ABSTRACT Because there were no previous reports on the molecular characterization of Potato virus V (PVV, genus Potyvirus, family Potyviridae), the complete genomic sequence of PVV isolate Dv42 was determined. The length of the single-stranded messenger-polarity RNA genome was 9,851 nt (nucleotides), followed by a poly(A) tail. The genome contained a 5'-terminal nontranslated region (5'-NTR; 204 nt), a single open reading frame (nucleotides 205-9406; 3,067 amino acids), and a 3'-NTR that was unusually long (446 nt) compared with that of Potato virus Y (PVY; 331-nt 3'-NTR), Potato virus A (PVA; 207-nt 3'-NTR), and other potyviruses that naturally infect Solanaceae species. Phylogenetic analysis with the cylindrical inclusion protein-encoding and coat protein (CP)-encoding regions indicated that PVV Dv42 was most closely related to Pepper mottle virus and PVY, respectively. Seven PVV isolates (including Dv42) collected from cultivated potatoes in the Netherlands, the United Kingdom, and Norway from 1964 to 1997 were uniform in serological properties and symptomatology in indicator hosts that could distinguish strains of PVY and PVA. The nucleotide sequences of the 5'-NTR, P1, CP, and 3'-NTR regions of the PVV isolates were determined and were 94.6 to 99.5, 96.3 to 98.8, 96.4 to 98.7, and 96.3 to 99.6% identical, respectively. The amino acid similarities for the P1 and CP were 95.8 to 98.6 and 96.0 to 97.8%, respectively. Phylogenetic analysis of the CP sequences of PVV revealed no significant grouping, in contrast to PVY and PVA, which were grouped largely according to the previously recognized strains based on host responses. However, the relatively few differences in the P1 sequences of PVV were correlated with the different countries of origin. Hence, the PVV isolates infecting potatoes in Europe seem to vary little genetically and may belong to a single strain.  相似文献

Elevated phosphate concentrations at Safaga and Quseir, Red Sea, Egyptian coast, wererelated to some ecological and nutritional aspects of the macro-alga Cystoseira myrica during1985. The results obtained were compared with those recorded for a non-polluted control site atGhardaqa. Chemical analysis of surface seawater samples revealed that the concentrations of dissolvedphosphate at Safaga and Quseir were 3 and 20 times as much as that at Ghardaqa, respectively.The standing crop of C. myrica showed two peaks in spring and autumn. Maximum biomass yield(491 g m-2) and number of individuals (127 m-2) of C. myrica were recorded in the spring at theGhardaqa site. Quseir C. myrica samples contained significantly higher protein-N, total-N andtotal-P values but lower carbohydrate values than the Safaga and Ghardaqa samples (LSD at 1%level). Tissue N:P molar ratios for C. myrica, Laurencia papillosa and Ulva lactuca collected fromGhardaqa ranged from 68-98, compared to 30-59 for the same species collected at thephosphorus polluted sites at Safaga and Quseir. The carbon levels and the C:N ratios of C. myricaexhibited no significant variations in the three studied sites. However, the C:P ratios of Ghardaqaplants were significantly higher than those of Safaga and Quseir.  相似文献

Vascular plants have various inducible resistance mechanisms as defense against pathogens. Mosses, small nonvascular plants (subkingdom Bryophyta), have been little studied in regard to their pathogens or modes of defense. Data here show that Erwinia carotovora, a bacterial plant pathogen that causes softrot in many dicotyledonous plants, can also cause soft rot symptoms in the moss Physcomitrella patens. Infection of moss by E. carotovora required pathogenicity factors similar to those required to infect vascular plants and, again as in vascular plants, salicylic acid (SA) induced moss to inhibit tissue maceration by Erwinia. These data reveal that SA-dependent defense pathways may have evolved before differentiation of vascular and nonvascular plants.  相似文献

A neurological disorder in Merino sheep, characterised clinically by progressive posterior ataxia and microscopically by Wallerian degeneration in thoracic segments of the spinal cord, is described. Animals of both sexes were affected, with the earliest onset of disease being at 5 months of age. Most affected animals died before 2 years of age. The clinical, pathological and epidemiological features suggest that this degenerative thoracic myelopathy is a previously unrecognised entity differing from other reported causes of ataxia in sheep in Australia.  相似文献

ABSTRACT Sequences of the coat protein (CP) and 3'-end nontranslated region (3'NTR) of 13 isolates and the helper component proteinase (HC) of nine isolates of potato A potyvirus (PVA) were determined and compared with the eight previously determined PVA CP and 3'NTR sequences and one HC sequence. CP amino acid (aa), 3'NTR nucleotide, and HC aa sequence identities were 92.9, 93.4, and 94.8%, respectively. Sequence data, serological tests, and the necrotic local lesions induced in the leaves of the potato hybrid 'A6' confirmed that tamarillo mosaic virus is a strain of PVA. The aa substitutions A6T and G7S in the CP N-terminus were correlated with loss of aphid transmissibility. Development of necrotic lesions or nonnecrotic symptoms in the systemically infected leaves or lack of systemic spread in potato cv. King Edward were used to place the PVA isolates into four strain groups, but this grouping was not correlated with any differences in CP, HC, or 3'NTR. Recognition of CP by three monoclonal antibodies was used to place the PVA isolates into three groups different from the four groups above. The epitopes of two mono-clonal antibodies were mapped by site-directed mutagenesis to the same lysine residue at the CP aa 34.  相似文献

Plant-pathogenic Streptomyces species produce a variety of different phytotoxic 4-nitroindol-3-yl-containing 2,5-dioxopiperazines (thaxtomins) that induce scab symptoms on potato tubers (Solanum tuberosum). The possible mutual synergistic or antagonistic effects of thaxtomins are unknown. Modified methodology using column chromatography allowed the purification of thaxtomin A in large quantities (27 mg, HPLC purity of 97%). Thaxtomin A ortho isomer, thaxtomin B, and C-14 deoxythaxtomin B (thaxtomin D) were also purified. All four compounds induced similar symptoms of reduced root and shoot growth, root swelling (10-200 ppb), or necrosis (200-1000 ppb) on micropropagated in vitro cultures of potato. The scab-resistant potato cvs. Sabina and Nicola were more tolerant to thaxtomins than was the scab-susceptible cv. Matilda. Thaxtomins applied in combinations showed additive effects but no synergism, whereas thaxtomins A and B displayed antagonism with thaxtomin A ortho isomer.  相似文献

Resistance to potato viruses was examined in the F1 hybrids (TET) obtained from a cross between a diploid (2n = 24), tuber-bearing interspecific hybrid 87HW13.7 (Solanum tuberosum W231 ×S. multi-dissectum PI 473354) and a diploid (2n = 24), nontuber-bearing wild potato species (S. brevidens CPC 2451) using S. phureja IvP35 (2n = 24) for rescue pollination. The parental plants were susceptible to PVX, whereas two hybrids (TET38.2 and TET38.9) and S. phureja IvP35 reacted with hypersensitivity to PVX. Two hybrids (TET 38.9 and TET 38.12) were extremely resistant to PVY°, which was similar to S. brevidens and S. phureja IvP35, whereas the remaining two hybrids were moderately resistant to PVY°. No resistance to PVA and PLRV was observed in the progenies, in contrast to S. brevidens which was extremely resistant to PVA and PLRV. Hypersensitivity to PVX in two progenies suggested (1) integration by somatic translocation or heterofertilization and expression of genes from the rescue pollinator S. phureja IvP35, or (2) transgressive or complementary gene action.  相似文献
