Steers of known percentage Brahman (B) and Angus (A) breeding (100% A, n = 6; F1 B x A, n = 6; and 100% B, n = 6) were used to determine the effect of calcium chloride injection on the calpain proteinase system and meat tenderness. The steers were slaughtered in six replications (at either 9 or 14 mm of backfat, determined ultrasonically), with each breed type represented. Calpains and calpastatin activities were measured on fresh, prerigor longissimus muscle samples. Carcass data were collected after a 24-h chill, and the short loin (IMPS #180), top sirloin (IMPS #184), and top round (IMPS #168) were removed from both sides of each carcass. The cuts from the right side were then injected at 5% (wt/wt) with CaCl2 solution (2.2%). Longissimus muscle calpain and calpastatin activities were also measured at 48 h postmortem from the injected and control sides of each carcass. Warner-Bratzler shear force was measured on steaks from the three subprimals aged 1, 2, 5, 15, or 31 d. Marbling scores and USDA quality grades were higher (P<.05) in A than in F1 B x A and B carcasses. Calpastatin activity was higher (P<.05) in muscle from B than in muscle from A and F1 B x A steers, and postmortem storage (O vs 48 h) and CaCl2 injection reduced (P<.05) the activity of the calpains and calpastatin. Strip loin and top sirloin steaks from A and F1 B x A steers were more tender (P<.05) than steaks from B steers; however, top round steak tenderness did not differ (P>.05) across breed type. Calcium injection improved strip loin and top sirloin steak tenderness, but it did not affect top round steak tenderness. Collectively, these data show that CaC12 injection can be used to improve meat tenderness, with similar responses shown in cattle containing 0, 50, and 100% B inheritance. However, even with CaCl2 injection, B steaks are less tender than their A and F1 B x A counterparts.  相似文献

Microbial products are sensed through Toll-like receptors (TLRs) and trigger a program of dendritic cell (DC) maturation that enables DCs to activate T cells. Although an accepted hallmark of this response is eventual down-regulation of DC endocytic capacity, we show that TLR ligands first acutely stimulate antigen macropinocytosis, leading to enhanced presentation on class I and class II major histocompatibility complex molecules. Simultaneously, actin-rich podosomes disappear, which suggests a coordinated redeployment of actin to fuel endocytosis. These reciprocal changes are transient and require p38 and extracellular signal-regulated kinase activation. Thus, the DC actin cytoskeleton can be rapidly mobilized in response to innate immune stimuli to enhance antigen capture and presentation.  相似文献

AIM: To assess the consequences of anthelmintic resistance on commercial sheep farms on liveweight (LW) gains and susceptibility to diarrhoea of growing lambs. METHODS: Five farms with a history of resistance to benzimidazole drenches were selected. On each farm, 150 ewe lambs were randomly allocated to 1 of 3 treatment groups (n=50). Group 1 was treated 5 times at 28-day intervals with oxfendazole (to which nematode resistance had been demonstrated); Group 2 was treated 5 times at 28-day intervals with levamisole or levamisole + oxfendazole (to which nematode resistance had not been demonstrated) and; Group 3 was treated with a combination of slow-release albendazole capsules and oral moxidectin (intended to totally suppress nematode infection). Differentiated faecal egg counts (FECs), FEC reduction tests (FECRT), LW gain and degree of faecal soiling around the breech (dag scores) were determined and compared between groups. RESULTS: Pre-treatment FECs in Group 1 were similar to or slightly lower than those of Group 2. Efficacy of oxfendazole in Group 1 varied between farms and over time within farms, the reduction in FECs after treatment varying from 0% to 95%. The genera identified as resistant on each farm also varied over time. Lambs from Group 3 had lower FECs, lower dag scores and higher growth rates than lambs from Group 2, which in turn had higher LW gains than lambs from Group 1. The overall LW gains during the 5 months of the study were 7.03+/-0.27 kg for Group 1, 8.33+/-0.27 kg for Group 2, and 9.91+/-0.31 kg for Group 3 (mean+/-SEM; means differed significantly from each other, p<0.01). CONCLUSIONS: The decrease in growth rates associated with the continued use of an incompletely effective anthelmintic were surprisingly small in situations where all lambs grazed together.  相似文献

  1. Although it is well established that human activities are linked to the loss of seagrasses worldwide, the influence of anthropogenic disturbances on the habitat fragmentation of seagrass meadows is less understood. This information is essential to identify how humans are modifying seascapes and what disturbances pose the greatest risk to seagrasses, which is pertinent given the rapid urbanization occurring in coastal areas.
  2. This study examined how the habitat fragmentation of an endangered seagrass Posidonia australis varied in relation to several anthropogenic disturbances (i.e. human population, marine infrastructure, terrestrial run-off and catchment land-usage) within 10 estuaries across 620 km of coastline in New South Wales, Australia.
  3. When comparing between estuaries, the fragmentation of P. australis meadows was significantly greater in estuaries adjacent to highly populated metropolitan centres – generally in the Greater Sydney region. At sites within estuaries, the density of boat moorings was the most important predictor of habitat fragmentation, but there was also evidence of higher fragmentation with increased numbers of jetties and oyster aquaculture leases.
  4. These results suggest that the fragmentation of seagrass meadows will become more pervasive as the human population continues to grow and estuarine development increases. Strategies to mitigate anthropogenic disturbances on seagrass meadow fragmentation could include prohibiting the construction of boat moorings and other artificial structures in areas where seagrasses are present or promoting environmentally friendly designs for marine infrastructure. This knowledge will support ongoing management actions attempting to balance coastal development and the conservation of seagrasses.

A three-year field trial on an upland loessial soil (Belmont silt loam) in New Zealand investigated the effects of ripping, application of fertilizer N and grazing management on the recovery of some physical, chemical and biochemical properties of soil and pasture productivity following removal (mining) of topsoil. Removing the top 31 cm of soil by mining (all of the A horizon and part of the AB horizon) also removed most of the soil's labile organic matter fractions, and to a lesser extent its total organic matter. After three years, the microbial C and mineralizable N in the 0–10 cm depth of mined soil had reached 65 and 62 per cent of the corresponding levels in unmined soil. Ripping to a depth of 30 cm, application of fertilizer N and lenient grazing of the pasture failed to enhance the recovery of soil fertility. A soil sampling depth of 20 cm provided a reasonable basis to assess the microbial biomass and potential fertility. Pasture productivity was, on average, 30 per cent lower on mined land than on unmined land over the first three years after mining. Application of N proved uneconomic in terms of farm production, although a good pasture response to N fertilizer was obtained. The results from this and related trials are summarized as a series of recommendations for the monitoring and regulation of topsoil mining. An incentive regime is also recommended to encourage land owners and/or topsoil miners to use successful restoration techniques on topsoil-mined land.
