A study was made to gain greater understanding of the development of translucent end tubers which caused problems for french fry processors duringthe processing season of 1971–72. Analysis of seven commercial and three experimental samples revealed a lower dry matter and higher reducing sugar content in the basal portion of the tubers. Normally Russet Burbank potatoes contain higher dry matter in the basal end. A study of dry matter changes during growth and development in 1971 and of moisture stressed potatoes in 1972 showed a reversal in dry matter content and an uneven distribution of reducing sugars, whereas the normally irrigated potatoes maintained a higher percentage of dry matter in the basal portion throughout. A decrease in pH of the tissue accompanied the loss of starch from the basal portion of moisture stressed potatoes. As postulated by other research workers, the loss of dry matter is attributed to initiation of active growth of the apical end, thereby utilizing carbohydrates from the basal end following a period of stress. Immaturity of tubers and storage at low temperatures immediately after harvest caused water stressed potatoes to develop increased numbers of translucent ends in storage.  相似文献

In organ cultures of intact rat pineal glands, N(6)O(2')-dibutyryl adenosine 3', 5'-monophosphate stimulates the conversion of tritiated trytophan to tritiated melatonin, as does L-norepinephrine. Potential sites of stimulation of melatonin production by dibutyryl cyclic adenosine monophosphate are discussed, based on observations that the dibutyryl analog also stimulates the conversion of serotonin labeled with carbon-14 to carbon-14-labeled melatonin without altering hydroxyin-dole-O-methyl transferase activity or intracellular accumulation of serotonin labeled with carbon-14.  相似文献

The hypothesis that endogenous cholecystokinin (CCK) released from the small intestine during feeding causes satiety was tested in rat pups, 9 to 12 days old. Intragastric administration of soybean trypsin inhibitor, a procedure that releases CCK from the small intestine, decreased the subsequent intake of a test meal. This effect was reversed by prior treatment with MK-329, a selective antagonist of CCK at alimentary-type CCK (CCK-A) receptors. Thus, endogenous, small intestinal CCK can cause satiety in the neonatal rat and this effect involves CCK-A receptors.  相似文献

Glufosinate is a widely used herbicide, which is difficult to detect by conventional analytical techniques. For many other herbicides, suitable antibodies have been raised for immunoassay development. Unfortunately, glufosinate is a very small molecule and difficult to immunize with. Thus, a derivatization-assisted immunoassay (DAIA) using the target analyte N-acetyl-glufosinate (NAG) was constructed. The activated hapten was synthesized by a new approach, using a homobifunctional cross-linker suberic acid bis(N-hydroxysuccinimide ester). The preparation of a suitable conjugate, the immunization, and the characterization of polyclonal antibodies are shown. The determination of the conjugation density (hapten density) of the immunogens was performed by four different methods (high-performance liquid chromatography with a refractive index detector, total reflection X-ray fluorescence, inductively coupled plasma mass spectrometry, and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry), which gave similar results. The limit of detection was 17 mug/L NAG in water for the direct competitive enzyme immunoassay. NAG is also a main metabolite of glufosinate in resistant transgenic plants. The antibodies might be useful for the selective detection of NAG in the presence of the parent compound glufosinate (cross-reactivity 0.13%) and other metabolites.  相似文献

2,4-Diacetylphloroglucinol (DAPG) is an antibiotic with broad-spectrum antibacterial and antifungal activities. It is a major determinant in the biological control of several plant diseases. DAPG is produced by Pseudomonas fluorescens both in vitro and in the rhizosphere of wheat. It is involved in the natural suppression of take-all disease known as take-all decline, which develops in soils following extended monoculture of wheat or barley. A one-step synthesis of DAPG from the commercially available 2-acetylphloroglucinol is described. This reaction involves the direct alkylation of 2-acetylphloroglucinol using acetic anhydride as the acetylation reagent, with boron trifluoride-etherate as the catalyst. This synthesis is simple and produces higher yields of DAPG (90%) as compared with previously described procedures. As ecological concerns are gaining equal status with agricultural concerns, the demand for natural biocontrol measures is increasing. There is tremendous pressure from society on agriculture to reduce the use of pesticides. A discussion is given on the agricultural and ecological importance of this natural antibiotic and its application as an alternative to reduce the use of synthetic pesticides.  相似文献

We studied discharges of organic carbon from eight contiguous small watersheds on the Atlantic Coastal Plain in Maryland for up to 24 yr. Six of these watersheds were second or third orderwith mixed-land-use, while two were first order (one completely forested and one highly dominated by cropland). These watershedshave perched aquifers, so all groundwater discharges as well assurface runoff were measured at V-notch weirs and flumes, whichincluded volume-integrating flow-proportional samplers. Interannual variations in annual and seasonal precipitation during this study spanned approximately the range of 160 yr weather records in the region. Annual total organic carbon (TOC)area yields from the overall Rhode River watershed varied 8-fold, correlations with precipitation were highly significant,and a power function regression explained 54% of the variance in annual TOC fluxes. TOC fluxes were higher from upland forestthan mixed land use, and highest from the cropland-dominated watershed. The fluxes from first order watersheds were more variable with precipitation. In the spring, TOC fluxes were highest and most correlated with precipitation, compared to other seasons. Precipitation volume also explained much of thevariance in annual and spring TOC concentrations from upland forest and cropland, with concentrations three to five times higher in very wet years than in very dry years. Variation in winter and summer air temperature was correlated with TOC concentrations from forested watersheds, and linear regressionsexplained 19 to 42% of the variance in TOC. A regression modelwas used to construct graphical and tabular summaries. Particulate organic carbon and dissolved organic carbon (DOC)concentrations and the ratio of DOC to TOC were highly correlated with water discharge for a second order, mixed land use watershed, and power function regressions explained 21 to 43% of the variance. For the first order, single-land-use watersheds the ratio of DOC to TOC was also highly correlatedwith discharge.  相似文献

Cooking ability or time-to-breakdown (TTB) of different portions of potato tubers was determined by taking cores 1.27 cm (1/2inch) in diameter and 0.95 cm (3/8 inch) long from 12 locations in the tubers and cooking until penetration of a weighted rod pressing on the cores. Cores from the stem portion, on the average, required over twice as long for penetration of the rod during cooking as compared to cores from the bud section. The cores from the middle longitudinal portion of the tuber took longer to cook than cores from either side. Low specific gravity (1.070) tubers took significantly longer to cook than cores from high specific gravity (1.090) potatoes. However, when specific gravity of individual cores was determined with a pycnometer and TTB determined for the same samples, no relationships could be established. Cores from the stem portion took longer for TTB regardless of specific gravity or starch content. The data suggested much greater influence of cell wall structure as would be obtained from different portions of the tuber on TTB than starch content of the cells. No differences in TTB were obtained from tubers of high or low fertility or between Norgold Russet and Russet Burbank varieties, except Norgold was more uniform in cooking time between the stem and bud portion than Russet Burbank.  相似文献

Often overlooked in the presence of neoplasia, PNDs constitute significant clinical entities in dogs with hematopoietic tumors. They may cause morbidity and mortality in such patients, with effects more severe than those caused by the associated tumor. Accurate clinical evaluation of these disorders is important in differential diagnosis and treatment, for failure to realize that cancer can produce many clinical signs similar to those of other diseases may lead to incorrect diagnosis and delayed therapy. Early recognition of the problem underlying the PND is essential to selecting the proper therapeutic approach and maximizing the patient's chances for remission and survival. The presence of these disorders may complicate or rule out the preferred therapy in some cases of hematopoietic neoplasia, because the addition of cytotoxic drugs may worsen the existing PND, predisposing the dog to a variety of complications. Appropriate management of the PND may be of more immediate importance than treatment of the tumor. The study and recognition of PNDs in dogs with hematopoietic tumors may be valuable for a number of reasons: to facilitate early diagnosis of the tumor, for the observed abnormalities may represent tumor cell markers; to allow assessment of premalignant states; to aid in the search for metastases; to help quantify and monitor response to therapy; to aid in the evaluation of tumor recurrence or progression; to aid in identifying specific pathophysiologic processes by which cancer produces systemic effects; and to provide insight into the study of malignant transformation. Recognition of PNDs is relevant to the diagnosis and treatment of many problems in veterinary cancer medicine. With increasing emphasis on diagnosis and treatment of canine hematopoietic tumors, PNDs will be recognized with greater frequency and will assume greater importance in the therapeutic management of those patients. Research in veterinary and human cancer medicine needs to be directed toward identifying more definitively those substances and pathways that are responsible for PNDs, because therapy directed toward arresting the specific pathophysiologic processes causing the PND may offer the best approach for successful management of cancer.  相似文献
