A 2 × 2 factorial experiment was conducted to investigate the effect of feeding a wheat‐based diet of two different hectolitre weights (66 vs. 74 kg/hl), achieved through different agronomical conditions, with or without the supplementation of a β‐glucanase and β‐xylanase enzyme mix on young pigs. The parameter categories which were assessed included growth performance, coefficient of apparent total tract digestibility (CATTD), faecal consistency, faecal microbial populations and faecal volatile fatty acid (VFA) concentrations. Sixty‐four pigs (11.6 kg SD 0.97) were assigned to one of four dietary treatments: (T1) low hectolitre weight wheat diet, (T2) low hectolitre weight wheat diet containing 0.1 g/kg β‐glucanase and β‐xylanase enzyme supplement, (T3) high hectolitre weight wheat diet and (T4) high hectolitre weight wheat diet containing 0.1 g/kg β‐glucanase and β‐xylanase enzyme supplement. The inclusion of wheat was 500 g/kg in the diet. The low hectolitre weight grain had a higher level of zearalenone, aflatoxin and ochratoxin contamination compared to the high hectolitre weight grain. The high hectolitre weight wheat had a higher gross energy (GE), crude protein (CP) and lysine contents compared to the low hectolitre weight wheat. Pigs offered the low hectolitre weight diet had a lower average daily gain (ADG) (p < 0.001), a lower gain to feed (G:F) ratio (p < 0.001) and a higher faecal score (more diarrhoea) (p < 0.001) compared to pigs offered the high hectolitre weight. The low hectolitre weight diet had a reduced CATTD (p < 0.05) of nitrogen (N) and gross energy (GE) compared with pigs offered the high hectolitre weight diet. In conclusion, the higher level of mycotoxins and lower content of GE, CP and lysine in the low‐quality wheat reduced ADG and the CATTD of nutrients in pigs offered this diet. The inclusion of a β‐glucanase and β‐xylanase enzyme mix had no effect on growth performance or nutrient digestibility.  相似文献

Ascorbic acid (AC) used as antioxidant in embryo culture is very sensitive and degrades unavoidably in aqueous solution. Methyl‐β‐cyclodextrin (CD) improved the stability of AC in solution to elevated temperature, light, humidity and oxidation. The aim of this study was to evaluate the effect of the complex AC‐CD during in vitro maturation (IVM) or in vitro culture (IVC) on oocyte developmental competence and subsequent embryo development and quality. AC‐CD (100 µM) was added to IVM media, and maturation level and embryo development were examined. Matured oocytes, their cumulus cells and produced blastocysts were snap‐frozen for gene expression analysis by RT‐qPCR. Besides, in vitro‐produced zygotes were cultured with 100 µM of AC‐CD and blastocysts were as well snap‐frozen for gene expression analysis. A group without AC‐CD (control?) and other with CD (control+) were included. No differences were found on maturation, cleavage or blastocyst rates. However, in matured oocytes, AC‐CD downregulated BAX, GPX1 and BMP15. In cumulus cells, AC‐CD downregulated BAX/BCL2 and GSTA4 while upregulated BCL2 and CYP51A1. The expression of SL2A1, FADS1, PNPLA and MTORC1 was downregulated in blastocysts derived from oocytes matured with AC‐CD, while in blastocysts derived from zygote cultured with AC‐CD, CYP51A1 and IGF2R were downregulated and PNPLA2 was upregulated. In conclusion, AC‐CD in both IVM and IVC media may reduce accumulated fat by increasing lipolysis and suppressing lipogenesis in blastocysts derived from both oocytes and zygotes cultured with AC‐CD, suggesting that CD improves the quality of embryos and bioavailability of AC during IVM and IVC.  相似文献

The breeding of South American camelids is the main economic activity of the high Andean region of South America and it, is potentially, the most profitable resource in of the Puna environmental conditions of the Puna. The duration of the gestation in alpaca is 339.7 ± 12 days. The objective of the present work was to macroscopically and microscopically describe the ontogenic development of the splanchnic cavities of the alpaca and to determine the gestational time in which the post‐cranial ossification centers are observed in the embryos/fetuses of this species, from day 21 to 107 of gestation. The documentation of normal ontogenic development, which is vacant for this period, is of the utmost importance to understand the consequences of the alterations at the different gestational times, as well as for the estimation of the gestational age in the case of abortions. Forty‐seven alpaca specimens of both sexes, at different times of their gestational development, collected during slaughter at local slaughterhouses of the Department of Huancavelica, Peru, were evaluated. Specimens were assigned to seven groups according to their morphological characteristics. The embryogenesis in the alpaca was characterized by a series of changes comparable to those occurring in other mammals with similar gestational periods. Despite these similarities, species differences were found in some organs as stomach, which are observed too in adult individuals.  相似文献

A technique was tested for obtaining water relations parameters from individual fascicles of loblolly pine needles by the pressure-volume curve method. Comparisons were made between parameters derived from (1) individual fascicles that were rehydrated in a pressure chamber after being removed from the shoot and (2) fascicles that were rehydrated on the shoot. Estimates of tissue osmotic potential for needles rehydrated by the two methods were significantly different for needles from terminal shoots, but not significantly different for needles from lateral shoots. Similarly, a significant difference in the estimated tissue elasticity at zero water potential was noted for needles on terminal shoots, but not for those on lateral shoots. It is suggested that differences due to rehydration method are related to the duration of the rehydration period and not the choice of technique. The use of fascicles of needles, rehydrated after detachment, allows repeated estimation of the tissue water relations of a single conifer shoot.  相似文献

The detection of gelatinase and beta-haemolysis activities was carried out in 83 faecal enterococci (43 Enterococcus faecalis, 33 E. faecium, five E. durans and two E. hirae) of poultry origin. In addition, the presence of genes of the gelE-fsrABC locus and of the cyl operon (cylL(L), cylL(S), cylA, cylB and cylM) were studied by polymerase chain reaction and correlated with gelatinase and beta-haemolysis production, respectively. Most of our E. faecalis isolates were gelatinase-positive (88%), being this activity not frequent in the other enterococcal species (2.5%). Only one of the 33 E. faecium isolates showed a positive gelatinase reaction. All enterococci that showed gelatinase activity harboured the gelE and fsrABC genes, although these genes were also detected in four E. faecalis and one E. durans gelatinase-negative isolates. Most of our non-E. faecalis gelatinase-negative isolates did not harbour gelE-fsrABC genes. A high proportion of faecal enterococci of poultry origin harboured genes of the cyl operon (71%), although only 7% contained the five cyl tested genes (all of them E. faecalis). Only one isolate of our series could express beta-haemolysis, harbouring the whole cyl operon. The cylL(S) genotype was the most prevalent in our enterococci (39%) and also the most prevalent among our E. faecalis isolates (60%). Other genotypes detected were the following ones (% of enterococci): cylA + cylB + cylM (13%), cylL(L) + cylA (4%), cylL(L) (4%), cylL(L) + cylA + cylB + cylM (2%), cylL(L) + cylA + cylM (1%) and cylA + cylM (1%). Both phenotypic and genotypic assays are important to evaluate the virulence potential of enterococci.  相似文献

The aims of this study were to determine the impact of body site, vigorous brushing and topical melatonin treatment on hair regrowth after clipping normal dogs. Siberian Husky dogs were randomly assigned to three groups of eight dogs each. All dogs had the lumbosacral region and both lateral thighs clipped. The left thigh and lumbosacral area received no treatment and were compared in all 24 dogs. Eight dogs had the right thigh treated with 0.1% melatonin twice daily for 2 months, and hair regrowth was compared with the left thigh. Eight dogs had the right thigh brushed twice daily for 2 months, and hair regrowth was compared with the left thigh. Eight dogs had neither thigh treated. Hairs were plucked before and 2 months postclipping, and the proportion of hair growth from the original length was calculated and compared as described above. Biopsy samples were collected before and after treatment to determine if brushing induced dermal inflammation and melatonin increased the proportion of anagen follicles. Proportionally, left thigh hairs were significantly longer compared to lumbosacral hairs 2 months postclipping. No significant differences in hair regrowth were noted between the nontreated thigh and the thigh treated with melatonin or brushed. No significant difference in dermal inflammation was noted before and after brushing. No significant differences were observed in the proportion of anagen follicles before and after topical melatonin treatment. Our results showed that the hairs in the lumbosacral region were proportionally shorter than lateral thigh hairs 2 months postclipping. Moreover, topical melatonin and brushing had no impact on hair regrowth after clipping normal dogs.  相似文献

Determination of site quality is a basic tool for proper selection of locations and species, in management of forest plantations.Throughout the Caribbean studies of site quality are few and are hampered by statistical limitations, inappropriate growth models, and limited data. We fitted growth curves for dominant height to evaluate and classify site quality of teak (Tectona grandis) plantations by using data from 44 permanent sample plots established since 1990 in 3 22 years old teak plantations in the Colombian Caribbean region. We used Korf’s and von Bertalanffy’s models to fit curves as nonlinear effects models. Both models, with a single random parameter, were considered as adequate for dominant height growth modelling, but Korf’s model was superior. There sulting curves were anamorphic and closely reflected high variability insite quality. Five site classes were clarified: at a base age of 12 years old,teak reached a mean dominant height of 24.8 m on the best sites, 9.8 m inthe worst sites, and in the averages sites, 15.8-18.8 m. Using this model,we identified the best and the worst sites for teak plantations in the Caribbean region. This model proved a useful tool, not only for site quality evaluation, but also for improved teak plantation planning and management.  相似文献

Seventy-six faecal samples were obtained from broilers at slaughterhouse level in Portugal. Samples were inoculated on cefotaxime-supplemented Levine agar plates. Cefotaxime-resistant Escherichia coli isolates were recovered from 32 samples (42.1%), obtaining a total of 34 E. coli isolates (one or two isolates per sample). Susceptibility to 16 antibiotics was studied by disk diffusion method, and 85% of the isolates presented a phenotype of multi-resistance that included antimicrobial agents of at least four different families. Extended-spectrum-beta-lactamases (ESBL) of the TEM and CTX-M groups were detected in 31 ESBL-positive E. coli isolates. Twenty-six isolates harboured the blaTEM-52 gene and two of them also harboured blaTEM-1b. The blaCTX-M-14 gene was identified in three isolates (in association with blaTEM-1b in one of them), and blaCTX-M-32 was demonstrated in two additional isolates. Three of the 34 cefotaxime-resistant isolates (9%) did not produce ESBLs, and two of them presented mutations at positions −42 (C → T), −18 (G → A), −1 (C → T), and +58(C → T) of the promoter/attenuator region of ampC gene. tet(A) and/or tet(B) genes were detected in all 34 tetracycline-resistant isolates, aadA in all 26 streptomycin-resistant isolates; cmlA in 3 of 6 chloramphenicol-resistant isolates, and aac(3)-II or aac(3)-I + aac(3)-IV genes in all 4 gentamicin-resistant isolates. Different combinations of sul1, sul2 and sul3 genes were demonstrated among the 22 trimethoprim–sulfamethoxazole-resistant isolates. Amino acid changes in GyrA and ParC proteins were identified in all 18 ciprofloxacin-resistant isolates. The results of this study indicate that the intestinal tract of healthy poultry is a reservoir of ESBL-positive E. coli isolates.  相似文献
