Although neutralizing antigenic sites of foot-and-mouth disease virus (FMDV) can be defined by selection of monoclonal antibody (MAb) escape mutants, no conformational neutralizing epitope on the major antigenic site located on the G-H loop of type Asia1 FMDV has been precisely mapped. In this study, we generated a potent neutralizing MAb 3E11, which recognized a conformation-dependent epitope and neutralized FMDV Asia1/YS/CHA/05 in vitro. Importantly, a dose of 5.5 NT(50) of the MAb 3E11 completely protected suckling mice from a dose of 10 LD(50) of homologous virus challenge in vivo. Through a 12-mer random peptide phage display, synthetic peptide analysis and constructing a series of FMDV Asia1/YS/CHA/05 mutants using reverse genetic system, we finely mapped the neutralizing epitope as the 12-amino acid peptide (141)SXRGXLXXLXRR(152). These results provide additional insights into the virus-MAb interaction at the amino acid level and may help in the development of an epitope-based Asia1 FMDV vaccine. 相似文献

Effects of vitamin C supplementation on growth performance and antioxidant status of layer ducklings
The study was conducted to evaluate the effects of increasing dietary vitamin C supplements on growth performance, antioxidant status and humoral immunity of layer ducklings. The results showed that the body weight and daily body weight gain of ducklings increased (p < 0.05) with increasing dietary vitamin C supplementations and reached a maximum at 400 mg vitamin C/kg feed. The dietary vitamin C supplementations reduced the malondialdehyde concentration (p < 0.05) and increased the activities of superoxide dismutase and glutathione peroxidase (p < 0.05) in serum and liver of ducklings at day 5 and 28. Additionally, feeding 400 mg/kg or 800 mg vitamin C/kg feed increased IgM and IgA concentrations in sera (p < 0.05) and serum IgG concentrations increased (p < 0.05) following supplementation in concentrations of 150-800 mg vitamin C/kg feed. In conclusion, the results suggested that dietary vitamin C supplements of 400 mg/kg feed provide optimal effects on growth performance, antioxidant status and parameters of humoral immunity. 相似文献

One of the crucial factors for evaluation of an effective genetically engineered vaccine is whether susceptible animals are protected from virus challenge after vaccination. In this study, a recombinant pseudorabies virus (PRV-P12A3C) that expressed capsid precursor polypeptide P12A and nonstructural protein 3C of foot-and-mouth disease virus (FMDV) was used as a vaccine. The expression of P12A3C and its immunogenicity and protective efficacy against FMDV challenge were measured. Humoral and cellular immune responses were evaluated after each immunization. Subsequently, each piglet was challenged with 1000 ID50 (50% infection dose) FMDV serotype O, named OR/80, which is used to produce vaccine in China. PRV-P12A3C induced a high level of neutralizing antibody and FMDV-specific lymphocytes. Inactivated vaccine provided 100% protection, and the vector strain (TK−/gE−/gI−) showed no protection. PRV-P12A3C induced 60% protection, compared with piglets that were vaccinated with TK−/gE−/gI−. 试验旨在考察枯草芽孢杆菌B1与中性蛋白酶协同发酵和酶解处理(简称菌酶协同酵解)制备菜籽肽的最佳工艺条件及对经菌酶酵解后的菜籽粕营养成分的分析。以菜籽肽得率为主要指标,通过对加酶量、发酵温度和料水比等酶解、发酵条件的单因素和正交试验,确定了菜籽肽以菌酶同时添加为最佳制备方式,以枯草芽孢杆菌接种量1.0%、中性蛋白酶添加量150 U/g、料水比1:1.0、发酵温度35℃、发酵时间48 h为最佳菌酶协同酵解条件。在此条件下,菜籽粕中硫代葡萄糖苷(硫苷)的降解率达74.23%,单宁和植酸的降解率分别为31.17%和16.39%。发酵后菜籽粕中粗蛋白含量达41.30%,比发酵前提高1.92%,菜籽肽含量达170.12 mg/g,比发酵前提高了3.7倍,还原糖含量比发酵前提高了57.68%,钙、磷含量分别比发酵前提高了26.83%和11.57%。试验表明,菌酶协同处理的方式对菜籽粕品质的提升有较好作用。 相似文献

Two experiments were undertaken to select the efficient method applied for goat estrous synchronization. In experiment 1, a total of 120 does (Capra hircus) were divided into five groups with a randomized block design, and the does of treatment 1 were synchronized by intravaginal sponges impregnated with 30 mg Levonorgestrel inserted for 10 days. Does of treatments 2, 3, and 4 were treated with further injection of 25 IU follicle-stimulating hormone (FSH), 0.05 mg prostaglandin F2 alpha (PGF2α), and 25 IU FSH + 0.05 mg PGF2α at sponge withdrawal, respectively. The does in the control group (n = 40) without estrous synchronization treatment and natural estrous does were observed. In experiment 2, a total of 140 does in five goat farms in breeding and non-breeding seasons were treated with the selective efficient procedure. The results presented that all the employed treatments were capable of inducing and synchronizing estrous goats. According to estrous response and economy, the use of intravaginal sponges impregnated with 30 mg Levonorgestrel and 0.05 mg PGF2α (treatment 3) is the first choice for estrous synchronization, and 95.0% of synchronized does demonstrated estrus, which was significantly higher than that of treatment 1 (P < 0.05) and control group (P < 0.01). The percentages of ovulating of treatments 3 and 4 were the same (95.0%), which were significantly higher than that of treatment 1 (P < 0.01). The ovulation rates among different groups were not significant (P > 0.05). When the selective procedure was applied to five goat farms, 85.7% (120/140) of does demonstrated estrus, and the kidding percentage, litter size, and prolificacy rate were 53.6%, 0.95, and 177%, respectively. 相似文献

根据中医利用解表、清瘟、凉血止血的中草药防治人类流感的原理,选取金银花、北板蓝根、贯叶连翘、双黄连、贯众等五种中草药所制成的制剂进行H5亚型禽流感病毒感染鸡胚后毒力作用的抑制实验。首先对鸡胚接种禽流感H5亚型病毒,然后对感染鸡胚分别注射上述几种中药制剂,再抽取胚液进行HA和HI实验,将其中对鸡源H5-AIV有抑制作用的中草药挑选出来,测定中草药对禽流感病毒的治疗指数(Therapeutic index,TI)。然后采用寇氏法计算药物半数有效量ED50。结果表明,金银花、北板蓝根、贯叶连翘、双黄连、贯众五种中草药制剂在对H5亚型禽流感病毒的鸡胚毒性方面均有较好的抑制作用。 相似文献

利用从柱状黄杆菌(Flavobacterium columnare)、嗜水气单胞菌(Aeromonas hydrophila)和温和气单胞菌(Aeromonas sobria)中提取的外膜蛋白(outer membrane protein,OMP)作为免疫原,注射接种团头鲂(Megalobrama amblycephala)后,通过测定受免鱼的交叉凝集抗体效价、肾脏和血液中吞噬细胞的吞噬活性和采用A.hydrophila活菌攻毒方法,比较了3种致病菌OMP对团头鲂的免疫原性。试验结果表明,从3种致病菌中提取的OMP对团头鲂均具有较强的免疫原性,受免鱼的血清中存在对3种致病菌的交叉凝集抗体,与对照鱼相比,肾脏和血液中吞噬细胞对吞噬原的吞噬活性和对A.hy-drophila活菌攻毒的相对免疫保护率(relative percent survival,RPS)明显上升。说明在3种致病菌的OMP上不同程度地存在共同保护性抗原。 相似文献
