The effects of feeding different levels of forages and concentrate, in a discontinuous feeding pattern, on the efficiency of feed utilisation and rumen function were studied using rumen fistulated sheep. Experiment 1 was a 4 × 4 latin square design to determine the whole tract digestibility and rumen characteristics of diets comprising 15% (C15), 25% (C25), 35% (C35) and 45% (C45) concentrate (energy-dense dairy pellets) with the rest of the diet being a combination of fresh short rotation ryegrass (Lolium mutiforum) and conserved (lucerne hay and maize silage) forages. In Experiment 2, the rumen degradation characteristics of feed ingredients were determined using the nylon bag technique. Daily dry matter intake (either expressed as g/kg LW or g/kg W 0.75) was 10% lower (p = 0.03) for the C15 diet compared with C25, C35 and C45 diets. The apparent in vivo digestibility of dry matter (DM) for C15 diet was 4% higher (p = 0.04) than the C35 and C45 diets which may be attributed to the high quality of the forage (ryegrass) used.Fibre digestibility decreased as proportion of concentrate in the diet increased. However, this was unlikely due to changes in the rumen fermentation pattern, as neither pH (6.1 ± 0.23) nor ammonia concentration (24.4 ± 6 mg/dl), were different (p > 0.05) among diets. Instead, the lower fibre digestion was most likely the result of different type and proportion of fibre among diets, as total rumen degradability and rate of fibre degradation in the rumen were higher (p = 0.001) for ryegrass than for other feedstuffs. There was no significant difference in total nitrogen balance and urinary allantoin excretion among diets, which indicated similar total microbial protein synthesis (MPS). The asynchrony observed, for N and energy availability in the rumen for different diets using Sinclair et al. [Sinclair, L.A., Garnsworthy, P.C., Newbold, J.R., Buttery, P.J., 1993. Effect of synchronizing the rate of dietary energy and nitrogen release on rumen fermentation and microbial protein synthesis in sheep. J. Agric. Sci. 120, 251–263] equation, was due to the feeding pattern used in this study leading to excess of N in relation to total organic matter digested in the rumen. In conclusion, feeding concentrates in the diets as PMR with conserved forages in a discontinuous feeding pattern may be valuable to develop feeding strategies in a pasture based system for high producing dairy cows without affecting the rumen system.  相似文献

Pharmacokinetics, urinary excretion and plasma protein binding of danofloxacin was investigated in buffalo calves following intravenous administration at the dose rate of 1.25 mg/kg to select the optimal dosage regimen of danofloxacin. Drug concentrations in plasma and urine were measured by microbiological assaying. In vitro plasma protein binding was determined employing the equilibrium dialysis technique. The distribution and elimination of danofloxacin were rapid, as indicated by values (mean ±SD) of distribution half-life (t1/2α = 0.16 ± 0.07 h) and elimination half-life (t1/2β = 4.24 ± 1.78 h), respectively. Volume of distribution at steady state (Vss) = 3.98 ± 1.69 L/kg indicated large distribution of drug. The area under plasma drug concentration versus time curve (AUC) was 1.79 ± 0.28 μg/mlxh and MRT was 8.64 ± 0.61 h. Urinary excretion of danofloxacin was 23% within 48 h of its administration. Mean plasma protein binding was 36% at concentrations ranging from 0.0125 μg/ml to 1 μg/ml. On the basis of pharmacokinetic parameters obtained, it is concluded that the revision of danofloxacin dosage regimen in buffalo calves is needed because the current dosage schedule (1.25 mg/kg) is likely to promote resistance.  相似文献

Recent advances in semiconductor technologies have given rise to the development of mid‐infrared (mid‐IR) spectrometers that are compact, relatively inexpensive, robust and suitable for in situ proximal soil sensing. The objectives of this research were to evaluate a prototype portable mid‐IR spectrometer for direct measurements of soil reflectance and to model the spectra to predict sand, clay and soil organic matter (SOM) contents under a range of field soil water conditions. Soil samples were collected from 23 locations at different depths in four agricultural fields to represent a range of soil textures, from sands to clay loams. The particle size distribution and SOM content of 48 soil samples were measured in the laboratory by conventional analytical methods. In addition to air‐dry soil, each sample was wetted with two different amounts of water before the spectroscopic measurements were made. The prototype spectrometer was used to measure reflectance (R) in the range between 1811 and 898 cm?1 (approximately 5522 to 11 136 nm). The spectroscopic measurements were recorded randomly and in triplicate, resulting in a total of 432 reflectance spectra (48 samples × three soil water contents × three replicates). The spectra were transformed to log10 (1/R) and mean centred for the multivariate statistical analyses. The 48 samples were split randomly into a calibration set (70%) and a validation set (30%). A partial least squares regression (PLSR) was used to develop spectroscopic calibrations to predict sand, clay and SOM contents. Results show that the portable spectrometer can be used with PLSR to predict clay and sand contents of either wet or dry soil samples with a root mean square error (RMSE) of around 10%. Predictions of SOM content resulted in RMSE values that ranged between 0.76 and 2.24%.  相似文献

Selenium (Se) is an essential nutrient for animals, humans, and microorganisms, but its role in the plants needs further exploration. It is considered beneficial at low levels, but is toxic at higher levels, and there is a fine boundary between these concentrations. Generally, Se levels less than 1 mg kg?1 have been found to be beneficial for the plants while higher levels cause toxicity in most of the agricultural crops. At low concentrations, Se can act as a plant growth regulator, antioxidant, anti-senescent, abiotic stress modulator, and defensive molecule against pathogens in plants. At higher concentrations, plants show various toxic symptoms, which include stunting of growth, chlorosis, withering, and drying of leaves, decreased protein synthesis premature and even death of the plant. The roles of selenium as enhancer and inhibitor of plant growth in various agricultural crops are discussed here with recent updates. Biofortification of some crops with Se using agronomic and genetic approaches is being explored to cultivate them in the regions having Se-deficiency in foods. Strategies of phytoremediation of Se in hyperaccumulators and transgenic plants overexpressing enzyme/proteins to increase Se tolerance are also described.  相似文献

Abstract

A pot experiment was conducted to study the interaction effects of phosphorus and copper on wheat. The soils used were calcareous loamy sand (ls) and non calcareous sandy loam (sl). Four levels of Cu (0, 5, 10 and 20?mg Cu kg?1 soil) and six levels of P (0, 25, 50, 100, 200 and 400?mg P kg?1 soil) were applied in all possible combinations with three replications. Soil pH decreased with Cu application while Olsen P increased with P application in both soils. Growth and yield of wheat improved significantly with graded levels of applied P. However, when any level of P was combined with 20?mg Cu kg?1 soil, severe iron chlorosis of leaves, a drastic reduction in growth and chlorophyll content was observed in calcareous ls only. The results indicated that it was Cu and not P that induced Fe deficiency in wheat grown in alkaline calcareous soil and the Cu requirement of the crop seemed to be much lower in the calcareous ls. Root dry matter, grain and straw yield decreased with increasing levels of applied Cu in ls but in sl maximum increase of 62.5, 74.3 and 63.7 per cent in root, grain and straw yield was observed with a combined application of 400?mg P and 5?mg Cu kg?1 soil over control. Accumulation of Cu in roots decreased the Fe absorption by roots which indicated that Fe chlorosis of wheat leaves is expected when Cu: Fe concentration ratio in root is > 0.30.  相似文献

ABSTRACT

A population of recombinant inbred lines (RILs) developed from a cross of Triticum aestivum cvs. WL711 and PH132 using the single-seed descent method was used to investigate the genetics of high protein content and its contribution to bread-making quality. Four-year data on protein content of parents and 124 RILs suggested that the difference in protein content between the two parents was controlled by two major genes with an additive effect. The individual gene gave intermediate protein content. Seven RILs with high protein content and without any yield penalty were selected, multiplied, and investigated for various milling, dough, and bread-making characteristics. PH132 and all seven selected RILs had higher protein content, higher sodium dodecyl sulphate (SDS) sedimentation value, longer dough development time, a lower mixing tolerance index, higher stability, and better loaf volume and loaf quality than the bread wheat cultivars WL711 and PBW343. High molecular weight (HMW) glutenin subunit composition of various high-protein RILs indicated a recombination of parental protein subunits. “2?” coded by Glu-A1 locus, 17 + 18 subunits coded by Glu-B1 locus, and “5 + 10” coded by Glu-D1 locus were predominant. One of the selected RILs, T-74, possessing 2 + 12 at Glu-D1 locus, also had superior bread-making quality, indicating that grain-protein content above a certain minimum value is a relatively more important determinant of bread-making quality than HMW glutenin subunits 5 + 10.  相似文献

Mature and healthy male house rats, Rattus rattus (n= 160) were fed on bait (cracked wheat: powdered sugar, 98:2) containing different concentrations of triptolide (0.1, 0.05, 0.025 and 0%) for 7 and 14 days in no‐choice and bi‐choice feeding tests in the laboratory. The objective of the study was to record the antifertility affects of triptolide after 30 and 60 days of termination of treatment. Results revealed no significant effect of triptolide treatment on weights of testis, epididymis, seminal vesicles and prostate gland of rats. Overall, sperm motility, live sperm count, sperm density and sperm morphology in the cauda epididymal fluid were found to differ significantly (P≤ 0.05) between untreated and treated groups of rats. The major effect of triptolide on sperm morphology was in the form of sperm head tail separation, which was up to 56.0% in rats treated for 14 days in no‐choice and autopsied after 30 days. A significant effect (P≤ 0.05) of triptolide treatment was observed on the histomorphology of the testis, which included a dose‐dependent decrease in diameter of seminiferous tubules, thickness of germinal epithelium and numbers of various spermatogenic cells. Cell associations in the seminiferous epithelial cycle were poorly developed in rats ingesting medium (4.7–5.1 mg/100 g bw) and high doses (6.9–7.2 mg/100 g bw) of triptolide than rats ingesting low doses (1.8–2.3 mg/100 g bw) and untreated rats. The cell stages affected had not recovered fully within the 60 day period following triptolide withdrawal. The present study suggests the potential of triptolide in reproductive management of Rattus rattus.  相似文献
