Target region amplification polymorphism (TRAP) is a relatively new PCR-based technique that detects large numbers of loci in a single reaction without extensive pre-PCR processing of samples. The aim of this study was to integrate TRAP markers in an EST-derived SSR linkage map of a RIL mapping population from the cross of the durum wheat cultivars Ciccio and Svevo, for a more general purpose of establishing a high-throughput system for genetic map saturation. Primer combinations producing PCR products with at least 4–5 polymorphic bands were selected and analyzed across the mapping population. The PCR reactions produced a total of 2,881 fragments with an average of 52 peaks per reaction. A total of 142 new TRAP markers were mapped and found to be randomly distributed in the genome. The total length of the map was 2,043.0 cM, with an average chromosome length of 145.9 cM. Homoeologous group one had the highest number of TRAP markers (38 loci) and the longest map length (407.9 cM) for a total of 87 markers, while the homoeologous group five had the lowest TRAP marker number (5 loci) and the shortest map length (232.5 cM). The distribution of markers among the seven homoeologous groups was random. The results indicate that TRAP is highly efficient in genetic mapping, generating a large number of markers scattered across the genome. A partial genetic linkage map was constructed on 71 doubled-haploid lines derived from a cross between the barley lines Tadmor
and WI2291 with 181 molecular markers. The segregating population was used to detect markers linked to the gene Mlg conferring resistance to powdery mildew (Erysiphe graminis f. sp. hordei) and to genes for quantitative resistance to scald (Rhynchosporium secalis). The gene Mlg on chromosome 4H was flanked by two AFLP markers at a distance of 2.0 and 2.4 cM, respectively. QTLs for resistance to scald
were detected on chromosomes 2H and 3H. This association of molecular markers with qualitative and quantitative disease resistance
loci represents a valuable starting-point for marker-assisted selection.

The plastome sequence of the European cultivated potato, Solanum tuberosum subsp. tuberosum (tbr, GenBank accession no. DQ386163), was compared with that of S. bulbocastanum, a wild potato relative (blb, GenBank accession no. DQ347958), in order to characterize the degree and type of variability
in different genomic regions, and develop molecular markers relevant to genetics, breeding and biotechnology of potato. One
hundred forty-two and 251 PICs (Potentially Informative Characters) were found in coding and non-coding sequences (NCSs),
respectively. Further, while variation in coding regions was almost exclusively due to nucleotide substitutions, 25% of PICs
in NCSs of tbr and blb were due to indels, most of them mononucleotide or longer tandem repeats (micro and minisatellites).
Four intergenic regions were selected for further analyses in other 16 tuber-bearing Solanum species. The rps16-trnQUUGgene spacer was found to be the most variable, forty-six PICs in this region distinguishing 18 haplotypes. Analysis of haplotype
relationships, based on variability in the four intergenic regions, confirmed that the most primitive species from Central
America were the most distant to S. tuberosum. Finally, polymorphic sites in the same regions were used to develop a set of CAPS (Cleaved Amplified Polymorphic Sequences)

Hydrophilic phenols are the most abundant natural antioxidants of virgin olive oil (VOO), in which tocopherols and carotenes are also present. The prevalent classes of hydrophilic phenols found in VOO are phenyl alcohols, phenolic acids, secoiridoids such as the dialdehydic form of decarboxymethyl elenolic acid linked to (3,4-dihydroxyphenyl)ethanol or (p-hydroxypheny1)ethanol (3,4-DHPEA-EDA or p-HPEA-EDA) and an isomer of the oleuropein aglycon (3,4-DHPEA-EA), lignans such as (+)-1-acetoxypinoresinol and (+)-pinoresinol, and flavonoids. A new method for the analysis of VOO hydrophilic phenols by direct injection in high-performance liquid chromatography (HPLC) with the use of a fluorescence detector (FLD) has been proposed and compared with the traditional liquid-liquid extraction technique followed by the HPLC analysis utilizing a diode array detector (DAD) and a FLD. Results show that the most important classes of phenolic compounds occurring in VOO can be evaluated using HPLC direct injection. The efficiency of the new method, as compared to the liquid-liquid extraction, was higher to quantify phenyl alcohols, lignans, and 3,4-DHPEA-EA and lower for the evaluation of 3,4-DHPEA-EDA and p-HPEA-EDA. 相似文献
Mozzarella cheese obtained from buffalo (Bubalus bubalis) milk is a typical Italian product certificated by means of the European Protected Designation of Origin (PDO). Mozzarella cheese can also be obtained from bovine milk or bovine/buffalo milk mixtures, but in this case, it cannot be sold as PDO product, and its label must report the actual ingredients. However, bovine milk in PDO products was frequently detected in the past, suggesting fraudulent addition or accidental contamination. Several methods based on end-point polymerase chain reaction (PCR) have been profitably applied in a large number of tests to detect the presence of undeclared ingredients, also in dairy products. In the present study we report a real-time PCR method able to quantify bovine milk addition to pure buffalo cheese products. We validated a normalized procedure based on two targets: bovine mitochondrial cytochrome b (cyt b) to detect and quantify the bovine DNA and nuclear growth hormone (GH) gene used as a universal reference marker. With the use of this real-time PCR assay, 64 commercial mozzarella di bufala cheese samples purchased at local supermarkets, dairy shops, or directly from cheese manufacturers were analyzed. The results obtained demonstrate that most of the commercial samples were contaminated with bovine milk. Therefore, this assay could be conveniently employed to carry out routine and accurate controls aimed not only to discourage any fraudulent behavior but also to reduce risks for consumer health. 相似文献

The contents of triacylglycerols and diacylglycerols in three kinds of olive fruit oils (pulp, seed, and whole fruit) were determined. The fatty acid composition and the quality ratios 1,2-diacylglycerols/1,3-diacylglycerols and 1,2-diacylglycerols/total diacylglycerols were also assessed. Seven major Italian olive varieties were considered. Results of univariate statistical analyses indicated that the above analytical parameters (glyceridic ratios excepted) were effective in discriminating between pulp and seed oils. The seed oil fraction did not determine any change in the glyceridic indices and the acylglycerol or fatty acid composition concerning the whole fruit oil (mixture of pulp and seed oil fractions), the weight (%) of seed ( approximately 2%) being by far lower than the weight (%) of pulp ( approximately 85%) (fruit weight basis). Based on the data of triacylglycerol or fatty acid composition, and using appropriate parametric or nonparametric multivariate statistics, the genetic origins (olive variety) of the three fruit oil kinds were characterized. 相似文献
Summary Groups of 10 barley genotypes were selected for high grain yield under either high yielding (two groups) or low yielding conditions (two groups). The genotypes had a similar average grain yield across a wide range of yielding conditions, but differed in their linear response over environments (environmental sensitivity). The genotypes selected for high grain yield under low yielding conditions were less sensitive to changing environments than genotypes selected for high grain yield under high yielding conditions. The higher stability of genotypes selected under low yielding conditions was shown by both the linear regression analysis and the comparison of coefficients of variation. The use of a safety-first index showed that the probability of a crop failure of genotypes selected for high grain yield under high yielding conditions was between 1.8 and 2.7 times higher than for genotypes selected for high grain yield under low yielding conditions. The development of new cultivars for areas where a large proportion of the crop is grown by subsistence farmers should therefore be based on selection under low yielding conditions. 相似文献
The forest-based sector has been at the forefront in operationally implementing the sustainability concept, its associated
principles and indicators for sustainable forest management. Several methods have been developed to study environmental impacts
of forestry activities, but none of the existing tools address all the dimensions of sustainability along the whole forest
wood chain (FWC) in a balanced way. Consequently, the decision was made to develop a tool for sustainability impact assessment
(ToSIA), the modelling framework for sustainability impact assessment of FWCs. The objective of the EU Project Eforwood was
to develop ToSIA, a decision support tool. Within ToSIA, a FWC is modelled as a number of interconnected processes. For each
process, a range of economic, environmental and social indicators and their respective values are calculated, thus representing
the three pillars of sustainability. By this method, the multifunctionality of forests can be assessed and supply chains can
be compared with respect to sustainability. Sensitivity analysis and scenario techniques can be applied to learn about the
effect of expected changes to the structure of the chain, the material flows and the indicator values. In order to provide
the tool with information about forest and logging operations, data were collected at two fundamental levels: (1) a regional
level with case studies in Scandinavia, Iberia and Baden-Württemberg and (2) a European level with a case study that reflects
conditions in the 27 countries of the European Union. This paper describes and details the harvesting and logging processes
for the European countries. The results are displayed for each of the three regional case studies as well as aggregated to
five principal areas in Europe: Eastern, Northern, Western, Central and Southwest Europe. 相似文献
The goal of sustainable coffee production requires multiple functions from agroforestry systems. Many are difficult to quantify and data are lacking, hampering the choice of shade tree species and agronomic management. Process-based modelling may help quantify ecosystem services and disservices. We introduce and apply coffee agroforestry model CAF2021 (https://doi.org/10.5281/zenodo.5862195). The model allows for complex systems with up to three shade tree species. It simulates coffee yield, timber and fruit production by shade trees, soil loss in erosion, C-sequestration, N-fixation, -emission and -leaching. To calibrate the model, we used multivariate data from 32 different treatments applied in two long-term coffee agroforestry experiments in Costa Rica and Nicaragua. Without any further calibration, the model was then applied to agroforestry systems on 89 farms in Costa Rica and 79 in Guatemala where yields had been reported previously in farmer interviews. Despite wide variation in environmental and agronomic conditions, the model explained 36% of yield variation in Costa Rica but only 15% in Guatemala. Model analysis quantified trade-offs between yield and other ecosystem services as a function of fertilisation and shading.
