The objective of this study was to determine the efficiency of a portable total mercury analyzer (OhioLumex RA-915+) in comparison with traditional analytical methods, such as inductively coupled plasma atomic emission spectrometry and cold vapor atomic absorption. The quick mercury analytical procedure with the direct mercury analyzer without sample pretreatment (such as sample digestion) was optimized for a variety of environmental samples, including contaminated soil and plant samples. The efficiency was evaluated using practical parameters, such as time required for analysis, sample amount, mercury species, accuracy, and precision/reproducibility, as well as using statistical analysis. Our results demonstrate that these three instrumental methods yielded similar mercury concentration values and statistical data, while the mercury direct analyzer had the advantages of not requiring for sample digestion and only requiring a small quantity of samples for distribution of mercury in a single root, a single root hair, and sub-regions of a single leaf of plants. These factors are used to justify use of the portable direct mercury analyzer under field conditions and validation of the results. 相似文献

【目的】介绍ANUSPLIN在黄土高原多沙粗沙区时间序列气象要素的空间插值过程,为相关人员在ANUSPLIN的参数设置、误差分析和协变量要素选择等方面提供参考。【方法】以薄盘样条函数为插值理论,以专用气象数据插值软件ANUSPLIN为实现工具,并引入一个或多个协变量线性子模型,来实现多个气象要素的空间插值。【结果】完成了黄土高原多沙粗沙区时间序列(1980~2000年)多个气象要素月平均数据的栅格化,计算了气象要素随其影响因子变化的关系。【结论】ANUSPLIN以薄盘光滑样条函数为理论基础,引入协变量线性子模型,能较好地提高气象要素空间插值精度,且能反映气象要素随其影响因子变化的比率关系。在大多数情况下,该区模型选择以样条次数为3次的局部薄盘光滑样条函数模型为最佳。温度的空间插值相对比较容易,且误差较小,1995-07平均相对误差为1%;风速、水汽压的误差中等;日照时数和降雨量的误差较大,个别情况相对误差可超过50%。 相似文献

根据植物葡萄糖-6-磷酸脱氢酶(G6PDH,EC1.1.1.49)氨基酸保守区域,设计简并引物,采用RT-PCR技术克隆得到甜瓜属野生种Cucumis hystrix Chakr.(2n=24)G6PDH基因cDNA片段;随后基于该序列设计特异引物,PCR方法筛选野生种BAC文库,获得2个BAC阳性单克隆。测序后获得了G6PDH基因全序列及其上游启动子序列,GenBank登录号:JQ771576。序列分析显示:G6PDH基因全长约6.5 kb,由15个外显子和14个内含子组成;内含子序列均符合5’-gt-ag-3’结构。外显子拼接后获得了G6PDH基因的开放阅读框(ORF)序列,序列全长1 551 bp,编码516个氨基酸,与黄瓜基因组网站公布的G6PDH基因核苷酸序列和氨基酸序列同源性分别为98.71%和99.03%。野生种G6PDH氨基酸序列N端缺少转运肽序列,确定为野生种胞质G6PDH。氨基酸序列与烟草、马铃薯、荷兰芹、猕猴桃、拟南芥、大豆、小麦、玉米、葡萄、杨树等植物胞质G6PDH同源性高达77%以上。系统发育树分析发现,甜瓜属胞质G6PDH与茄科植物最先聚类,植物胞质G6PDH在分子进化水平上与物种进化相符。启动子结构分析表明:序列含有启动子基本元件TATA-box、CAAT-box,还含有丰富的光响应元件,与环境胁迫响应相关的不同顺式作用元件,促进高水平转录的5UTR Py-rich stretch元件和提高表达的CAT-box元件等。 相似文献

This study assessed the foliar uptake of 15N-labelled nitrogen (N) originating from wet deposition along with leaf surface conditions, measured by wettability and water storage capacity. Foliar 15N uptake was measured on saplings of silver birch, European beech, pedunculate oak and Scots pine and the effect of nitrogen form (NH 4 + or NO 3 ? ), NH 4 + to NO 3 ? ratio and leaf phenology on this N uptake was assessed. Next to this, leaf wettability and water storage capacity were determined for each tree species and phenological stage, and the relationship with 15NH 4 + and 15NO 3 ? uptake was examined. Uptake rates were on average five times higher (p?<?0.05) for NH 4 + than for NO 3 ? and four times higher for deciduous species than for Scots pine. Developing leaves showed lower uptake than fully developed and senescent leaves, but this effect was tree species dependent. The applied NH 4 + to NO 3 ? ratio did only affect the amount of N uptake by senescent leaves. The negative correlation between measured leaf contact angles and foliar N uptake demonstrates that the observed effects of tree species and phenological stage are related to differences in leaf wettability and not to water storage capacity. 相似文献

Microbial activity is affected by changes in the availability of soil moisture. We examined the relationship between microbial activity and water potential in a silt loam soil during four successive drying and rewetting cycles. Microbial activity was inferred from the rate of CO2 accumulating in a sealed flask containing the soil sample and the CO2 respired was measured using gas chromatography. Thermocouple hygrometry was used to monitor the water potential by burying a thermocouple in the soil sample in the flask. Initial treatment by drying on pressure plates brought samples of the test soil to six different water potentials in the range -0.005 to -1.5MPa. Water potential and soil respiration were simultaneously measured while these six soil samples slowly dried by evaporation and were remoistened four times. The results were consistent with a log-linear relationship between water potential and microbial activity as long as activity was not limited by substrate availability. This relationship appeared to hold for the range of water potentials from ?0.01 to ?8.5 MPa. Even at ?0.01 MPa (wet soil) a decrease in water potential from ?0.01 to ?0.02 MPa caused a 10% decrease in microbial activity. Rewetting the soil caused a large and rapid increase in the respiration rate. There was up to a 40-fold increase in microbial activity for a short period when the change in water potential following rewetting was greater than 5 MPa. Differences in microbial activity between the wetter and drier soil treatments following rewetting to the original water potentials are discussed in terms of the availability of energy substrate. 相似文献

The Schelde estuary is heavily polluted with many different (micro)pollutants. This results in high concentrations in the abiotic and biotic compartments of the Schelde estuary and in various effects. the present day suboxic and anoxic conditions in the upper estuary are probably responsible for the high distribution coefficients for some trace metals in the lower Schelde estuary (Western Schelde). a future reduction in discharges with a high biological oxygen demand will therefore not only result in higher oxygen concentrations, but probably also in lower distribution coefficients for trace metals in the Western Schelde. Simulations show that lower trace metal distribution coefficients will probably result in higher concentrations of dissolved (bioavailable) trace metals, even with substantially reduced discharge rates, due to the desorption of trace metals from resuspended bottom sediments. 相似文献

The metabolism and fate of ethametsulfuron-methyl ?methyl 2-[[[[[4-ethoxy-6-(methylamino)-1,3, 5-triazin-2-yl]amino]carbonyl]amino]sulfonyl]benzoate? in rutabaga were investigated. After 72 h, absorption and translocation of [(14)C]ethametsulfuron-methyl in rutabaga did not change for the duration of the study (50 days). Less than 4% of recovered radioactivity was present in the rutabaga root. Ethametsulfuron-methyl was metabolized through a proposed unstable alpha-hydroxy ethoxy intermediate that dissipated 3 days after treatment to two major metabolites, O-desethylethametsulfuron-methyl and N-desmethyl-O-desethylethametsulfuron-methyl, as determined by liquid chromatography-mass spectrometry. It was estimated that at a spray dose of 30 g of active ingredient ha(-)(1) and a harvest weight of 0.5 kg, the edible portion of the rutabaga root would contain no ethametsulfuron-methyl and approximately 1.3 ppb total of both identified metabolites. Residue analysis and toxicological assessment show that ethametsulfuron-methyl and its metabolites should pose little or no risk to consumers of rutabagas. 相似文献

Until now, most tillage erosion experiments were conducted under controlled soil and operating conditions. However, soil condition, tillage depth, speed and direction generally show substantial within-field variation. In this study, a series of tillage experiments were set up to investigate the erosivity of tillage under normal operating conditions. The effect of a typical tillage sequence, including multiple mouldboard, chisel and harrow passes, on soil translocation and tillage erosion was studied during a period of 3 years. Soil translocation in excess of 10 m was observed while the average net translocation rates ranged between 0 and 0.9 m. The results suggest that the annual tillage transport coefficient, associated with mechanized agriculture, is in the order of 781 kg m−1 yr−1. The experimental results also show that the tillage transport coefficient of a sequence of tillage operations can be reasonably well predicted from information provided by the farmer and by summing the transport coefficients obtained from controlled, single pass experiments. However, a Monte Carlo simulation showed that a relatively high number of tillage operations are required to obtain accurate estimates of the tillage transport coefficients in multiple pass experiments. 相似文献

The purpose of this review is to examine the current knowledge of the role of trees in providing shelter for pastures, crops, and livestock, for controlling erosion of soils and improving productivity and sustainability of agricultural production in Australia — and the extent to which this knowledge has been applied.Land degradation — tree loss and associated soil salinity, water and wind erosion, soil acidification, soil structural decline and nutrient degradation — is evidence that our primary production systems are not sustainable. We have sought increased production without proper consideration of the ecological context of that system. About half of Victoria's crop and pasture lands are affected or at risk, and in Western Australia about 25% of the cleared agricultural land is wind-eroded and 60% is potentially susceptible, salinity affects 0.43 m ha and half of the divertible surface water is affected by salinity. Similar problems occur in other States. At least 43 m ha or 13% of our rangelands are seriously degraded by wind erosion caused by overgrazing, often coinciding with drought or a run of drier years.Minimum tillage and stubble management for erosion control in cropping has been a major extension and research activity in Australian agriculture. Severe weather, combined with imperfect adoption of appropriate grazing and crop management systems, shows the weakness of complete reliance on these methods of erosion control. An effective system must accommodate the impact of extreme events, which are the most damaging. However, the complementary use of windbreaks to reduce soil erosion is rare, and their establishment has not been promoted, despite the wide-spread adoption of this technology by other countries.In the cropping and higher rainfall grazing areas, the systematic planting of 10% of the land in a net of shelterbelts/timberbelts/clusters could achieve a 50% windspeed reduction; this would substantially improve livestock and pasture production in the short and long-term. Wind erosion could be dramatically reduced and crop production probably increased by the use of windbreaks. Wheat and oat yield at Rutherglen (Victoria), and lupin yield at Esperance (Western Australia), were increased in the sheltered zone by 22% and 47%, and 30%, respectively.In semi-arid and dry temperate areas, planting of 5% of the land to shelter could reduce windspeed by 30–50% and soil loss by up to 80%. This planting would also contribute substantially to achieving other objectives of sustainable agriculture. Agroforestry — particularly timberbelts applications — will be important in the long-term strategy for achieving revegetation. If some of the trees yield a marketable product then the adoption of the system will be more readily achieved.In the arid (pastoral) areas there is an urgent need to promote the ethic that preservation and improvement of the perennial grass and shrub vegetation is critical for the protection of the soil and maintenance of land capability. Control of animal grazing remains the sole means of preventing erosion in much of this zone. While satellite imagery allows us to assess the condition of leasehold lands, we have failed to achieve stocking policies that will halt the degradation of our rangelands. 相似文献
