The fixation of atmospheric nitrogen by organisms has usually' been measur'ed by the increase in t6tal nitrogen using the Kjeldahl method and, recently, by enriching with 15N isotope using a mass spectrometer after a suitable period of incubation. However, the Kjeldahl method is in accurate, laborious and not sensitive. The isotope UN method is 30 to 100 times more sensitive than the Kjeldahl method, but it is expensive and requires that the experiment is conducted only on a small scale, creating conditions quite different from the natural environment. The method also requires a mass spectrometer. More recently, DILWORTH (1) and SCHOLLHORN and BURRIS (2) observed that acetylene is reduced to ethylene by extracts from Clostridium pasteurianum. Ethylene production in the acetylene-reducing function of nitrogen-fixing microorganisms was determined by gas chromatographic techniques. The acetylene reduction method has been used in the detection of nitrogenase in biological systems (3,4,5,6). The method is reported to be simple, economical and a thousand times more sensitive than the liN method (6). This paper is a preliminary report on the use of the acetylene reduction method in studies on nitrogen fixation of various kinds of materials in the rice fields. 相似文献

The fate of added nitrogen in submerged soils was studied using 15N-labelled ammonium sulfate and alanine. After 8 weeks of incubation 25 and 22%, respectively, of nitrogen from ammonium sulfate and alanine were recovered in the soil. Under the experimental conditions used nitrogen added to presubmerged soils was lost rapidly outside of the soil-water system, regardless of whether the nitrogen was organic or inorganic. Fractionation studies revealed that the amount of tagged N incorporated into exchangeable ammonium, residual fractions, volatilized as NH3 and chemically fixed nitrogen was not enough to account for the nitrogen loss. The nitrogen loss was attributed to nitrification and subsequent denitrification during the incubation period. The effect of N-Serve [2-chloro-6-(trichloromethyl)pyrimidine] on nitrification of 15N-labelled (NH4)2SO4-in submerged soils was studied. About 15% more nitrogen was recovered from non-presubmerged soils, and less nitrate was accumulated in presubmerged soils where N-Serve coated (NH4)2SO4 was applied, than from soils where (NH4)2SO4 was applied without N-Serve. Presubmerged soils provided a more favorable environment for nitrification than for denitrification under the experimental conditions used. 相似文献

Soils of the northwestern part of the Taal Volcano in the Philippines representing four geomorphological units (upper, middle, and lower slopes and alluvial plains) were investigated and related to soil productivity. Results revealed that the soils on the upper and middle slopes contained higher amounts of organic matter and available P and displayed a low P retention together with more favorable physical properties such as loamy soil texture, loose and friable and well-drained soils compared to those on the lower slopes and in the alluvial plains. Due to these favorable soil characteristics, sustained agricultural production was higher at the upper elevations than at the lower elevations. Year-round multistorey / mixed cropping systems of cultivation in the upper and middle landscapes were also made possible because the higher precipitation was evenly distributed coupled with cooler temperatures compared the conditions on the lower slopes and in the alluvial plains. On the other hand, the soils on the lower slopes and in the alluvial plains had a clayey texture and contained a lower amount of organic matter and available P, in addition to the lower precipitation, resulting in reduced land utilization, as indicated by the limited types of crops grown and lower yield of crops. 相似文献

储藏条件是影响糙米储藏品质变化的关键因素,研究了糙米在5℃、20℃、30℃及自然室温下,分别以真空加脱氧剂、真空、CO2气体封入、自然空气封入、纸袋包装五种包装(气体)条件下,储藏6个月,糙米及其精米和米饭的有关理化学特性变化情况。结果表明,30℃储藏区米饭品质劣化严重, 储藏中糙米过干燥也可能影响米饭品质,真空与脱氧剂并用、真空、CO2气体封入、空气封入四种包装条件对糙米储藏品质的影响差异不明显,该研究结果对稻米的储藏有一定的参考价值。 相似文献

An indirect competitive enzyme immunoassay for hen egg white lysozyme (HEL) used as a food additive was investigated. Anti-HEL antibodies were obtained from B10A mouse ascites immunized by intraperitoneal injection of HEL. HEL samples to be assayed were extracted from foods with 1% gelatin in borate buffer. Goat anti-mouse IgG (H+L)-peroxidase complex was used as a second antibody, and 3,3',5,5'-tetramethylbenzidine was used as a substrate for the peroxidase. The working range for quantitative analysis was 1-50 ng/mL, because in this range the binding inhibition curve of anti-HEL antibodies to HEL-coated plates by HEL was linear. Even after losing the lysozyme activity by heat treatment, HEL could be detected by indirect competitive enzyme immunoassay. Recoveries of HEL by this assay were greater than 85% for Japanese noodles and Japanese traditional-style confectioneries, 53-95% for Miso and cooked beans, and 30-85% for fried fish pastes. HEL contents of 55 commercial foods were determined; HEL was detected in 19 samples in the range 25-20,000 ng/g. HEL as a food additive was detected more frequently in plant-derived foods than in foods of animal origin. 相似文献

Understanding how molecules can restructure the surfaces of heterogeneous catalysts under reaction conditions requires methods that can visualize atoms in real space and time. We applied a newly developed aberration-corrected environmental transmission electron microscopy to show that adsorbed carbon monoxide (CO) molecules caused the {100} facets of a gold nanoparticle to reconstruct during CO oxidation at room temperature. The CO molecules adsorbed at the on-top sites of gold atoms in the reconstructed surface, and the energetic favorability of this reconstructed structure was confirmed by ab initio calculations and image simulations. This atomic-scale visualizing method can be applied to help elucidate reaction mechanisms in heterogeneous catalysis. 相似文献

The objectives of this study are to propose a model for explaining the genotypic and environmental variation in above-ground biomass growth via photosynthesis and respiration processes from transplanting to heading for different rice genotypes grown under a wide range of environments, and to identify the physiological traits associated with genotypic difference in the biomass growth based on model analysis. Cross-locational experiments were conducted with nine different rice genotypes at eight locations in Asia covering a wide climate range under irrigated conditions with sufficient nitrogen application. The crop growth rate observed during the period from transplanting to heading ranged from 3.4 to 19.4 g m−2 d−1 among the genotypes grown at the eight locations. About one-third of the data sets were utilized for model calibration and the remaining sets were used for model validation. An above-ground biomass growth model was developed by integrating processes of single leaf photosynthesis as a function of stomatal conductance and leaf nitrogen content, growth and maintenance respiration and crop development. To rigorously examine the validity of this process model, measured data were input as external variables for leaf area index (LAI) development and leaf nitrogen content per unit leaf area. The model well explained the observed dynamics in above-ground biomass growth (R2 = 0.95*** for validation dataset) of nine rice genotypes grown under a variety of environments in Asia. The model simulation suggested that genotypic difference in the biomass growth was closely related to the difference in the stomatal conductance and leaf nitrogen content, as well as to LAI. This paper proposes the model structure, algorithms and all parameter values contained in the model, and discuss its effectiveness as a component of a more comprehensive model for simulating dynamics of biomass growth, LAI development and nitrogen uptake as a function of genotypic coefficients and environments. 相似文献

Astaxanthin is a naturally occurring red carotenoid pigment classified as a xanthophyll, found in microalgae and seafood such as salmon, trout, and shrimp. This review focuses on astaxanthin as a bioactive compound and outlines the evidence associated with its potential role in the prevention of atherosclerosis. Astaxanthin has a unique molecular structure that is responsible for its powerful antioxidant activities by quenching singlet oxygen and scavenging free radicals. Astaxanthin has been reported to inhibit low-density lipoprotein (LDL) oxidation and to increase high-density lipoprotein (HDL)-cholesterol and adiponectin levels in clinical studies. Accumulating evidence suggests that astaxanthin could exert preventive actions against atherosclerotic cardiovascular disease (CVD) via its potential to improve oxidative stress, inflammation, lipid metabolism, and glucose metabolism. In addition to identifying mechanisms of astaxanthin bioactivity by basic research, much more epidemiological and clinical evidence linking reduced CVD risk with dietary astaxanthin intake is needed. 相似文献
