A laboratory assay was designed to determine the insecticidal efficacy of Beauveria bassiana (Balsamo) Vuillemin (Hyphomycetes: Moniliales) and diatomaceous earth (Diafil 610) against Rhyzopertha dominica (F.) (Coleoptera: Bostrychidae). The fungus B. bassiana was applied at 2.23 × 107, 2.23 × 108 and 2.23 × 109 conidia kg−1 of wheat individually as well as mixed with 200 and 400 ppm of Diafil 610. The conditions for the trials were 30 ± 2oC with 55% r.h. and the counts for mortality were made after 8, 16 and 24 d. All the dead adults were removed after each count and the vials were kept for the next 60 d to assess the emergence of the F1 generation. The findings from these studies proved that the extended exposure interval and the highest combined dose rate of the entomopathogenic fungus and the diatomaceous earth gave the maximum mortality of the beetles. The emergence of the progeny was also highly suppressed where the maximum dose rate of the synergized treatments was applied. The rate of mycosis and sporulation in the cadavers of R. dominica was maximum where the low dose rates of B. bassiana were applied.  相似文献

In 2010, an H5N1 highly pathogenic avian influenza virus (HPAIV) was isolated from feces of apparently healthy ducks migrating southward in Hokkaido, the northernmost prefecture of Japan. The H5N1 HPAIVs were subsequently detected in domestic and wild birds at multiple sites corresponding to the flyway of the waterfowl having stopovers in the Japanese archipelago. The Hokkaido isolate was genetically nearly identical to H5N1 HPAIVs isolated from swans in the spring of 2009 and 2010 in Mongolia, but less pathogenic in experimentally infected ducks than the 2009 Mongolian isolate. These findings suggest that H5N1 HPAIVs with relatively mild pathogenicity might be selected and harbored in the waterfowl population during the 2009-2010 migration seasons. Our data provide "early warning" signals for preparedness against the unprecedented situation in which the waterfowl reservoirs serve as perpetual sources and disseminators of HPAIVs.  相似文献

1. Domestic fowl and free-living birds were examined for the presence or absence of antibodies against hydropericardium syndrome (HPS) using an indirect haemagglutination assay.

2. Two-hundred and eighty serum samples of commercial (45 broilers, 20 adult layers and 15 Fayoumi fowl) and wild birds, including 65 peafowl, 45 pigeons, 10 crows, 30 house sparrows, 10 doves, 15 ducks, 10 parrots and 15 guinea fowl, were collected and examined.

3. The percentage of HPS-positive serum samples was 80% in house crows, 78% in pigeons, 7% in house sparrows and 6% in peafowl.

4. The sera obtained from parrots, doves, ducks and guinea fowl were all negative.

5. This study suggests that crows and pigeons could be carriers of the HPS agent.  相似文献

‘Phosphate solubilizing bacteria' (PSBs) are able to release unavailable P from native and applied P sources into plant‐available soil pool through their solubilizing and acidifying effects. The effects of three indigenous and one exotic PSBs on P solubilization from different P sources, plant biomass production, and P‐uptake efficiency of maize (Zea mays L.) were examined in an incubation and greenhouse study. For incubation study, surface (0–15 cm) soil was collected from an arable field (Inceptisols) and amended with rock phosphate (RP), single superphosphate (SSP), poultry manure (PM), and RP+PM with and without PSBs. The amended soil was incubated in the control environment at 25 ± 2°C for a total of a 100‐d period to establish relative potential rate of P solubilization of added P sources. A complementary greenhouse experiment was conducted in pots by growing maize as a test crop. Growth characteristics, P‐uptake, and P‐utilization efficiency (PUE) were determined. Phosphate solubilizing bacteria generated a solubilization effect on different P sources by releasing more P into plant‐available soil pool, i.e., 14.0–18.3 µg g?1 in RP, 5.0–9.9 µg g?1 in SSP, 1.4–4.4 µg g?1 in PM, and 4.5–7.8 µg g?1 in RP+PM compared to their sole application without PSBs. The available P from inorganic SSP declined continuously from the mineral pool (after day 30) and at the end 40% of applied P was unaccounted for. However, P losses were reduced to 28 and 27% when PSBs (PSB1 and PSB3) were applied with superphosphate treatments. In the absence of PSBs, the recoveries of applied P (in soil) from RP, SSP, PM and RP+PM were 4, 25, 9, and 12%, respectively, those had been increased to 14, 30, 12 and 15% in the presence of PSBs. Similarly, the plant biomass in RP+PSBs treatments compared to the RP without PSBs increased between 12–30% in first sampling (30 DAG) and 13–30% in the second sampling (60 DAG). The P utilization efficiency (PUE) in plants supplemented with PSBs was 20–73% higher compared to those without PSBs. The detection of oxalic and gluconic acids in culture medium treated with PSBs (7.8–25.0 and 25–90 mg L?1, respectively) confirmed the production of organic acids by the indigenous bacterial isolates. This study indicate that low P recovery both in plant and soil can likely be improved by using indigenous PSBs and organic amendment poultry manure, which allowed a more efficient capture of P released due to P solubilization.  相似文献

Exophiala sp. LHL08, a gibberellin-producing strain, was investigated to assess its effects on cucumber plant growth and heat (40°C) stress tolerance. The results reveal that Exophiala sp. associated plants had significantly higher plant growth attributes (shoot length, plant biomass, chlorophyll contents, and leaf area) than control under heat stress. Endophytic association helped the plants to obtain adequate water to reduce the leaf electrolytic leakage under stress. High-temperature-induced oxidative stress was less pronounced in Exophiala sp. associated plants as shown by enhanced levels of total polyphenol and reduced activities of glutathione, superoxide anion, and lipid peroxidation. To tolerate heat stress and rescue plant growth, the endophyte association significantly increased catalase and peroxidase activities of the host plants as compared to control plants. Contents of palmitic, stearic, oleic, and α-linolenic were significantly decreased in the Exophiala sp.-inoculated plants than control plants under heat stress. Contents of flavonoids like genistein and daidzein were produced in higher quantities, while glycitein content was almost same in endophyte-associated plants under heat stress than control plants. Contrarily, stress-responsive endogenous abscisic acid and jasmonic acid were significantly activated in non-inoculated control treatments as compared to endophyte-inoculated plants under heat stress. The findings of the study reveal that association of Exophiala sp. with cucumber host plants can modulate heat stress by influencing physiological and biochemical contents of plants under heat stress.  相似文献

ABSTRACT

Climate change is imposing high temperature resulting in prolonged drought episodes and shrinking of fresh water resources across the globe. In this scenario, even drought tolerant crops like quinoa are also losing significant yield. However, this study was planned to investigate the impact of drought on quinoa at critical growth stages and bacterial inoculation to improve drought tolerance. Drought was imposed by maintaining 25% pot water holding capacity (PWC) at multiple leaf, flowering, and seed filling stage (DSFS), while 80% PWC was considered as control. Three strains of plant growth promoting rhizobacteria (PGPR) named as: Bacillus licheniformis, Pseudomonas fluorescens, and Azospirillum brasilense were inoculated with quinoa seeds before sowing with respect to drought treatments. PGPR inoculation mitigated the drastic effects of drought by improving crop growth, net assimilation rate, water use efficiency, leaf chlorophyll, and phenolic contents, all of these ultimately contributed to improvement in grain yield and its contributing attributes. Moreover, PGPR markedly improves the grain quality attributes including protein, phosphorus, and potassium contents. Principal component analysis linked the different scales of study and demonstrated the potential of physio-biochemical traits to explain the quinoa yield variations under drought condition with response to PGPR inoculation. Among different PGPR, A. brasilense was found most effective both under normal and drought conditions. Overall, DSFS has more detrimental effects among critical growth stages of quinoa and A. brasilense can be used as a shotgun tactic to ameliorate drought stress in quinoa.  相似文献

ABSTRACT

Sulfur (S) availability is a characteristic of conservation tillage. We studied the effects of S on sunflower yield and fatty acid profile under conventional and conservation tillage in silty clay soil of D.I. Khan, Pakistan. Conventional tillage consisted of disk plowing followed by tiller and rotavator, while conservation tillage comprises tiller and disc harrowing. Treatments comprise six sulfur levels (S1 = 0, S2 = 20 kg S ha?1 at sowing, S3 = 10 kg S ha?1 at sowing +10 kg S ha?1 25 days after sowing (DAS), S4 = 40 kg S ha?1 at sowing, S5 = 20 kg S ha?1 at sowing + 20 kg S ha?1 25 DAS, and S6 = 20 kg S ha?1 at sowing + 10 kg S ha?1 25 DAS + 10 kg S ha?1 50 DAS) replicated thrice. After 2 years of experimentation, soil samples were collected (from 0 to 0.30 m depth) for organic matter (OM), total soil N (TSN), and available sulfur analysis. Conservation tillage significantly increased OM, TSN, and S contents in 0–0.30 m soil compared to conventional tillage. Likewise, sunflower plants showed higher achene yield, better fatty acid profile, and net economic returns in conservation tillage than in conventional tillage. Application of 20 kg S ha?1 at sowing and 10 kg S ha?1 at each 25 and 50 DAS recorded significantly higher achene yield, higher oil, and linoleic acid contents; however, oleic and oleic/linoleic ratio did not increase with higher S rates. Conservation tillage was effective in increasing OM, TSN, and available S besides increasing yield and economics in silty clay soil.  相似文献

In this research we explore the potential of precision surface irrigation to improve irrigation performance under the warabandi system prevalent in the Indus Basin Irrigation System. Data on field dimensions, field slopes along with characteristic soil infiltration properties and outlet discharge were collected through a survey of a sample tertiary unit of Maira Branch Canal, Khyber Pakhtunkhwa Province, Pakistan. The performance of all fields in the tertiary unit was analysed and reported in aggregate, with detailed results of one field presented for illustration. The objective is to determine the optimum field layout, defined as the number of border strips, for the observed field characteristics to maximize performance. The results indicate that performance improvement is relatively easily achievable through changes in field layout within current irrigation services. Estimated application efficiency is sensitive to the selected depth of application, and it is important that a practical depth of application is selected. We recommend a depth of application of 50 mm and show how this is achievable and leads to a low quarter distribution uniformity of 0.750 and an application efficiency of 80 %. We also explore the feasibility of a 10-day warabandi rather than the 7-day warabandi and show that there is no significant change in the performance under a 10-day warabandi.  相似文献
