A longitudinal study was performed in a Swedish dairy herd where Neospora caninum had been isolated from a stillborn calf. Starting in autumn 1994, blood samples from all female animals in the herd were collected once yearly until 1999. The sera were analysed for presence of IgG1 antibodies to N. caninum by the iscom ELISA, and by an avidity ELISA to establish the timing of infection. In addition, data on reproductive performance were compiled. During the study the percentage of seropositive female animals increased from 63% to 87%. In 1994 a large number of young animals tested seropositive although their dams were seronegative, indicating that a transmission of the parasite other than the vertical had recently occurred. Low avidity values supported this assumption. The annual abortion rate increased from a mean of 2% before the initiation of the study to 9% in 1994-1998. During the same time, as judged by the avidity data, a large proportion of the animals shifted from being recently to being chronically infected. The source of the external infection in the herd could not be identified.  相似文献

The shortage of the n-3 fatty acids eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on the international markets has led to increasing substitution of fish oil by plant oils in Atlantic salmon (Salmo salar) feed and thereby reducing the EPA and DHA content in salmon. However, the minimum required levels of these fatty acids in fish diets for securing fish health are unknown. Fish were fed with 0, 1 or 2% EPA or DHA alone or in combination of both over a period, growing from 50 to 400 g. Primary head kidney leucocytes were isolated and stimulated with Toll-like receptor (TLR) ligands to determine if EPA and DHA deficiency can affect expression of important immune genes and eicosanoid production. Several genes related to viral immune response did not vary between groups. However, there was a tendency that the high-level EPA and DHA groups expressed lower levels of IL-1β in non-stimulated leucocytes. These leucocytes were also more responsive to the TLR ligands, inducing higher expression levels of IL-1β and Mx1 after stimulation. The levels of prostaglandin E2 and leukotriene B4 in serum and media from stimulated leucocytes were lower in both low and high EPA and DHA groups. In conclusion, leucocytes from low EPA and DHA groups seemed to be less responsive towards immunostimulants, like TLR ligands, indicating that low levels or absence of dietary EPA and DHA may have immunosuppressive effects.  相似文献

 In a cropping systems experiment in southeastern Norway, ecological (ECO), integrated (INT) and conventional (CON) forage (FORAGE) and arable (ARABLE) model farms were compared. After 5 experimental years, topsoil was sampled in spring from spring grain plots and incubated for 449 days at controlled temperature (15  °C) and moisture content (50% water-holding capacity). There were no detectable differences between model farms in terms of total soil C or N. For INT and CON, however, values of microbial biomass C and N, microbial quotient (Cmic/Corg), and C and N mineralization were, or tended to be, higher for FORAGE than for ARABLE. For the ECO treatment, values were similar for FORAGE and ARABLE and did not differ significantly from that of CON-FORAGE. For INT and CON, the metabolic quotient (qCO2) was lower for FORAGE than for ARABLE. Again, for the ECO treatment, values were similar for FORAGE and ARABLE and did not differ significantly from that of CON-FORAGE. We estimated the sizes of conceptual soil organic matter pools by fitting a decomposition model to biomass and mineralization data. This resulted in a 48% larger estimate for CON-FORAGE than for CON-ARABLE of physically protected biomass C. For physically protected organic C the difference was 42%. Moreover, the stability of soil aggregates against artificial rainfall was substantially greater for CON-FORAGE than for CON-ARABLE. On this basis, we hypothesized that the lower qCO2 values in the FORAGE soils were mainly caused by a smaller proportion of active biomass due to enclosure of microorganisms within aggregates. Altogether, our results indicated a poorer inherent soil fertility in ARABLE than in FORAGE rotations, but the difference was small or absent in the ECO system, probably owing to the use of animal and green manures and reduced tillage intensity in the ECO-ARABLE rotation. On the central portion of the west Florida continental shelf, radionuclide activities show unusually wide variations: radium-226 activities up to 350 disintegrations per minute per 100 liters, radon-222 activities up to 1300 disintegrations per minute per 100 liters, and deficiencies of radon-222 as low as -10 disintegrations per minute per 100 liters. Florida's phosphate-rich strata seerm to be the principal source of the radionuclides, with the transfer occurring directly from sediments or indirectly in streams, ground-water flow, and geothermal springs. Winter storm fronts may enhance radon degassing in the shelf waters.  相似文献

Sallow (Salix caprea L.) and rowan (Sorbus aucuparia L.) constitute small proportions of the deciduous tree volume in Scandinavia, but are highly preferred winter forage for moose and red deer, which occur at historically high densities. Thus, a possible decline of these tree species has been indicated. Against this background, we have reviewed the life histories of relevance for browsing, as well as the basic biology and genetics of sallow and rowan. The species show similarities with respect to short lifespan, small size and sympodial growth pattern, which are risk factors in a browsing context. They also have high juvenile growth rate, important for growing quickly out of reach of browsers. Sallow depends strongly on disturbance for establishment and is more demanding with respect to soil and light conditions than rowan, possibly important for the substantially lower abundance of sallow on the Norwegian Forest Inventory plots. Similarly, the relative recruitment of small size classes of sallow is less than for rowan. Although recruitment is reported to be hampered in wintering areas with high moose or red deer densities, the inventory data, however, dating only back to 1994, do not suggest a general decrease in any of the species. Sallow and rowan saplings show low mortality in moose and deer dominated areas and the species can be characterised as rather resilient to browsing. Of more concern is that browsing can constrain the development of mature rowan and sallow trees locally, with possible consequences for associated epiphytic biodiversity.  相似文献

Plasma kinetics and tissue sites of degradation for native and chemically modified low density lipoproteins have been investigated in (Oncorhynchus mykiss). Native and modified LDL labelled with125I-tyramine-cellobiose, (a residualizing adduct), were injected intravenously, and plasma and organ samples analyzed. Native LDL were cleared with a half life of about 30 hours, and mainly catabolized in the liver. Acetylation of LDL resulted in accelerated clearance (t1/2=2 h) and catabolism in the kidneys. Methylation of LDL had only minor effects on catabolism. The cellular localization of lipoprotein uptake was visualized in kidney by fluorescence microscopy. Native LDL were endocytozed by spheroid, parenchymal cells, supposedly steroid-producing cells. Acetylated, fluorescent LDL were found in vacuoles of flattened, sinusoidal lining endothelial cells. Our data show that catabolism of native low density lipoproteins in salmonids takes place mainly via hepatic receptors. A scavenger receptor pathway, for modified lipoproteins (mainly localized in the kidney) is also operative in trout.  相似文献

The experience of pain arises from both physiological and psychological factors, including one's beliefs and expectations. Thus, placebo treatments that have no intrinsic pharmacological effects may produce analgesia by altering expectations. However, controversy exists regarding whether placebos alter sensory pain transmission, pain affect, or simply produce compliance with the suggestions of investigators. In two functional magnetic resonance imaging (fMRI) experiments, we found that placebo analgesia was related to decreased brain activity in pain-sensitive brain regions, including the thalamus, insula, and anterior cingulate cortex, and was associated with increased activity during anticipation of pain in the prefrontal cortex, providing evidence that placebos alter the experience of pain.  相似文献

Wolffish (Anarhichos lupus) larvae were first fed in a low water-level raceway system, with two new types of formulated dry feed diets. Water level in the rearing tank was 1.5 cm. The fish showed a high preference for feeding from the surface. During the first week the fish experienced a drop in dry weights. The highest SGR of 5.8% was obtained between day 35–60. The best feeding regime gave a survival rate of 82 (2.5)%. Results indicate failure to initiate feeding as a main cause of mortality in the fed groups. Death by starvation occurred after 30–40 days. It is speculated whether the large difference in growth rates between the two feeding regimens is due to differences in carbohydrate levels between the two feed types. The rearing tank technology proved, with minor adjustments, to be self-cleaning and well suited for first feeding of wolffish.  相似文献

Liver is the main catabolic tissue for low density lipoprotein in rainbow trout (Gjøen and Berg 1992). We have investigated the interaction of LDL with isolated trout liver cells and liver membranes. 125I-TC labelled trout LDL bound to isolated trout liver cells in a time dependent and saturable manner with an apparant Kd of 20.1 g/ml, suggesting the existence of a specific binding site on the surface of these cells. The binding was Ca2+ dependent assessed by the 50% reduction obtained by 5 mM EDTA. Saturable binding to isolated trout liver membranes could also be demonstrated, but with lower affinity as compared to intact cells. Degradation of 125I-TC-LDL in hepatocytes was also saturable as degradation could be inhibited about 60% by a 100 fold surplus of unlabelled LDL. The rate of degradation increased with temperature up to 20°C. Both cell association (binding + uptake) and degradation were reduced down to 20% of control in the presence of microtubular and lysosomal inhibitors. Effects of different incubation temperatures (2, 8, 14 and 20°C) and hepatocyte membrane fatty acid composition on the rate of internalization and lysosomal degradation of the ligand, mannosylated albumin, that is taken up by receptor-mediated endocytosis, were investigated in rainbow trout (Oncorhynchus mykiss, Walbaum). The fish were kept at a water temperature ranging from 9 to 14°C and fed pelleted diets coated with either capelin oil (control), EPA/DHA-concentrate (rich in n-3 polyunsaturated fatty acids) or soybean oil (rich in n-6 unsaturated fatty acids) for at least 3 months prior to sampling. The endocytic uptake mediated by the mannose receptor was very efficient at all temperatures studied. Lysosomal degradation, on the other hand, came to a halt below 8°C. The activation energies for uptake and degradation were 54.6 and 164.2 kJ/mol respectively. No negative effects of increased amounts of either n-3 or n-6 fatty acids were observed on the endocytic parameters studied. On the contrary, multivariate analysis indicated a positive relationship between high levels of n-6 fatty acids and low unsaturation index in the phosphatidylcholine (PC) fraction of the hepatocytes and the internalization rate of 2°C, meaning that the rate of receptor-mediated endocytosis may be affected by membrane fatty acid composition.  相似文献
