Both epidemiological and experimental data indicate that a diet rich in fiber may reduce cancer risk. One possible mechanism is by adsorbing carcinogens and transporting them out of the body without metabolic activation. We investigated the role of fiber lignification and feruloylation on the adsorption of four of the most relevant heterocyclic aromatic amines in food: 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx), 2-amino-3-methylimidazo[4,5-f]quinoline (IQ), 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), and 2-amino-9H-pyrido[2,3-b]indole (AalphaC). Adsorption experiments, under conditions mimicking the small intestine, were carried out using nonlignified and artificially lignified primary maize walls with defined lignin and ferulate/diferulate concentrations and defined lignin compositions. Lignin concentration and composition both influenced the adsorption of heterocyclic aromatic amines, especially the more hydrophobic types. Heterocyclic aromatic amine adsorption increased with lignin concentration. 2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyridine and 2-amino-9H-pyrido[2,3-b]indole were better adsorbed by guaiacyl-rich lignins, 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline by syringyl-rich lignins, whereas the adsorption of 2-amino-3-methylimidazo[4,5-f]quinoline was not clearly influenced by lignin composition. Nonlignified cell walls adsorbed lesser amounts of heterocyclic aromatic amines. Variations in cell wall feruloylation had no effect on heterocyclic aromatic amine adsorption. 相似文献

CASE DESCRIPTION: 4 North American porcupines were evaluated because of diarrhea or neutropenia (or both) that developed after treatment with fenbendazole for intestinal parasites. CLINICAL FINDINGS: Complete blood cell count abnormalities included severe neutropenia in all affected porcupines and mild anemia in some of them. In 2 porcupines, postmortem findings included bone marrow hypoplasia and intestinal crypt cell necrosis. TREATMENT AND OUTCOME: Affected porcupines received supportive care including fluid supplementation and broad-spectrum antimicrobials. The 2 surviving animals recovered after 9 to 33 days of treatment. CONCLUSIONS AND CLINICAL RELEVANCE: Fenbendazole is an anthelminthic that may be used in an extralabel manner for the treatment of intestinal parasitism in wildlife species. The drug inhibits mitosis and can affect rapidly dividing cell lines, such as those in the bone marrow and intestinal crypt mucosa. Fenbendazole may not be an appropriate anthelminthic choice in North American porcupines. 相似文献

OBJECTIVES: To test a non-glycosylated recombinant human bone morphogenetic protein-2 (ngly-rhBMP-2)/fibrin composite, which has been shown experimentally to enhance healing of bone defects in rodents, in a clinical case series of dogs and cats undergoing treatment for fracture non-unions and arthrodesis. METHODS: A ngly-rhBMP-2/fibrin composite was applied in 41 sites in 38 dogs and cats for which a cancellous bone autograft was indicated, replacing the graft. RESULTS: Bridging of the bone defect with functional bone healing was achieved in 90 per cent of the arthrodesis and fracture nonunions treated in this manner. CLINICAL SIGNIFICANCE: This prospective clinical study demonstrates the beneficial effects of ngly-rhBMP-2 in a specially designed fibrin matrix on the treatment of bone defects, and validates the use of this composite as an alternative to bone autografts in dogs and cats. 相似文献

Goal, Scope and Background Numerous xenobiotics released into surface waters are transferred to suspended particulate matter and finally attached to
sediments. Aquatic organisms may be exposed to them by direct particle feeding, by physical contact with contaminated surfaces
as an exposure route, and by the uptake of dissolved contaminants after equilibration via the free water phase. In order to
assess potential sediment toxicity, each of these exposure routes has to be addressed. This paper presents a newly developed
particle contact assay that uses the fermentation performance of a specific Saccharomyces cerevisiae strain for the assessment
of toxic effects in sediments. The test procedure is based on the characteristic feature of growing yeast cells to attach
to sediment particles, which are also relevant for the accumulation of contaminants. The physical contact with lipophilic
contaminants mirrors an exposition pathway for the direct uptake into the cells. In order to quantitatively characterize the
toxic effects of particle attached pollutants on the fermentation performance, unpolluted native reference sediment was spiked
with representatives for widely distributed anthropogenic contaminants.
Methods Saccharomyces cerevisiae was established as sensitive eukaryotic microorganism for the ecotoxicological assessment of particle
attached anthropogenic contaminants in freshwater sediments. For this purpose, yeast cells were cultivated in sediment samples
and the resulting fermentation performance was continuously measured. Sediments artifically spiked with HCB, PCB, g-HCH, DDT,
and benzo(a)pyrene and solutions of each contaminant were comparatively investigated by means of their adverse effects on
yeast fermentation performance. Additionally, four native river sediments characterized by increasing levels of pollution
were assessed by the yeast particle contact assay, and simultaneously by standard aquatic tests with algae, daphniae, and
luminescent bacteria using pore water and elutriates. Results of the bioassays were related to specific sediment contamination
with respect to metals and organic priority pollutants.
Results and Discussion In sediments spiked with PCB and benzo(a)pyrene fermentation, performance was affected extensively below concentrations inhibiting
fermentation in contaminant solutions. This suggests a high efficiency of the exposure route by physical contact. The fermentation
performance was only slightly affected by single lipophilic pollutants, whereas mixtures of individually spiked sediments
caused critically reduced fermentation performance suggesting additive synergistic effects. Native river sediments modestly
to critically polluted by hazardous organic compounds lead to a slightly to dangerously reduced fermentation performance in
the yeast contact assay. These inhibitory effects were much less pronounced in the standard bioassays conducted with algae,
daphniae and luminescent bacteria, applying pore waters and elutriates as sample matrices. Using pore water, inhibition was
measured only in the most polluted sediment, elutriates lead to a slight inhibition of the algal growth in the undiluted sample
only. These results indicate an improved sensitivity of the yeast particle contact assay compared to the standard assays,
due to uptake and physical cell contact as additional routes of exposure.
Conclusion The yeast particle contact assay is a valuable tool for the assessment of ecotoxicological potential in freshwater sediments.
Since the assay addresses physical contact as an exposure route, it indicates bioavailability of lipophilic compounds in sediments.
Outlook The sensitive indication of bioavailable contaminants associated to sediment particles by the newly developed yeast particle
contact assay recommends it as a complementary microbial bioassay in a test battery for assessing major pathways of contaminants

OBJECTIVE: To determine prevalence of Holstein bulls with chromosomal anomalies, particularly the 1/21 centric fusion (CF), at a commercial artificial insemination (AI) company in the United States. DESIGN: Cross-sectional cytogenetic prevalence study. ANIMALS: All 606 Holstein bulls at a commercial AI company were cytogenetically screened to detect CF, chimerism, and other chromosomal abnormalities. PROCEDURE: Lymphocytes from heparinized blood samples were cultured by standard cytogenetic techniques, and chromosome spreads were prepared for microscopic examination. Chromosomal abnormalities were detected by examining 10 chromosome spreads per bull. Pedigree analysis was performed. RESULTS: None of the bulls had any type of CF. However, 6 bulls were identified as chimeras (i.e., contained lymphocytes with male [XY] and female [XX] chromosomes). One bull was sire or maternal grandsire to 85 of the bulls tested, and 739 of 1,212 (61%) sire and maternal-grandsire possibilities were accounted for by just 18 bulls. CONCLUSIONS AND CLINICAL RELEVANCE: Analysis of these results supports previous indications that CF is extremely rare in Holstein bloodlines available commercially via AI in the United States. However, chimeric bulls are more common, and they reportedly have decreased reproductive performance. Therefore, identification of chimeric sires in the AI facility reported here and the possibility of de novo onset of CF at any time indicates that early cytogenetic screening should be encouraged for prospective bulls intended for use in AI programs. 相似文献

An assessment was made of the risks of bovine spongiform encephalopathy (BSE) occurring in Argentina. Most of the factors associated with the origin and development of the BSE epidemic in the UK are essentially absent. For example, Argentina's large sheep and cattle industries are based on low-cost production systems using grass. Concentrated feeds are not used for sheep, rarely for beef cattle and to a comparatively modest extent for dairy cows. Particularly important are the facts that scrapie (and BSE) has never been reported in Argentina—very small amounts of waste tissues from sheep are rendered to produce meat and bone meal (MBM)—and MBM is not used in concentrated feeds for cattle. We conclude that Argentina has an exceptionally low risk of BSE due to scrapie. There is a very small risk of BSE having been introduced via live animals imported from countries with BSE, but this could only give rise to isolated cases because MBM is not fed to cattle.
A surveillance programme has been carried out based largely on a histological examination of brains from three categories of old dairy cows: animals reported on the suspicion of having neurological disease; animals in poor condition at slaughter; healthy animals randomly selected in the abattoir. No evidence of transmissible spongiform encephalopathy was seen in several sections from each of a total of 1019 brains. We conclude that, for most practical purposes, Argentina may be considered to be free from BSE. 相似文献

133 samples of food were investigated for comparison of a recent commercially available 1-2 test (Biocontrol) with a cultural standard method (non-selective pre-enrichment in buffered peptone water, selective enrichment in Rappaport-Vassiliadis-medium and selenite brilliant-green mannitol enrichment broth, inoculation on two selective agars) for presence of Salmonella. Magnetic reconnection of solar coronal loops is the main process that causes solar flares and possibly coronal heating. In the standard model, magnetic field lines break and reconnect instantaneously at places where the field mapping is discontinuous. However, another mode may operate where the magnetic field mapping is continuous but shows steep gradients: The field lines may slip across each other. Soft x-ray observations of fast bidirectional motions of coronal loops, observed by the Hinode spacecraft, support the existence of this slipping magnetic reconnection regime in the Sun's corona. This basic process should be considered when interpreting reconnection, both on the Sun and in laboratory-based plasma experiments. 相似文献
