Context
Knowledge of how environmental gradients generate changes in community composition across forest landscapes (β-diversity) represents a critical issue in the era of global change, which exerts especially powerful impacts by shifting disturbance regimes.Objectives
We analyzed the response of tree communities to increased disturbance rates that were linked to European settlement at the temperate-boreal interface of eastern Canada. We tested whether disturbance has led to spatial homogenization or heterogenization, and to decoupling or strengthening of community-environment relationships.Methods
We used a reconstruction of pre-industrial tree communities based on historical land survey records (1854–1935), together with modern data, to assess changes in tree β-diversity patterns. Then, β-diversity was partitioned into fractions explained by spatial (dbMEM) and environmental variables (latitude, elevation, slope, drainage and surface deposits) in order to assess changes in spatial structures and community-environment relationships.Results
In pre-industrial times, environmental variables explained only a small proportion of β-diversity since dominant taxa were present across the range of environmental gradients, whereas habitat specialists were very rare. Between pre-industrial and modern times, our analysis highlights an increase in β-diversity and the proportion of β-diversity that was explained by environmental variables. Increased disturbance rates have favored early-successional habitat specialist taxa and reduced the habitat breadth of pre-industrial generalists, thereby increasing the strength of community-environment relationships.Conclusions
Our results support that disturbance can alter the strength of community-environment relationships and also suggest that functional traits of species within the regional pool could predict whether or not disturbance alters such relationships.752.

The population and range of feral pigs in the United States are rapidly expanding, yet key knowledge gaps exist regarding their role in the ecology and transmission of foodborne pathogens. Our objectives were to estimate the prevalence of Campylobacter jejuni and Campylobacter coli shedding among feral pigs throughout Texas and to identify risk factors for positive status. Faecal samples were collected from feral pigs in Texas from February 2014 through May 2015, and target organisms were detected using PCR assays. The prevalence of C. jejuni shedding was 1.6% (6/370), and the prevalence of C. coli shedding was 3.5% (13/370). C. coli shedding was significantly more common (p = .008) among female pigs than among male pigs. Feral pigs may represent a source of human campylobacteriosis. 相似文献

Clostridium difficile is a common nosocomial pathogen in humans and animals that causes diarrhea and colitis following antibiotic therapy. Isolates of C. difficile obtained from faecal material from 20 human patients and 6 equine subjects with antibiotic-associated diarrhea were investigated regarding production of toxins A and B, their capacity to adhere to the human intestinal Caco-2 cell line and equine intestinal cells, and for the presence of fimbriae. The results showed that most (17/20) of the human clinical isolates produced both toxins A and B. One of the human isolates proved toxin A-negative/toxin B-positive. All (6/6) horse isolates were positive for both toxins A and B. Both the human and horse isolates possessed the capacity to adhere, to varying degree, to human and equine intestinal cells. It appeared that human isolates produced greater amounts of toxin B, and that there was a host-species dependency on ability to attach to intestinal epithelial cells. We conducted an immunological assay of blood samples taken from 85 swine‐specialist veterinarians attending the Congress of the Mexican Association of Swine Specialist Veterinarians in Mexico in 2011. Serum samples were assayed for Porcine rubulavirus (PorPV), Encephalomyocarditis virus (EMCV) and Leptospira spp. antibodies. Using a hemagglutination inhibition test, we registered 2.3% and 27% seropositivity for PorPV and EMCV, respectively. Using viral neutralization tests, we registered 5.8% and 47% seropositivity for PorPV and EMCV, respectively. For Leptospira spp., we registered a seropositivity of 38.8%. The variables (sex, age, years of exposure, number of visited farms, biosecurity level and region) showed no significant effect (P > 0.05) on the seropositivity for EMCV, PorPV and Leptospira spp. except for number of visited farms on HI seropositivity for EMCV (P < 0.05; odds ratio: 1.38). The data obtained provide information on the epidemiology of emerging diseases with zoonotic potential in occupational risk groups. 相似文献

Flavobacterium columnare is a ubiquitous Gram-negative bacterium that causes columnaris disease in a wide variety of fish worldwide. Timely detection of this bacterium is important to prevent its spreading and to reduce the economic loss to fish farmers. We developed a TaqMan-based real-time polymerase chain reaction (PCR) targeting a 113 bp nucleotide region of the chondroitin AC lyase gene of F. columnare G4. Specificity of the assay evaluated with 20 isolates of F. columnare and 15 other taxonomically or ecologically related bacteria revealed that the primers and probe were 100% specific for detection of F. columnare. The sensitivity limit of detection of F. columnare in pure cultures, over a range of dilutions [3.1 × 100–3.1 × 106 colony-forming units (CFU) mL−1], was observed to be ∼3 bacterial cells. The lowest limit of detection in nucleic acids from pure culture of F. columnare was 5.4 fg and the assay was linear with the log of amount of nucleic acid (R2=0.994) over that range (5.4 ng–5.4 fg). In tissues (blood, gills and kidney) of F. columnare experimentally infected fish, the bacterial numbers measured by TaqMan real-time PCR ranged from 3.4 × 100 to 9.5 × 105 CFU mL−1. In both F. columnare experimentally infected and spiked samples, positive PCR results were confirmed by bacteriological culture with 100% agreement. The TaqMan real-time PCR developed in this study is specific, sensitive and reproducible for the detection and quantitation of F. columnare in infected fish. 相似文献

A variety of tools are available to quantify uncertainty in age‐structured fish stock assessments and in management forecasts. These tools are based on particular choices for the underlying population dynamics model, the aspects of the assessment considered uncertain, and the approach for assessing uncertainty (Bayes, frequentist or likelihood). The current state of the art is advancing rapidly as a consequence of the availability of increased computational power, but there remains little consistency in the choices made for assessments and forecasts. This can be explained by several factors including the specifics of the species under consideration, the purpose for which the analysis is conducted and the institutional framework within which the methods are developed and used, including the availability and customary usage of software tools. Little testing of either the methods or their assumptions has yet been done. Thus, it is not possible to argue either that the methods perform well or perform poorly or that any particular conditioning choices are more appropriate in general terms than others. Despite much recent progress, fisheries science has yet to identify a means for identifying appropriate conditioning choices such that the probability distributions which are calculated for management purposes do adequately represent the probabilities of eventual real outcomes. Therefore, we conclude that increased focus should be placed on testing and carefully examining the choices made when conducting these analyses, and that more attention must be given to examining the sensitivity to alternative assumptions and model structures. Provision of advice concerning uncertainty in stock assessments should include consideration of such sensitivities, and should use model‐averaging methods, decision tables or management procedure simulations in cases where advice is strongly sensitive to model assumptions. 相似文献

- Extreme climate changes during the Cenozoic Era strengthened different biogeographical barriers that decreased the connectivity among populations, triggering lineage diversification of different species worldwide.
- The mitochondrial DNA control region was employed to explore the phylogeography of Sphyrna zygaena, a globally distributed species threatened by unsustainable, illegal, unreported and unregulated fisheries triggered by the international shark fin trade. It is listed as ‘Vulnerable’ by the IUCN Red List and its trade is regulated by CITES Appendix II.
- Only 13 haplotypes were found with low genetic diversity levels (hd = 0.686 ± 0.014; π = 0.00206 ± 0.00004) compared with other species of the Sphyrnidae family. The species has a very strong phylogeographic population structure among the Atlantic, Indian and Pacific Oceans (ΦST = 0.79132). Worldwide, there are six distinct populations with some haplotype sharing.
- These populations are probably connected by a stepping-stone dispersal of a small number of migrants per generation from the Indo-Pacific towards the Atlantic. Modelling suggests that S. zygaena diverged into two lineages around 6.96 million years ago which have been isolated in glacial refuges in the Atlantic and Indo-Pacific oceans; and after deglaciation, a population expansion probably permitted secondary contact.
- Conservation plans to establish differentiated management units should be adopted in each of the identified populations. Among these, the Eastern Central Atlantic and West Indo-Pacific are the most important areas for the species considering the historical migration routes that act as a bridge connecting the Atlantic and Indo-Pacific Oceans while the Gulf of Guinea connects the Atlantic populations. Still, further studies are required to know if these populations are also linked with nursery areas for the species.
- The results herein can help to delimit the main evolutionarily significant units to implement effective policies to establish differentiated management units as starting points to genetic monitoring programmes for Sphyrna zygaena.

Three newly formed Port wine pigments were isolated by Toyopearl HW-40(s) gel chromatography and semipreparative HPLC. Furthermore, the pigments were identified by mass spectrometry (LC/MS) and NMR techniques (1D and 2D). These anthocyanin-derived pigments showed UV-visible spectra different from those of the original grape anthocyanins. These pigments correspond to malvidin 3-glucoside linked through a vinyl bond to either (+)-catechin, (-)-epicatechin, or procyanidin dimer B3 [(+)-catechin-(+)-catechin].
