The secretion of O2 by rice roots results in aerobic conditions in the rhizoshere compared to the bulk flooded soil. The effect of this phenomenon on the adsorption/desorption behavior and on the availability of phosphorus (P) in a flooded soil was investigated in a model experiment. An experimental set‐up was developed that imitates both O2 release and P uptake by the rice root. The results showed that O2 secretion significantly reduced P adsorption/retention and increased P desorption/release in the “rhizosphere” soil, compared to the anaerobic bulk soil. The P uptake by an anion exchange resin from both unfertilized and P‐amended soil was significantly increased. The results confirm that the O2 secretion is an important mechanism to enhance P availability and P uptake of rice under flooded conditions, where the “physico‐chemical” availability of P in the anaerobic bulk soil is strongly reduced. The decrease of P availability in the P‐amended flooded bulk soil was mainly associated with the almost complete transformation of the precedingly enriched Al‐P fraction into Fe‐bound P with extremely low desorption/release characteristics during the subsequent flooding.  相似文献

The ubiquitous muscle protein tropomyosin has been identified as the major shrimp allergen and is suggested to be a cross-reacting allergen. Previously, only a few methods for the detection of tropomyosin in food have been published. A quantitative sandwich enzyme-linked immunosorbent assay (ELISA) for the detection of tropomyosin from crustaceans in foods has been developed and validated. A polyclonal rabbit antitropomyosin capture antibody and the biotinylated conjugate of the same antibody for detection were the basis for the ELISA, which was specific for crustaceans. The ELISA was able to quantitate tropomyosin in various food matrixes, had a detection limit of 1 microg/g, and cross-reacted to some extent with cockroach. Recoveries ranged from 63 to 120%, and the intra and interassay coefficients of variation were <6 and <14%, respectively.  相似文献

A 90‐day laboratory incubation study was carried out using six contrasting subtropical soils (calcareous, peat, saline, noncalcareous, terrace, and acid sulfate) from Bangladesh. A control treatment without nitrogen (N) application was compared with treatments where urea, ammonium sulfate (AS), and ammonium nitrate (AN) were applied at a rate of 100 mg N (kg soil)–1. To study the effect of N fertilizers on soil carbon (C) turnover, the CO2‐C flux was determined at nine sampling dates during the incubation, and the total loss of soil carbon (TC) was calculated. Nitrogen turnover was characterized by measuring net nitrogen mineralization (NNM) and net nitrification (NN). Simple and stepwise multiple regressions were calculated between CO2‐C flux, TC, NNM, and NN on the one hand and selected soil properties (organic C, total N, C : N ratio, CEC, pH, clay and sand content) on the other hand. In general, CO2‐C fluxes were clearly higher during the first 2 weeks of the incubation compared to the later phases. Soils with high pH and/or indigenous C displayed the highest CO2‐C flux. However, soils having low C levels (i.e., calcareous and terrace soils) displayed a large relative TC loss (up to 22.3%) and the added N–induced TC loss from these soils reached a maximum of 10.6%. Loss of TC differed depending on the N treatments (urea > AS > AN >> control). Significantly higher NNM was found in the acidic soils (terrace and acid sulfate). On average, NNM after urea application was higher than for AS and AN (80.3 vs. 71.9 and 70.9 N (kg soil)–1, respectively). However, specific interactions between N‐fertilizer form and soil type have to be taken into consideration. High pH soils displayed larger NN (75.9–98.1 mg N (kg soil)–1) than low pH soils. Averaged over the six soils, NN after application of urea and AS (83.3 and 82.2 mg N (kg soil)–1, respectively) was significantly higher than after application of AN (60.6 mg N (kg soil)–1). Significant relationships were found between total CO2 flux and certain soil properties (organic C, total N, CEC, clay and sand content). The most important soil property for NNM as well as NN was soil pH, showing a correlation coefficient of –0.33** and 0.45***, respectively. The results indicate that application of urea to acidic soils and AS to high‐pH soils could be an effective measure to improve the availability of added N for crop uptake.  相似文献

The effect of complex natural organic ligands on the weathering kinetics of aluminum oxide was investigated in laboratory experiments. A peat-derived humic substance and root exudates obtained from ectomycorrhizal (Picea abies — Hebeloma crustuliniforme) and non-mycorrhizal Norway Spruce trees; and γ-Al2O3 were used as a model system. The experimental weathering rates are in accordance with a surface-controlled dissolution mechanism. The effect of the humic material on dissolution rates appears to depend on the degree of protonation of the humic (macro)molecules: we observed dissolution-enhancement or -inhibition at pH 3 and 4, respectively. Ectomycorrhizal exudates proved to be effective weathering agents at pH 4, as opposed to humic material and non-mycorrhizal exudates. Our results suggest that (i) the role of humic materials in mineral weathering and podzolization is different from what is commonly thought, and (ii) mineral weathering rates in the rhizosphere may be higher than in the bulk soil.  相似文献

Under‐ as well as overfertilization with nitrogen (N) will result in economic loss for the farmer due to reduced yields and quality of the products. Also from an ecological perspective, it is important that the grower makes the correct decision on how much and when to apply N for a certain crop to minimize impacts on the environment. To aggravate the situation, N is a substance that is present in many compartments in different forms (nitrate, ammonium, organic N, etc.) in the soil‐plant environment and takes part in various processes (e.g., mineralization, immobilization, leaching, denitrification, etc.). Today, many N‐recommendation systems are mainly based on yield expectation. However, yields are not stable from year to year for a given field. Also the processes that determine the N supply from other sources than fertilizer are not predictable at the start of the growing season. Different methodological approaches are reviewed that have been introduced to improve N‐fertilizer recommendations for arable crops. Many soil‐based methods have been developed to measure soil mineral N (SMN) that is available for plants at a given sampling date. Soil sampling at the start of the growing period and analyzing for the amount of NO ‐N (and NH ‐N) is a widespread approach in Europe and North America. Based on data from field calibrations, the SMN pool is filled up with fertilizer N to a recommended amount. Depending on pre‐crop, use of organic manure, or soil characteristics, the recommendation might be modified (±10–50 kg N ha–1). Another set of soil methods has been established to estimate the amount of N that is mineralized from soil organic matter, plant residues, and/or organic manure. From the huge range of methods proposed so far, simple mild extraction procedures have gained most interest, but introduction into practical recommendation schemes has been rather limited. Plant‐analytical procedures cover the whole range from quantitative laboratory analysis to semiquantitative “quick” tests carried out in the field. The main idea is that the plant itself is the best indicator for the N supply from any source within the growth period. In‐field methods like the nitrate plant sap/petiole test and chlorophyll measurements with hand‐held devices or via remote sensing are regarded as most promising, because with these methods an adequate adjustment of the N‐fertilizer application strategy within the season is feasible. Prerequisite is a fertilization strategy that is based on several N applications and not on a one‐go approach.  相似文献

Micrometeorological measurements of nighttime ecosystem respiration can be systematically biased when stable atmospheric conditions lead to drainage flows associated with decoupling of air flow above and within plant canopies. The associated horizontal and vertical advective fluxes cannot be measured using instrumentation on the single towers typically used at micrometeorological sites. A common approach to minimize bias is to use a threshold in friction velocity, u*, to exclude periods when advection is assumed to be important, but this is problematic in situations when in-canopy flows are decoupled from the flow above. Using data from 25 flux stations in a wide variety of forest ecosystems globally, we examine the generality of a novel approach to estimating nocturnal respiration developed by van Gorsel et al. (van Gorsel, E., Leuning, R., Cleugh, H.A., Keith, H., Suni, T., 2007. Nocturnal carbon efflux: reconciliation of eddy covariance and chamber measurements using an alternative to the u*-threshold filtering technique. Tellus 59B, 397–403, Tellus, 59B, 307-403). The approach is based on the assumption that advection is small relative to the vertical turbulent flux (FC) and change in storage (FS) of CO2 in the few hours after sundown. The sum of FC and FS reach a maximum during this period which is used to derive a temperature response function for ecosystem respiration. Measured hourly soil temperatures are then used with this function to estimate respiration RRmax. The new approach yielded excellent agreement with (1) independent measurements using respiration chambers, (2) with estimates using ecosystem light-response curves of Fc + Fs extrapolated to zero light, RLRC, and (3) with a detailed process-based forest ecosystem model, Rcast. At most sites respiration rates estimated using the u*-filter, Rust, were smaller than RRmax and RLRC. Agreement of our approach with independent measurements indicates that RRmax provides an excellent estimate of nighttime ecosystem respiration.  相似文献

Effect of arbuscular mycorrhizal fungi (AMF) on heavy metal tolerance of alfalfa (Medicago sativa L.) and oat (Avena sativa L.) on a sewage-sludge treated soil In pot experiments with a sewage sludge treated soil, the influence of two arbuscular mycorrhizal fungi (AMF) isolates of Glomus sp. (T6 and D13) on plant growth and on the uptake of heavy metals by alfalfa (Medicago sativa L.) and oat (Avena sativa L.) was investigated. Alfalfa showed an increase of biomass with mycorrhizal infection only to a small extent. In oat AMF inoculation increased the growth of both root and shoot by up to 70% and 55% respectively. Mycorrhization raised the P-content and -uptake in alfalfa, but not in oat, in both roots and shoots. Mycorrhizal alfalfa showed lower Zn-, Cd- and Ni-contents and uptake in roots and shoots. The root length was significantly decreased in mycorrhizal alfalfa plants (up to 38%). The translocation of heavy metals into the shoot of mycorrhizal alfalfa was slightly increased. Mycorrhizal infection of oat led to higher concentrations of Zn, Cd and Ni in the root but to less Zn in the shoot. The translocation of heavy metals to the oat shoot was clearely decreased by mycorrhizal colonisation. This may be based on the ability of fungal tissues to complex heavy metals at the cell walls, thus excluding metals from the shoot. This conclusion is supported by the enhanced root length (up to 78%) of mycorrhizal oat plants in this experiment. The mycorrhizal infection seemed to protect plants against heavy metal pollution in soils. It was obvious that different host plants reacted in different ways.  相似文献

Forest fungi not only have important functions within the forest ecosystem, but picking their fruit bodies is also a popular past time, as well as a source of income in many developing and developed countries. The expansion of commercial harvesting in many parts of the world has led to widespread concern about overharvesting and possible damage to fungal resources. In 1975, we started a field research project to investigate the effects of mushroom picking on fruit body occurrence. The three treatments applied were the harvesting techniques picking and cutting, and the concomitant trampling of the forest floor. The results reveal that, contrary to expectations, long-term and systematic harvesting reduces neither the future yields of fruit bodies nor the species richness of wild forest fungi, irrespective of whether the harvesting technique was picking or cutting. Forest floor trampling does, however, reduce fruit body numbers, but our data show no evidence that trampling damaged the soil mycelia in the studied time period.  相似文献
