Rubus spectabilis Pursh. was introduced as a donor of earliness in summer fruiting raspberries. F, hybrids between R. ideaus and R. spectabilis were more or less sterile but flowered very early. Fertility was almost fully restored in the BC[ generation. Some BC, individuals were early summer fruiting while some were early primocane fruiting. Using these as parents, further generations were raised and individuals with cultivar potential have been selected in the BC2 generation. The F, showed resistance to cane diseases and this resistance was largely retained in BC, and BC3 summer fruiting progenies. The two F, individuals used as parents were highly resistant to Phytophthora root rot and ten 2nd or 3rd backcross derivatives of R. spectabilis were moderately resistant. 相似文献

Compact growth is an important quality criterion in horticulture. Many Campanula species and cultivars exhibit elongated growth which is suppressed by chemical retardation and cultural practice during production to accommodate to the consumer’s desire. The production of compact plants via transformation with wild type Agrobacterium rhizogenes is an approach with great potential to produce plants that are non-GMO. Efficient transformation and regeneration procedures vary widely among both plant genera and species. Here we present a transformation protocol for Campanula. Hairy roots were produced on 26–90% of the petioles that were used for transformation of C. portenschlagiana (Cp), a C. takesimana × C. punctata hybrid (Chybr) and C. glomerata (Cg). Isolated hairy roots grew autonomously and vigorously without added hormones. The Cg hairy roots produced chlorophyll and generated plantlets in response to treatments with cytokinin (42 µM 2iP) and auxin (0.67 µM NAA). In contrast, regeneration attempts of transformed Cp and Chybr roots lead neither to the production of chlorophyll nor to the regeneration of shoots. Agropine A. rhizogenes strains integrate split T-DNA in TL- and TR-DNA fragments into the plant genome. In this study, regenerated plants of Cg did not contain TR-DNA, indicating that a selective pressure against this T-DNA fragment may exist in Campanula. 相似文献

Transformation of Kalancho? blossfeldiana ‘Molly’ using the naturally occurring bacterium Agrobacterium rhizogenes is a non-GMO strategy to breed compact plants. In the present study, crosses resembling a commercial breeding strategy were made to determine if the improved ornamental quality observed in the T1 generation was inherited to the offspring. F1 lines were produced by crossing the commercial Kalancho? cultivar ‘Sarah’ with a selected T1 line. Subsequently, F2 populations were produced by self-pollination of individual selected lines. The rol-genes were inherited to the progeny and the presence of rol-genes was confirmed in all F1 and many F2 plant lines exhibiting dwarfism. Screening of F1 and F2 plants showed that the rol-genes were inherited together. Besides decreased plant height, several F1 and F2 lines containing rol-genes exhibited changes in plant diameter, number of branches, flower diameter and time to first open flower and duration of flowering compared to the WT ‘Molly’. Furthermore, increased ethylene tolerance was observed in several lines containing rol-genes compared to the WT ‘Sarah’. Screening of three selected F2 populations derived from self-pollination of F1 lines containing rol-genes indicated a shift in distribution towards a lower mean plant height within the entire plant population compared to a control population of plants without rol-genes. Moreover, one of these F2 populations also exhibited earlier flowering compared to the control population. Compact potted plants and lines without delayed flowering and with improved ethylene tolerance were obtained and are valuable in commercial breeding programmes without using recombinant DNA technology. 相似文献

There is growing interest in the applications of soil enzymes as early indicators of soil quality change under contrasting agricultural management practices. However, despite there being an abundant literature on this subject, most comparative assessments have been based on a limited number of experimental farms and, therefore, conclusions are not as robust as desired. In this study, we compare 18 pairs of organic and neighbouring conventional olive orchards in southern Spain. These sites were selected to allow the definition of the relative contributions of site-landscape features, soil type, and time since organic accreditation and tillage intensity, on the soil quality. Soils were analysed for physico-chemical properties, the activities of dehydrogenase, β-glucosidase, arylsulphatase, acid and alkaline phosphatase, and potential nitrification. The geometric mean of the assayed enzymes (GMea) was validated with an independently performed Principal Component Analysis (PCA), and used as a combined soil quality index. The effects of tillage intensity and time since organic accreditation on the improvement of soil quality were also evaluated within the subset of organic farms. Overall for the 18 sites, contrasted management practices did not differ in their impact on basic soil physico-chemical properties, except for loss of on ignition and available inorganic N which were higher and lower in organic farms, respectively. Organic management resulted in significantly higher soil enzyme activities. However, differences were not significant in some of the paired comparisons when considered individually. This highlights the need for extensive comparative assessment, as in this study, to draw clear conclusions concerning the changes to soil quality under sustainable management practices. The GMea was significantly correlated with the first axis of the PCA and shown to be appropriate for condensing the set of soil enzyme values to a sole numerical value. Soil quality changes in organic versus conventional farms, as measured by the GMea, ranged from −23% to 97%, and was highly dependent on time since organic accreditation (r = 0.88; P < 0.01). The appearance of 2,4,6-trichloroanisole (TCA) in cork stoppers is of great concern because it can cause off-flavors in bottled wine. To prevent this sensorial defect, there should not be any traces of 2,4,6-trichlorophenol (TCP), 2,3,4,6-tetrachlorophenol (TeCP), or pentachlorophenol (PCP) in the finished corks, because they are the direct precursors of TCA. In the course of this study two methodologies based upon an extraction with ethanol/water mixtures to determine the chlorophenolic content in cork matrices were developed. The cork extract is preconcentrated using both solid-phase extraction and solid-phase microextraction methodologies. The latter was optimized by applying a full two-level factorial design. Finally, spiked ground corks at nanogram per gram levels of each chlorophenol were analyzed under optimal conditions and by applying both procedures. The obtained results demonstrate that chlorophenols can be detected in corks contaminated at the nanogram per gram level and, thus, these approaches can be successfully applied as quality control measures in the cork industry. 相似文献

In this study the estimation of reflectivity at 1730 MHz (l-band), measured with a microwave digital cordless telephony (DCT) patch antenna, is presented as an easy-to-handle and non-destructive new method to assess the relative water content (RWC) of poplar leaves and filter discs at different levels of dehydration. The accuracy of this new method has been contrasted with the R1300/R1450 index, determined by a portable near infrared (NIR) spectrometer. The close correlations found between RWC and the reflectivity at a frequency of 1730 MHz, both for filters and leaves, indicate that microwave determinations are rather independent of the physical properties of the material analysed. On the contrary, the differences found between poplar leaves and leaf filters in the relationships established between RWC and the R1300/R1450 index demonstrate a strong influence of the properties of the material in NIR reflectance measurements, specifically as they relate to changes in leaf thickness during dehydration. It should be noted that the amount of energy received by the leaf for the microwave technique (0.1 mW) was much lower than that received for the measuring of the R1300/R1450 index (2.5 W). Moreover, R-square coefficients were higher for microwaves than for the R1300/R1450 index. The use of a technologically simple, low cost and portable device, based on a microwave DCT patch antenna, could yield a solid support for the development of a commercial apparatus enabling the determination of plant water status under field conditions. 相似文献

Biological control of Tetranychus urticae Koch (Acari: Tetranychidae), a key pest of clementines, can be improved in this crop with the establishment of a ground cover of Festuca arundinacea Schreber (Poaceae). This cover houses an abundant and diverse community of predatory Phytoseiidae mites including Euseius stipulatus (Athias-Henriot), Neoseiulus barkeri Hughes, Neoseiulus californicus (McGregor) and Phytoseiulus persimilis Athias-Henriot and a dense population of the grass thrips Anaphothrips obscurus Müller (Thysanoptera: Thripidae) throughout the year. The aim of this study was to determine whether the presence of this thrips species could be related to the improvement in the biological control of T. urticae. Therefore, the capacity of the abovementioned phytoseiids to feed and reproduce on A. obscurus and their feeding preferences when T. urticae and A. obscurus were simultaneously offered, were analyzed. The results show that E. stipulatus, N. barkeri and N. californicus have a type II functional response when offered A. obscurus nymphs, whereas P. persimilis barely feeds on this thrips species. Furthermore, N. barkeri and N. californicus can reproduce feeding only on thrips. Regarding prey preference, the Tetranychus spp.-specialist P. persimilis preferably preyed on T. urticae, the generalists N. barkeri and E. stipulatus preferred A. obscurus, and the selective predator of tetranychid mites N. californicus showed no preference. Therefore, we hypothesize that the enhanced biological control of T. urticae observed could be related to A. obscurus becoming an alternative prey for non-specialist phytoseiids, without altering the control exerted by the T. urticae-specialist P. persimilis and likely reducing intraguild predation. 相似文献
