OBJECTIVE: To determine whether exposure of canine cancer cells to histone deacetylase (HDAC) inhibitors S(+)-N-hydroxy-4-(3-methyl-2-phenyl-butyrylamino)benzamide (OSU-HDAC42) or suberoylanilide hydroxamic acid (SAHA) results in increased histone acetylation and decreased cell viability and whether any changes in viability involve induction of apoptosis or alterations in progression of the cell cycle. Sample POPULATION: 9 canine cancer cell lines. PROCEDURES: Cells from 9 canine cancer cell lines were treated with dimethyl sulfoxide vehicle, OSU-HDAC42, or SAHA, then assays of cell viability were performed. Histone acetylation was assessed by use of western blot analysis. Apoptosis was assessed via ELISA to detect fragmentation of cytoplasmic nucleosomal DNA and western blot analysis to detect cleavage of caspase 3. Cell cycle analysis was performed by use of propidium iodide staining and flow cytometry. RESULTS-Concentrations of OSU-HDAC42 and SAHA required to achieve 50% inhibition of cell viability (IC(50)) were reached in cells of 6 and 4 canine cancer cell lines, respectively, and ranged from approximately 0.4 to 1.3 microM for OSU-HDAC42 and 0.6 to 4.8 microM for SAHA. Cells from T-cell lymphoma, mast cell tumor, osteosarcoma, and histiocytic sarcoma lines were most sensitive to HDAC inhibition, with IC(50)s of < 1 microM for OSU-HDAC42 and < 5 microM for SAHA. Induction of apoptosis was indicated via cleavage of caspase 3 and increases in cytoplasmic nucleosomes and the subG(1) cell population. CONCLUSIONS AND CLINICAL RELEVANCE: Micromolar concentrations of HDAC inhibitors OSU-HDAC42 and SAHA induced histone acetylation, cytotoxicity, and apoptosis in canine cancer cells. The Shuttle Experiment conducted between Hawaii and Tahiti from January 1979 to June 1980 was designed to observe the changing equatorial ocean structure and circulation, to study the variations and interactions of the four major equatorial ocean currents, and to develop a scientific basis for their monitoring by simple observations of thermal structure and sea level. Preliminary analyses of the results show that the equatorial thermal structure remains intact during a normal year and that only the positions and intensities of the currents are subject to change. The water transport of the equatorial undercurrent varied from 25 x 10(6) cubic meters per second in January to 51 x 10(6) cubic meters per second in July, but also exhibited strong short-term pulsations. The equatorial surface flow responded strongly to the winds at periods of 1 month and longer. An array of drifter buoys in the equatoral countercurrent was subject to very little dispersion while traveling over 4500 kilometers in 4 months. Low-frequency fluctuations in the North Equatorial Countercurrent can be monitored by means of the difference in sea level between Fanning and Majuro. 相似文献

Sixty male and 60 female crossbred pigs were allocated to an experiment to investigate the effects of porcine somatotropin (pST) administration (0 or 6 mg/d) and dietary lysine content on growth performance, tissue deposition, and carcass characteristics over the live weight range of 80 to 120 kg. Pigs receiving pST were given diets containing 6.9, 7.8, 8.8, 9.7, 10.6, or 11.5 g lysine/kg, whereas control pigs received diets containing 4.8, 5.8, 6.9, 7.8, 8.8 or 9.7 g lysine/kg. These dietary levels ranged from 0.40 to 0.70 g available lysine/MJ of DE for pST-treated pigs and from 0.28 to 0.58 g available lysine/MJ of DE for control pigs. Pigs were individually housed in pens, and there were five replicates of each treatment. All diets contained 14.5 MJ of DE/kg and were offered for ad libitum consumption to pigs between 80 and 120 kg live weight. Growth rate increased exponentially and food conversion ratio (FCR) decreased exponentially with increasing levels of lysine. In addition, there was a significant sex x pST interaction such that pST reduced the sex difference in FCR. Growth rate was faster in boars than in gilts and was increased by pST at the higher levels of dietary lysine. Similarly, FCR was lower for boars than for gilts and was decreased by pST at the higher dietary lysine levels. The optimum growth rate and FCR were defined as the lysine level at which growth rate and FCR were 95% and 105%, respectively, of the lysine plateau. The optimum growth rate and FCR were achieved at similar dietary lysine contents and were approximately 0.35 and 0.52 g available lysine/MJ of DE for control and pST-treated pigs, respectively. Protein deposition in the carcass increased exponentially with increasing dietary lysine level, was higher in boars than in gilts, and was increased by pST at the higher dietary lysine contents. Sex had no effect on dietary lysine required to maximize protein deposition. The dietary lysine contents required to ensure 95% of plateau protein deposition of 104 and 153 g/d were 0.39 and 0.55 g available lysine/MJ of DE for control and pST-treated pigs, respectively. Two studies were conducted to determine the effect of diets containing low-phytate barley or supplemented with phytase on P balance and excretion in grower pigs. In Exp. 1, eight 32-kg barrows were assigned to a repeated, 4 x 4 Latin square design and fed 4 diets that contained 96% barley: normal-phytate hulled barley (HB), low-phytate hulled barley (LPHB), normal-phytate hull-less barley (HLB), and low-phytate hull-less barley (LPHLB). The barley cultivars contained 0.16, 0.05, 0.24, and 0.03% phytate, respectively. Inorganic P (iP) was added to the HB and HLB diets to meet the 1998 National Research Council recommendation of available P (aP, 0.23%), whereas LPHB and LPHLB contained sufficient aP. The diets were fed at 2.5 times the maintenance requirement for ME. The apparent total tract digestibilities (ATTD) of P did not differ between the hulled and hull-less barley diets, but P retention (%) and excretion were greater in pigs fed the hull-less barley diets (P < 0.05). The ATTD of P was greater and P excretion was 35% lower in pigs fed the low-phytate compared with the normal-phytate diets (P < 0.001). The amount of P retained (g/d) was greater (P < 0.001) in pigs fed low-phytate barley, reflecting an ATTD of P of 65 and 49% for low-phytate and normal-phytate barley, respectively (P < 0.001). In Exp. 2, eight 21-kg barrows were assigned to a repeated, 4 x 4 Latin square design and fed 4 diets based on barley and soybean meal (SBM): HB-SBM, HB-SBM + iP, HB-SBM + phytase, and LPHB-SBM. The HB-SBM and HB-SBM + phytase diets were deficient in aP, whereas the HB-SBM + iP and LPHB-SBM diets had adequate aP. The feeding regimen was similar to that of Exp. 1. Adding iP to the HB-SBM diet did not affect the ATTD but increased the amount of P retained (g/d) and excreted (P < 0.001). The ATTD and amount of P retained (g/d) did not differ among pigs fed the HB-SBM + iP, HB-SBM + phytase, and LPHB-SBM diets. However, pigs fed the HB-SBM + phytase and LPHB-SBM diets excreted 32 and 29% less P, respectively, than pigs fed the HB-SBM + iP diet (P < 0.05), confirming that low-phytate barley is as effective as supplemental phytase in improving P digestibility and utilization and decreasing P excretion in grower pigs. 相似文献

Lactoferrin concentration (LFC) in normal and mastitic milk of dairy goats were examined. LFC in bulk milk collected from 70 dairy goat farms and individual milk samples from 10 goats with mastitis were measured by enzyme-linked immunosorbent assay (ELISA) and their reaction time in methylene blue reduction test (MBRT) monitored. Bulk milk samples were categorized into three grades, such as high, normal and low, based on the reaction time in MBRT. The mean LFC in milk that was considered high quality (167 microg/ml) was significantly lower than that of those graded as normal (218 microg/ml) and low quality (304 microg/ml), while mean LFC in mastitic milk was 587 microg/m l. The correlation coefficient between milk LFC and MBRT time was found to be -0.7. Three goats were inoculated with Staphylococcus aureus into one of their udder halves. The mean milk LFC was found to be significantly higher (1500 microg/ml) than the control (30 microg/ml). These findings suggest that milk LFC may be useful as an index for intramammary infection in goats. 相似文献

Preliminary results from the rearward-looking electrostatic analyzer of the plasma science experiment during the Mariner 10 encounter with Venus are described. They show that the solar-wind interaction with the planet probably involves a bow shock rather than an extended exosphere, but that this is not a thin boundary at the point where it was crossed by Mariner 10. An observed reduction in the flux of electrons with energies greater than 100 electron volts is interpreted as evidence for somne direct interaction with the exosphere. Unusual intermittent features observed downstream of the planet indicate the presence of a comet-like tail hundreds of scale lengths in length. 相似文献

The fungal pathogen Verticillium dabliae was less likely to cause symptoms of verticillium wilt on cotton seedlings that had been previously exposed to spider mites than on unexposed cotton seedlings. Conversely, populations of the spider mite Tetranychus urticae grew less rapidly on seedlings that had been inoculated with V. dabliae than on uninoculated controls. Changes caused by pathogen or herbivore attack reduced the suitability of the host plant to a diversity of organisms. This result suggests that highly unrelated organisms that share a host plant may interact strongly. 相似文献
