Recently, RccHan(TM):WIST (Wistar Hannover) rats were introduced to toxicity studies in Japan. The present study was performed to obtain control data for general toxicological parameters as an aid for interpretation of results in toxicity studies using this strain of rats. Four test groups comprising of 25 male and 25 female RccHan(TM):WIST rats were housed for 2, 4, 13 or 26 weeks from 6 weeks of age and observed and examined for clinical observation, body weight, food consumption, urinalysis, hematology, blood chemistry, organ weight, necropsy and/or histopathology. Ophthalmological examination was not conducted in this study, and the data in this report were obtained from an ongoing 104-week background study in RccHan(TM):WIST rats. These data were compared with the historical control data of CD(SD) (Sprague-Dawley) and/or F344 (Fischer) rats. The body weights of RccHan(TM):WIST rats were lower than those of CD(SD) rats and higher than those of F344 rats. The ophthalmological examination revealed a greater incidence of focal corneal opacity. Histopathology revealed focal mineralization of the cornea and Berlin blue-positive pigmentation in the epididymal interstitium as well as hepatocytes. Other than the above, some minor differences were found in urinalysis, hematology, blood chemistry and organ weights as compared with CD(SD) rats.  相似文献

In equilibrium environments where rainfall is relatively stable, grazing animal–vegetation dynamics are density-dependent; it is therefore appropriate to calculate carrying capacities and use them to define sustainable animal populations. In contrast, nonequilibrium environments are characterized by fluctuations in characteristics such as rainfall, resulting in fluctuations in plant biomass and in the corresponding carrying capacity. Herders adapt to such environments by moving opportunistically to pastures with better conditions. Studies since the 1990s have significantly improved our understanding of the continuity and integration of equilibrium and nonequilibrium systems. However, it remains unclear how and where such continuous, integrated rangeland systems result in qualitatively different land use patterns by local herders along a climatic gradient. Here, we developed a simple model that uses key environmental factors to predict a threshold representing the boundary between equilibrium and nonequilibrium land use systems, and we used an area of Mongolian rangeland as an example. We found a threshold in the proportion of usable pasture that corresponded to a specific range of rainfall values. Comparison of our results with previous ones supported our hypothesis about this threshold. The threshold behavior suggested that it is important to identify and monitor the boundary between equilibrium and nonequilibrium land use systems so that managers can respond to climatic change. National governments and aid agencies must understand the threshold process before they can identify focal areas where management regime change is required and propose appropriate policies that will support herders in the long term. Our study provides a simple, low-cost tool to evaluate ecosystems in this context.  相似文献

A total of 366 novel simple sequence repeat (SSR) markers were developed in Japanese chestnut (Castanea crenata), comprising 220 genomic SSRs derived from enriched genomic libraries and 146 expressed sequence tag (EST)–SSRs obtained from large-scale EST sequencing analysis. Thirty accessions, comprising Japanese, Chinese (C. mollissima), European (Castanea sativa), and American chestnuts (Castanea dentata), were used for evaluation of SSR polymorphism and transferability across species. The EST–SSRs showed less polymorphism than the genomic SSRs and were more transferable. The mean observed heterozygosity (HO) and the mean expected heterozygosity (HE) of genomic SSRs in the Japanese chestnuts were 0.63 and 0.68, respectively; those of EST-SSRs were each 0.47. Although about 80% of the genomic SSRs were amplified in all 4 species, more than 95% of the EST–SSRs were transferable across all 4 species. The many novel SSRs developed in this study will be applicable for the construction of genetic linkage maps, QTL analysis of phenotypic traits, high-throughput genotyping of marker-assisted selection, and association genetics.  相似文献

We reviewed studies dealing with regeneration under variable conditions in boreal and hemiboreal forests as affected by different microsite types by tree species functional groups. Generally, the importance of storm-induced microsites for regeneration dynamics in boreal forests depends on several factors: (1) distribution and type of microsites (generated by storm characteristics and stand conditions); (2) viable seed supply (stand history, species dispersal traits and status of surviving trees) and their species’ life history strategy; (3) climatic and site conditions (pre-storm conditions and storm-induced changes); and (4) delayed storm effects, such as retarded falling of trees, favoured vegetation growth, etc. Studies acknowledging the significance of microsites were mostly related to intermediate or severe events, causing sufficient changes in resource levels and growth conditions, and influencing extrinsic factors such as frost heaving, erosion and browsing. Also, the dispersal traits of available tree species, including sprouting and response of surviving trees, such as canopy expansion, should be considered in evaluating microsite importance in individual cases. In intermediate to severe windstorm events, pioneer species are generally profiting most from the additional offer in microsites, requiring bare mineral soil and elevated locations for their establishment and growth. Under gap dynamics, shade-tolerant species benefit from dead wood and elevated locations as these offer safe sites in stands with abundant understorey vegetation.  相似文献

In uneven-aged conifer–broadleaved mixed forests in Hokkaido, northern Japan, single-tree selection cutting has been a common management practice since the early twentieth century. This practice is expected to produce timber without major changes in stand structure or tree species composition. The demographic response of forests to this practice has often been unexpected, and degradation of stand properties has been widely observed. We propose here a sustainable management regime of selection cutting, based on an individual-based forest dynamics simulation model, SORTIE-ND. Our simulations, based on demographic data from 15 long-term monitoring stands, suggest that selection cutting using a lower cutting intensity together with a longer rotation period and reduced removal of small trees and conifer species is more appropriate than traditional systems in terms of maintaining stand structure and tree species composition, as well as being profitable financially. Supplemental regeneration practices, which can counter accidental mortality incurred during harvesting operations, would also be necessary to ensure tree recruitment.  相似文献

Radiation mutant 415-1, which is the first known diploid pollen-part self-compatible mutant of pears (Pyrus spp.), has a decreased ability to produce pollen. To determine whether the self-compatibility trait is associated with this defect, we directly analyzed the genotypes of individual pollen grains by using polymerase chain reaction amplification of DNA from single pollen grains. We isolated single pollen grains from 415-1 and succeeded in genotyping the S-RNase gene and three simple sequence repeat (SSR) markers in linkage group 17. Out of 173 individual pollen grains, 28 (16 %) were S-heteroallelic. These pollen grains had two alleles each of the S-RNase gene and of two linked SSR loci, all on a duplicated chromosomal segment, but only one allele of a non-duplicated locus farther away on the same chromosome. The segregation ratio of each marker in the pollen from 415-1 was approximately the same as that observed in outcross progeny. This suggests that the decrease in frequency of pollen with the duplicated S-haplotype occurred during meiosis or pollen formation, but that the probability of fertilization by S-heteroallelic pollen is equal to that of single-allelic pollen. However, the partial sterility in 415-1 can also be attributed to one or more unidentified lethal mutations unlinked to the duplicated segment encompassing the S-haplotype. Single-pollen genotyping can be used in a variety of applications in genetic research because in cases where all pollen genotypes are proportionately represented in the progeny, segregation ratios can be obtained without producing the next generation.  相似文献

Using an F1 population from a cross between Japanese pear (Pyrus pyrifolia Nakai) cultivars ‘Akiakari’ and ‘Taihaku’, we performed quantitative trait locus (QTL) analysis of seven fruit traits (harvest time, fruit skin color, flesh firmness, fruit weight, acid content, total soluble solids content, and preharvest fruit drop). The constructed simple sequence repeat-based genetic linkage map of ‘Akiakari’ consisted of 208 loci and spanned 799 cM; that of ‘Taihaku’ consisted of 275 loci and spanned 1039 cM. Out of significant QTLs, two QTLs for harvest time, one for fruit skin color, and one for flesh firmness were stably detected in two successive years. The QTLs for harvest time were located at the bottom of linkage group (LG) Tai3 (nearest marker: BGA35) and at the top of LG Tai15 (nearest markers: PPACS2 and MEST050), in good accordance with results of genome-wide association study. The PPACS2 gene, a member of the ACC synthase gene family, may control harvest time, preharvest fruit drop, and fruit storage potential. One major QTL associated with fruit skin color was identified at the top of LG 8. QTLs identified in this study would be useful for marker-assisted selection in Japanese pear breeding programs.  相似文献

Gametophytic self-incompatibility in Japanese pear (Pyrus pyrifolia Nakai) is controlled by the single, multi-allelic S-locus. Information about the S-genotypes is important for breeding and the selection of pollen donors for fruit production. Rapid and reliable S-genotype identification system is necessary for efficient breeding of new cultivars in Japanese pear. We designed S allele-specific PCR primer pairs for ten previously reported S-RNase alleles (S1S9 and Sk) as simple and reliable method. Specific nucleotide sequences were chosen to design the primers to amplify fragments of only the corresponding S alleles. The developed primer pairs were evaluated by using homozygous S-genotypes (S1/S1S9/S9 and S4sm/S4sm) and 14 major Japanese pear cultivars, and found that S allele-specific primer pairs can identify S-genotypes effectively. The S allele-specific primer pairs developed in this study will be useful for efficient S-genotyping and for marker-assisted selection in Japanese pear breeding programs.  相似文献

内蒙古半干旱地区沙质过牧草地的沙漠化过程   总被引:24,自引:17,他引:24  
为了掌握我国北方半干旱地区沙质放牧草地的沙漠化过程,在内蒙古东部的科尔沁沙地进行了为期5年的放牧试验。结果表明,在牧草利用率为70%的过牧条件下,草地植被盖度、高度、个体大小和根量急剧降低,第5年分别只有10.1%、1.0cm、2.5cm^3/株和18.7g/m^2(0-30cm土层内),是禁牧区的12.0%.3.02%.0.86%和22.1%。同时,家畜觅食活动增加,对草地践踏作用增强,如第5年其单位面积的蹄印6数和每只羊单位面积的蹄印数达到36.4n/m^2和8.5n/m^2,分别是轻牧区的9.6倍和8.25倍,土壤紧实度是禁牧区的3.75倍。植被盖度、高度、根量和植物个体普遍降低,家畜喜食植物首先从草地中退出后,草地开始出现小块裸斑,随着过牧的继续,这种裸斑迅速增加和扩大,使之相互连接成为大片裸地,如试验当年过牧区裸斑数仅为19个,而且73.7%≤2.5m的裸斑,到第3年裸斑数迅速增至358个,>2.5m的裸斑比例从26.3%增加到31.3%,裸地率由1.8%增加到35.8%,第5年裸斑平均长度由2.4m增加到4.1m,>2.6m的裸斑比例增至45.7%,裸地率达到了51.6%。当地面出现裸斑后,即使冬季不进行放牧,在风力的作用下草地中的裸地面积也会自然扩大,如第5年秋季过牧区的裸地率为51.6%,到第6年春季自然扩展到71.6%。裸地出现以后,冬春季在风力作用下地面迅速出现风蚀,过牧区第3年70%的测点发生风蚀,平均风蚀深度8.5cm,第5年90%的测点发生风蚀,平均风蚀深度25cm,最大风蚀深度90cm,地面呈现出凹凸不平的风沙地貌。研究结果表明,试验期间夏季放牧啃食对地表裸露的贡献率为41.3%,而冬春季节土壤风蚀对地表裸露的贡献率高达58.7%,这说明在内蒙古半干旱风沙区地表一旦裸露,马上就会因风蚀而迅速扩大。  相似文献

Wood is generally used as the interior material in museum storage rooms. Recently, however, the effects of volatile organic compounds (VOCs) from wood on artifacts has become a topic of great concern. The VOCs from four species of wood (western red cedar, spruce, kiri, and sugi) and their effects on artifact materials (two types of metal, seven types of pigment) were investigated using a deterioration-accelerating test, gas chromatography-mass spectrometry, X-ray diffraction analysis, and X-ray photoelectron spectroscopy. The results suggested that the influences on artifact materials varied greatly with wood species, and depended on specific components such as hinokitiol or acetic acid rather than the amount of total volatile organic compounds (TVOCs). It is a very serious problem that of the four species of wood, western red cedar (rich in hinokitiol), which has been recommended as an interior material for museum storage rooms, showed the heaviest deterioration on metal samples, and only this type of wood discolored enpaku (white lead) and rokushou (malachite, verdigris). In such storage rooms, museum artifacts should be carefully monitored. When selecting an interior material for a storage room or studying methods of preventing deterioration, it is very important to consider fully the characteristics of wood VOCs, not only the amount of TVOC.Part of this paper was presented at the 24th (Tokyo, June 2002) and 25th (Kyoto, June 2003) Annual Meeting of the Japan Society for the Conservation of Cultural Property  相似文献
