To acquire epidemiological data on the bovine viral diarrhea virus (BVDV) and identify cattle persistently infected (PI) with this virus, 4,327 samples from Holstein dairy cows were screened over a four-year period in Beijing, China. Eighteen BVD viruses were isolated, 12 from PI cattle. Based on genetic analysis of their 5''-untranslated region (5''-UTR), the 18 isolates were assigned to subgenotype BVDV-1m, 1a, 1d, 1q, and 1b. To investigate the innate immune responses in the peripheral-blood mononuclear cells of PI cattle, the expression of Toll-like receptors (TLRs), RIG-I-like receptors, interferon-α (IFN-α), IFN-β, myxovirus (influenza virus) resistance 1 (MX1), and interferon stimulatory gene 15 (ISG15) was assessed by qPCR. When compared with healthy cattle, the expression of TLR-7, IFN-α, and IFN-β mRNA was downregulated, but the expression of MX1 and ISG-15 mRNA was upregulated in PI cattle. Immunoblotting analysis revealed that the expression of interferon regulatory factor 3 (IRF-3) and IRF-7 was lower in PI cattle than in healthy cattle. Thus, BVDV-1m and 1a are the predominant subgenotypes in the Beijing region, and the strains are highly divergent. Our findings also suggest that the TLR-7/IRF-7 signaling pathway plays a role in evasion of host restriction by BVDV. 相似文献
Aquaculture research has focused on polysaccharides as they are among the most promising new-generation immunostimulant used to control aquatic diseases. The aim of this study was to investigate the immunomodulatory effects of fermented wheat bran polysaccharides (FWBPs) on juvenile common carp (Cyprinus carpio). Carps were fed different FWBP amounts (0%, 0.1%, 0.2%, and 0.4%) for 8 weeks and then their skin mucus and serum immune parameters, hepatopancreas antioxidant indicators, and immune-related gene expression in the intestines, kidneys, and spleen were measured. The skin mucus IgM levels significantly increased in 0.1% FWBP group, but decreased in 0.4% FWBP group. The skin mucus protease and the serum alkaline phosphatase activities increased significantly in the 0.2%, 0.1%, and 0.4% FWBP groups, respectively. The serum total Ig levels increased noticeably in the 0.1% and 0.2% FWBP groups. The highest and lowest serum lysozyme activities were observed in the 0.1% and 0.4% FWBP groups, respectively. The hepatopancreatic total superoxide dismutase activity was higher in the 0.1% FWBP group than in the control. The malondialdehyde levels decreased significantly in the 0.2% and 0.4% groups. The intestinal mRNA levels of the LZM-C and IL-10 genes were significantly higher in the 0.2% than in the 0.4% FWBP group; TNF-α was significantly upregulated in the 0.1% group. The gene expression in the kidneys did not differ significantly among the treatments, except for a significant increase in the IL-10 expression in the 0.1% treatment. Significantly elevated expression of LZM-C in 0.2% group and IL-10 in 0.1% group was observed in the spleens. TNF-α and IL-1β were significantly downregulated in the 0.4% group. These results suggest that FWBPs could be used as immunostimulant feed additives in common carp cultures.
Mung bean trypsin inhibitor (MBTI) of the Bowman-Birk family was purified to homogeneity with a molecular mass of approximately 9 kDa on tricine-sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and 8887.25 Da as determined by matrix-assisted laser desorption/ionization-quadrupole ion trap-time-of-flight mass spectrometry (MALDI-QIT-TOF MS). Using blue scad myofibrillar proteins as targets, it was found that, in the absence of MBTI, proteolysis of myofibrillar proteins, especially myosin heavy chain (MHC), could be identified after incubation at 55 °C for 2 h, while in the presence of MBTI, with a final concentration of 25 ng/mL, proteolysis of these proteins was greatly suppressed even after incubation for 3 h. Although cysteine proteinase inhibitor E-64 was also effective in preventing protein degradation, inhibitors for metallo- and asparatic proteinases did not reveal obvious inhibitory effects. Our present results strongly suggested that the naturally occurring legume bean seed protein MBTI can be used as an effective additive in preventing marine fish blue scad surimi gel softening, which is quite possibly caused by myofibril-bound serine proteinase (MBSP). 相似文献
Agarose is a natural seaweed polysaccharide and widely used in the medicine, food, and biological fields because of its high gel strength, non-toxicity, and electrical neutrality. The sulfate group is one of the main charged groups that affect the performance of agarose. In the present study, a simple, eco-friendly, and efficient method was explored for agarose preparation. After desulfation with hydrogen peroxide (H2O2), the sulfate content of agar reached 0.21%. Together with gel strength, electroendosmosis, gelling and melting temperature, the indicators of desulfated agar met the standards of commercially available agarose. Notably, the desulfated agar can be used as an agarose gel electrophoresis medium to separate DNA molecules, and the separation effect is as good as that of commercially available agarose. Further, the H2O2 desulfation process was analyzed. The addition of a hydroxyl radical (HO•) scavenger remarkably decreased the H2O2 desulfation rate, indicating that HO• has a certain role in agar desulfation. Sulfate content detection indicated that sulfur was removed from agar molecules in the form of sulfate ions (SO42−) and metal sulfate. The band absence at 850 cm−1 indicated that the sulfate groups at C-4 of D-galactose in sulfated galactan were eliminated. 相似文献
