The invasive brown marmorated stink bug, Halyomorpha halys (Stål), has become a serious pest in mid-Atlantic apple orchards. Because no decision support tools exist for H. halys management, calendar-based insecticide applications have been the only successful technique for mitigating H. halys injury in orchards. Following the identification of the two-component aggregation pheromone of H. halys, we conducted a study to determine whether biological information generated by traps baited with this pheromone and pheromone synergist could be used to make management decisions. In this study, experimental apple orchards were treated with a H. halys-targeted insecticide only when a predetermined cumulative threshold of either 1, 10, or 20 adults per trap was reached. Once threshold was reached, two alternate row middle sprays 1 week apart were triggered and the threshold was reset. For comparison, some orchards were also subjected to a weekly alternate row middle spray or left untreated as controls. At harvest, significantly less fruit injury was observed when treated weekly or using a threshold of 1 or 10 H. halys adults per trap to trigger insecticide applications. Orchards treated using a cumulative threshold of 20 adults per trap or when left unsprayed had significantly higher fruit injury. In addition, insecticide applications were reduced by 40% using a threshold of 10 adults per trap. Our results suggest that baited pheromone traps can be used as decision support tools to trigger insecticide applications when needed to mitigate risk and effectively manage H. halys in apple orchards.  相似文献

Many populations are spatially structured with frequent extinction–colonization events. A clear understanding of these processes is necessary for making informed and effective management decisions. Due to the spatially and temporally dynamic nature of many systems, population connectivity and local extinction–colonization processes can be difficult to assess, but graph theoretic and occupancy modeling approaches are increasingly being utilized to answer such vital ecological questions. In our study, we used 6 years of egg mass counts from 34 ponds for Rana sylvatica to parameterize spatially explicit demographic network models. Our models revealed that the studied populations have spatial structure with strong source–sink dynamics. We also assessed the colonization and persistence probability of each pond using multi-season occupancy modeling. We observed extreme fluctuation in reproductive effort among years, resulting in variable levels of connectivity across the landscape. Pond colonization and persistence were most influenced by local population dynamics, but colonization was also affected by precipitation. Our demographic network model had moderate ability to predict reproductive effort, but accuracy was hindered by variation in annual precipitation. Source populations had higher colonization and persistence rates as well as a greater proportion of ravine habitat within 1,000 m than sink populations. By linking a spatially explicit connectivity model with a temporal occupancy/persistence model, we provide a framework for interpreting patterns of occupancy and dispersal that can serve as an initial guide for future habitat management and restoration.  相似文献

The sponge cake baking test is accepted and routinely used as a standard quality evaluation tool of soft white wheat for Asian markets, but its lengthy and laborious procedure makes it unsuitable for the routine evaluation of a large number of wheat breeding lines. We simplified the sponge cake baking procedure in the egg‐whipping step and improved its consistency by replacement of the hand mixing of cake batter with mechanical mixing, using a wire whisk or a BeaterBlade paddle. Egg whipping and mechanical batter mixing conditions were optimized by comparing foam density, sponge cake volume, and crumb grain to those obtained by the conventional procedure. Foam density, sponge cake volume, and crumb grain comparable to the conventional 100 g flour procedure were obtained with modifications, including extension of whipping time without heat input using a 5 L KitchenAid mixer, one‐time water addition at 3 min before the completion of egg whipping instead of twice, as in the conventional procedure, and cake batter mixing with a KitchenAid wire whisk or a BeaterBlade paddle. For baking a 50 g flour cake, egg foam of appropriate density was obtained with increased whipping speed and shortened egg‐whipping time (8 min). The modified sponge cake baking procedure yielded egg‐foam density, cake volume, and crumb grain similar to the conventional procedure and effectively differentiated soft wheat flours of different quality. Sponge cake volume of 14 soft white wheat flours ranged from 1,134 to 1,426 mL with the conventional procedure, from 1,113 to 1,333 mL with the modified procedure of batter mixing with a wire whisk, from 1,108 to 1,360 mL with the modified procedure of batter mixing with a BeaterBlade paddle, and from 577 to 719 mL with the modified method of 50 g of flour and batter mixing with a wire whisk. The modified methods with the BeaterBlade paddle and wire whisk exhibited significant correlation in cake volume with a conventional procedure (r = 0.931, P < 0.001 and r = 0.925, P < 0.001, respectively).  相似文献

Various methods have been developed to mitigate the adverse effects of the Federal Columbia River Power System on juvenile Pacific salmon out-migrating through the Columbia River basin. In this study, we found that hatchery-reared spring Chinook salmon Oncorhynchus tshawytscha in the river are in varying degrees of health, which may affect delayed mortality and the assessment of the effectiveness of management actions to recover listed stocks (e.g., barging fish downstream versus leaving fish in the river). A laboratory disease challenge with Listonella anguillarum was completed on fish from Rapid River Hatchery and Dworshak National Fish Hatchery (NFH) with different out-migration histories: (1) transported by barge, (2) removed from the river before barging, or (3) left to travel in-river. Barged fish from Rapid River Hatchery experienced less mortality than fish from Dworshak NFH. No statistical differences were found between the hatcheries with fish that had in-river out-migration histories. We suggest that the stressors and low survival associated with out-migration through the hydropower system eliminated any differences that could have been present. However, 18-25% of the fish that were barged or collected before barging died in the laboratory before the disease challenge, compared with less than 2% of those that traveled in-river. Owing to disproportionate prechallenge mortality, the disease-challenged populations may have been biased; thus, they were also considered together with the prechallenge mortalities. The synthesis of prechallenge and disease-challenged mortalities and health characteristics evaluated during out-migration indicated that the benefit of barging was not consistent between the hatcheries. This finding agrees with adult survival and delayed mortality estimates for the individual hatcheries determined from adult returns. The results suggest that the health status of fish and their history before entering the hydropower system (hatchery of origin and out-migration path) are critical variables affecting the conclusions drawn from studies that evaluate mitigation strategies.  相似文献

We have provided a detailed structural analysis of porcine alveolar macrophage TLR3 extracellular domain (ECD). The porcine TLR3-ECD contains 18 leucine-rich repeats (LRRs) consisting of blocks of consensus motifs and non-consensus motifs containing insertions. Excluding the N-terminal and C-terminal LRRs, porcine TLR3 has two LRRs with insertions, resulting in one LRR of 39 amino acids and another LRR of 34 amino acids. Furthermore, we have conducted the first examination of the regulated expression of porcine alveolar macrophage TLR3 during in vivo co-infection with influenza virus and Bordetella bronchiseptica. There was a bi-phasic upregulation of porcine TLR3 during influenza virus infection (day 1 and day 10 post-infection). Co-infection resulted in an enhanced expression of porcine TLR3 only at day 1 post-infection. Interestingly, B. bronchiseptica induced an upregulation in alveolar macrophage TLR3 expression at day 10 post-infection. Based on our work and that of others, TLR3 likely plays a key role in the immune response of lung cells to influenza virus infection in several mammalian species.  相似文献

A 5.25-year-old cynomolgus macaque (Macaca fascicularis) was found to have a marked leukocytosis due to a lymphocytosis on routine quarantine laboratory data prior to inclusion in a preclinical research study. The majority of lymphocytes were characterized as intermediate to large with round to convoluted nuclei, coarse to clumped chromatin, rare prominent nucleoli, and moderate amounts of lightly basophilic cytoplasm that frequently contained small magenta granules and/or clear vacuoles. The animal had tested negative for several viruses and other etiologic agents found in nonhuman primates 1 week prior to shipment to the research facility. However, further evaluation of the blood smear revealed rare hemoflagellates, and later testing using real-time PCR and ELISA was confirmatory for Trypanosoma cruzi (T cruzi). Trypanosoma cruzi is a zoonotic pathogen responsible for Chagas disease in people and can have negative consequences on study results when positive animals are inadvertently used for preclinical research. This case report describes a marked large granular lymphocytosis in an otherwise healthy macaque as the only indication of infection with T cruzi in an animal believed to be negative for the infection. Additionally, it highlights the diagnostic limitations of screening tests to rule out diseases in animals intended to be used in preclinical studies.  相似文献

The ability of a hand‐held chlorophyll meter (SPAD‐502 Chlorophyll Meter3, Minolta Camera Co., Ltd., Japan) to determine the N status of cotton (Gossypium hirsutum L.) was studied at field sites in Alabama and Missouri. Meter readings on the uppermost fully‐expanded leaf were compared to leaf‐blade N and petiole NO3‐N at first square, first bloom and midbloom as to their seed cotton yield predictive capability. Nitrogen was applied at rates of 0, 45, 90, 135, 180 and 225 kg ha‐1 to establish a range of cotton chlorophyll levels, tissue N concentrations, and seed cotton yields. A typical curvilinear cotton yield response to N fertilizer was observed in Alabama experiments.

Because of adverse weather conditions, cotton yield in Missouri experiments did not respond to N. Chlorophyll meter readings were significantly correlated to leaf‐blade N concentration at all three stages of growth for all experiments. In Alabama, chlorophyll meter readings compared favorably to leaf‐blade N and petiole NO3‐N with respect to their seed cotton yield predictive capability at all three stages of growth. It appears that hand‐held chlorophyll meters would be as reliable as leaf‐blade N and petiole NO3‐N for predicting supplemental N fertilization requirements of cotton. However, more research will be required prior to use of chlorophyll meter readings for routine cotton‐N recommendation purposes.  相似文献
