P adsorption/retention and increased P desorption/release in the “rhizosphere” soil, compared to the anaerobic bulk soil. The P uptake by an anion exchange resin from both unfertilized and P‐amended soil was significantly increased. The results confirm that the O2 secretion is an important mechanism to enhance P availability and P uptake of rice under flooded conditions, where the “physico‐chemical” availability of P in the anaerobic bulk soil is strongly reduced. The decrease of P availability in the P‐amended flooded bulk soil was mainly associated with the almost complete transformation of the precedingly enriched Al‐P fraction into Fe‐bound P with extremely low desorption/release characteristics during the subsequent flooding.  相似文献

Plasma concentrations of endothelin (ET)-1 and -3 were determined simultaneously in dogs with various pathophysiological conditions, because these peptides may display different pharmacological profiles. The study pays special attention to the characterization of plasma ET immunoreactivity (ET-IR), using high-pressure liquid chromatography (HPLC) analysis with off-line detection by radioimmunoassay (RIA). In most sick dogs evaluated total plasma ET-1-IR concentration did not differ from that of healthy dogs. However, HPLC analysis of their total plasma ET-1 -IR revealed distinct ET-IR profiles. Big-ET-1, which is barely detectable in control dogs, does represent the predominant ET in sick dogs. Regardless of the pathophysiological conditions, considerable amounts of high-molecular weight ET-1-IR, most likely aggregated ET-material, was found consistently. With respect to ET-3, we constantly observed moderately increased concentrations, though no major difference of molecular pattern was evident between healthy and sick dogs. The data show a distinct regulation of ET-1 and ET-3 in dogs. Furthermore, specific molecular forms of ET-IR were found to occur in various diseases. The endothelins may therefore prove to be of diagnostic importance in the pathophysiology of vascular diseases.  相似文献

Helical computed tomography angiography was used to evaluate the renal vascular anatomy of potential feline renal donors. One hundred and fourteen computed tomography angiograms were reviewed. The vessels were characterized as single without bifurcation, single with bifurcation, double, or triple. Multiplicity was most commonly seen for the right renal vein (45/114 vs. 3/114 multiple left renal veins, 0/114 multiple right renal arteries, and 8/114 multiple left renal arteries). The right kidney was 13.3 times more likely than the left to have multiple renal veins. Additional vascular variants included double caudal vena cava and an accessory renal artery. For the left kidney, surgery and computed tomography angiography findings were in agreement in 92% of 74 cats. For the right kidney, surgery and computed tomography angiography findings were in agreement in 6/6 cats. Our findings of renal vascular anatomy variations in cats were similar to previous reports in humans. Identifying and recognizing the pattern of distribution of these vessels is important when performing renal transplantation.  相似文献

Investigations on the characterization of plant available phosphate in Thuringian calcareous soils Phosphate availability in Thuringian calcareous soils and its characterization by CAL‒, NaHCO3-, H2O- and CaCl2-soil tests was investigated in laboratory and pot experiments. Soil CaCO3 contents > 10% increase the pH value of CAL solution and thus decrease phosphate extractability. The increase of pH also causes an inadequate assessment of plant available phosphate by H2O- and CaCl2-soil tests. The CAL soluble P content of the soil corrected by the pH value of the extraction solution was most suitable for the forecast of P uptake of corn in a pot trial. From both parameters P availability indices for calcareous soils can be calculated which are comparable with those in soils containing < 5% CaCO3. The equation, which still has to be verified in field experiments, reads as CAL-Pcorrected = CAL-Pmeasured · (1 + 0.83·〈pH-valueCAL solution — 4.1〉).  相似文献
