There is an urgent need for new antibiotics to treat hospital- and community-associated methicillin-resistant Staphylococcus aureus (MRSA) infections. Previous work has indicated that both terrestrial and marine-derived members of the napyradiomycin class possess potential anti-staphylococcal activities. These compounds are unique meroterpenoids with unusual levels of halogenation. In this paper we report the evaluation of two previously described napyradiomycin derivatives, A80915A (1) and A80915B (2) produced by the marine-derived actinomycete, Streptomyces sp. strain CNQ-525, for their specific activities against contemporary and clinically relevant MRSA. Reported are studies of the in vitro kinetics of these chemical scaffolds in time-kill MRSA assays. Both napyradiomycin derivatives demonstrate potent and rapid bactericidal activity against contemporary MRSA strains. These data may help guide future development and design of analogs of the napyradiomycins that could potentially serve as useful anti-MRSA therapeutics. 相似文献

The source ofS. phureja cytoplasm, aS. phureja×S. chacoense F1, used as maternal parent favoured lower berry and seed set. During successive backcrosses progeny expressions approached
those of the recurrent ssp.tuberosum pollen parents. A low level of pollen sterility occurred in BC2 plants, unrelated to direction of cross or cytoplasmic factors. By BC4, seed set and fertility were as good as ssp.tuberosum, and recurrent backcrossing could be terminated without loss of seed production or fertility.

The Mediterranean sponge Hamigera hamigera (family Anchinoideae) was studied since its total extract showed deterrent activity in a fish feeding assay. Eight compounds were isolated from the biologically active fractions and four of these proved to be new natural products, hamigeroxalamic acid (1), hamigeramine (2), hamigeramide (3) and hamiguanosinol (5). In extensive farmer‐led trials practicing conservation farming (CF) in three regions of Zambia (Mongu: sandy soils; Kaoma: sandy or loamy sand soils; Mkushi: sandy loam or loamy soils), we studied the effects of biochar made of maize cobs (0, 2, and 6 t ha?1 corresponding to 0, 0.8, and 2.5% per basin) at different fertilizer rates of NPK and urea on crop yield of maize (Zea mays) and groundnuts (Arachis hypogaea). Conservation farming in this case combines minimum tillage (how basins), crop rotation and residue retention. For the first time, the effect of biochar on in situ soil nutrient supply rates [determined by buried Plant Root Simulator (PRS?) exchange resins] was studied, as well as the effects of biochar on elemental composition of maize. Effects of 0–10% (w:w) biochar addition on soil physical and soil chemical properties were determined in the laboratory. At all sites there was a consistent positive response in crop yield upon the addition of biochar. However, due to a great variability between farms there were no significant differences in absolute yields between the treatments. In the sandy soils at Mongu, relative yields (i.e., percentage yield with biochar relative to the same fertilizer rate without biochar) of maize grains and maize stover were significantly increased at recommended fertilizer rates (232 ± 60%) and at half the recommended rate (128 ± 6%), respectively. In addition, biochar significantly increased concentrations of K and P in maize stover. In situ soil nutrient supply rates as measured by PRS?‐probes were highly spatially variable with no consistent effects of the different treatments in the three regions. By contrast, the fraction of plant available water (Vol.‐%) significantly increased upon the addition of biochar in all three soils. The increase caused by 10% biochar addition was of factor 2.5 in Mongu (from 4.5% to 11.2%) and 1.2 in both Kaoma (from 14.7% to 18.2%) and Mkushi (from 18.2% to 22.7%). Cation exchange capacity, pH, and exchangeable K significantly increased upon the addition of 10% (w:w) biochar in all three regions with a subsequent increase in base saturation and decrease of available Al3+. Our findings suggest that the addition of biochar in combination with CF might have a positive impact on crop growth and that this positive effect is mainly caused by increases in plant‐available water and decreased available Al. 相似文献

With the aim of determining whether the arbuscular mycorrhizal (AM) inoculation would give an advantage to overcome salinity
problems and if the phosphorus (P) concentration can profoundly influence zucchini (Cucurbita pepo L.) plant responses to AM, a greenhouse experiment was carried out with AM (+AM) and non-AM (−AM). Plants were grown in sand
culture with two levels of salinity (1 and 35 mM NaCl, giving electrical conductivity values of 1.8 and 5.0 dS m−1) and P (0.3 and 1 mM P) concentrations. The percentages of marketable yield and shoot biomass reduction caused by salinity
were significantly lower in the plants grown at 0.3 mM P, compared to those grown at 1 mM P. However, even at high P concentration,
the absolute value of yield and shoot biomass of +AM zucchini plants grown under saline conditions was higher than those grown
at low P concentration. The +AM plants under saline conditions had higher leaf chlorophyll content and relative water content
than −AM. Mycorrhizal zucchini plants grown under saline conditions had a higher concentration of K and lower Na concentration
in leaf tissue compared to −AM plants. The P content of zucchini leaf tissue was similar for +AM and −AM treatments at both
low and high P concentrations in the saline nutrient solution. The beneficial effects of AM on zucchini plants could be due

We determined the effects of removal of leaves, stem axillary buds, or the entire shoot on root suckering (adventitious shoot formation by roots) and basal stem sprouts in 3- and 4-year-old potted seedlings of aspen (Populus tremuloides Michx.). The greatest number of root suckers (67.9 +/- 8.5 per plant) emerged after excision of the entire shoot. Defoliated and debudded stems were the major source of inhibitory agents for root suckering, although axillary buds and developing new leaves also exerted a significant inhibitory effect. Removal of mature leaves had only a minor effect on root suckering. Removal of a continuous band of bark (girdling) at the base of the stem consistently stimulated growth of adventitious shoots from the stem below the girdle and occasionally promoted root suckering. Exogenous application of indole-3-acetic acid to excised stumps inhibited root suckering and basal stem sprouting. Naphthylphthalamic acid (NPA), an auxin polar transport inhibitor, had no effect on root suckering or stem sprouting when it was applied to the bark of the basal stem. However, NPA significantly increased root suckering when it was applied to the exposed surface of xylem after girdling. These results suggest that polar transport of auxin in the xylem parenchyma is an important inhibitor of root suckering. On decapitated stems, vacuum extraction of xylem sap from the root system lowered the frequency of root suckering compared with decapitation alone, indicating that substance(s) originating in the root system also play a significant role in controlling root suckering. 相似文献

OBJECTIVE: To compare the efficacy of modified-live virus (MLV) vaccines containing either type 1 bovine viral diarrhea virus (BVDV) or types 1 and 2 BVDV in protecting heifers and their offspring against infection associated with heterologous noncytopathic type 2 BVDV challenge during gestation. DESIGN: Randomized controlled study. ANIMALS: 160 heifers and their offspring. PROCEDURES: After inoculation with a placebo vaccine, 1 or 2 doses of an MLV vaccine containing type 1 BVDV, or 1 dose of an MLV vaccine containing both types 1 and 2 BVDV, heifers were bred naturally and challenge exposed with a type 2 BVDV field isolate between 62 and 104 days of gestation. Pregnancies were monitored; after parturition, virus isolation and immunohistochemical analyses of ear-notch specimens were used to determine whether calves were persistently infected. Blood samples were collected at intervals from heifers for serologic evaluation and virus isolation. RESULTS: Persistent infection was detected in 18 of 19 calves from heifers in the control group and in 6 of 18 calves and 7 of 19 calves from heifers that received 1 or 2 doses of the type 1 BVDV vaccine, respectively. None of the 18 calves from heifers that received the type 1-type 2 BVDV vaccine were persistently infected. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that the incidence of persistent BVDV infection among offspring from dams inoculated with 1 dose of the MLV vaccine containing types 1 and 2 BVDV was decreased, compared with 1 or 2 doses of the MLV vaccine containing only type 1 BVDV. 相似文献

Five procyanidin fractions with different structural complexities were obtained after fractionation of a grape seed extract. The procyanidin fraction's abilities to inhibit lipid peroxidation induced by 2,2'-azobis-2-methyl-propanimidamide dihydrochloride in a liposomal membrane system were examined. The antioxidant capacities of all fractions were evaluated through monitoring oxygen consumption and by measuring the formation of conjugated dienes. All tested fractions provided protection of membranes against peroxyl radicals by increasing the induction time of oxidation. This effect increased up to fraction II but decreased with the increase of the structural complexity of further procyanidin fractions, possibly due to steric hindrance effects exhibited by the more complex fractions. In addition, the antiradical properties and the reducing power of these fractions were determined by using 2,2-diphenyl-1-picrylhydrazyl and ferric reducing/antioxidant power methods, respectively. Moreover, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium-bromide reduction and DNA synthesis were measured in Michigan Cancer Foundation 7 (MCF-7), a human breast cancer cell line, treated with catechin or procyanidin fractions in order to evaluate the effect of these compounds on cell viability and proliferation. The results obtained showed that at 30 microg/mL, fractions I and II decreased cell viability and proliferation, which was not observed with 60 microg/mL of the same fractions. Catechin was also able to decrease cell viability and proliferation at 30 and 60 microg/mL. It is interesting to notice that the procyanidin fractions that exhibited higher antioxidant activity were the same to affect cell viability and proliferation. 相似文献

Four anthocyanin-pyruvic adducts were synthesized through the reaction of cyanidin 3-O-glucoside, cyanidin 3-O-rutinoside, cyanidin 3-O-sophoroside, and cyanidin 3-O-sambubioside with pyruvic acid, structurally characterized by liquid chromatography-mass spectrometry (LC-MS) and nuclear magnetic resonance (NMR), and their chromatic properties were studied (pH and SO2 stability assays). Overall, these pigments were shown to have a higher resistance to discoloration toward pH variations and also in the presence of SO2, being that this resistance to discoloration was explained by a higher protection of the chromophore group against the water or bisulfite nucleophilic attack that gives rise to the colorless hemiacetal form. Only slight differences in the protection against the nucleophilic attack of water and bisulfite were found to occur between all of the cyanidin-pyruvic acid adducts studied. Indeed, anthocyanin-pyruvic acid adducts with glucose or sambubiose attached to the 3-O position of the flavylium moiety were shown to have smaller bleaching constants compared with similar pigments that possess a rutinosyl or sophorosyl moiety. The study of the pigments (A-D and cyanidin-3-O-glucoside) color parameters, namely, chroma (C), lightness (L), and the hue angle (h(a,b)), obtained from the CIELAB system, revealed that different patterns of sugars in the anthocyanin-pyruvic acid adduct moiety affected the referred three parameters of color. The loss of saturation (DeltaC < 0) and the increase of lightness (DeltaL > 0) presented by the cyanidin-pyruvic acid adduct solutions at acidic pH values (1.0 and 2.0) showed that they are much less colored than the cyanidin-3-O-glucoside. For higher pH values (5.0 and 7.0), the reverse trend was observed. This means that the cyanidin-pyruvic acid adducts A-D are much more colored than the anthocyanin at these pH values. The higher coloring capacity of these pigments at higher pH values may be an important feature, indicating a putative application of these compounds in food products. 相似文献
