Abstract

The choice of prospective type of farming requires knowledge about the specific relationships that exist between farm management practices and base environmental conditions. Nowadays the protection of soil organic carbon is one of the main tasks, because organic carbon in addition to soil fertility can act in elimination of soil contamination and carbon sequestration. Field experiments were focused on the effect of intensive farming without organic inputs versus grassland on organic carbon content. Organic carbon content (Cox) and humic substance fractions (C-humic acids and fulvic acid fractions), hot water extractable carbon and selected microbial characteristics in Eutric Cambisol were monitored during the period 1999–2010. A priming effect of soil cultivation was detected immediately after tillage. Arable soil with ‘intensive’ crop sequences (exclusively cash crops, cereals, oil plants) and with an optimal level of chemical inputs (mineral fertilizers, pesticides), but without organic farmyard manure had lower content of all carbon forms compared with grassland. 13C NMR spectroscopy and thermal analysis (TGA) were applied to characterize humic acid (HA) structure and stability. More carbon, less oxygen and more aromatic compounds were detected in grassland HA. Slight differences were found in HA thermo-oxidative stability and degradability, which was probably caused by changes in elemental composition and structure. Even the land use had no significant effect on basic microbiological characteristics (basal respiration, microbial biomass and qCO2); the physiology of the microbial community of grassland was altered by a higher ability to utilize L- and D-glutamic acid. The L/D ratio of glutamic acid mineralization indicated no occurrence of stress in soil for both types of farming. It has been demonstrated that although losses of carbon as a result of land-use conversions are generally more rapid, gains of carbon in grassland followed by changes in management practices can also occur.  相似文献

The relative efficiency of single-pass electrofishing of longer stream sections vs. double-pass electrofishing of shorter reaches was evaluated in small (<5 m wide) wadable lowland streams in the Lake Balaton basin, Hungary. Two hundred meters long stream sections at 8 sites were divided into ten 20 m long sampling units each. These units were used to estimate the representativeness of species richness, species composition and relative abundance data at each level of sampling effort (single vs. double-pass, and number of sampling units pooled) using rarefaction and similarity-based approaches. Assemblage variables showed strong response to the length of the stream sampled (number of sampling units pooled). However, no differences were found between the single- and double-pass methods at any level of sample size for any assemblage variable. Estimates of species richness and species occurrence distributions required more sampling effort than estimates of species relative abundances, using any evaluation method. If a proxy estimate of sample representativeness cannot be obtained in the field, a minimum sampling of single-pass electrofishing of 100 m long sections may be necessary even in small wadable streams with low level of habitat and assemblage diversity to get a relatively unbiased picture on assemblage characteristics.  相似文献

The aim of the presented work was to study the effect of pressing method, pasteurization, cultivar, and vintage on the content of (+)-catechin, (-)-epicatechin, and nine procyanidins in grape juice. The results showed that the concentration of these flavan-3-ols in the juice was influenced, in decreasing order of importance, by pressing method, cultivar, pasteurization, and vintage. Cold pressing without maceration was the least and hot pressing after maceration at 60 degrees C for 60 min the most effective method for extracting the flavan-3-ols. Pasteurization increased the concentration of catechins in cold-pressed juices, but it decreased concentrations in hot-pressed juices. The concentration of most procyanidins was increased by pasteurization. Among the white cultivars, Seyval and Niagara were highest in procyanidins and Elvira and Chardonnay were highest in catechins. Vincent, Foch, and Baco were the red cultivars highest in catechins, and Vincent also had the highest content of procyanidins.  相似文献

Peniciadametizine A (1); a new dithiodiketopiperazine derivative possessing a unique spiro[furan-2,7''-pyrazino[1,2-b][1,2]oxazine] skeleton, together with a highly oxygenated new analogue, peniciadametizine B (2); as well as two known compounds, brasiliamide A (3); and viridicatumtoxin (4), were isolated and identified from Penicillium adametzioides AS-53, a fungus obtained from an unidentified marine sponge. The unambiguous assignment of the relative and absolute configuration for the spiro center C-2 of compound 1 was solved by the combination of NMR and ECD measurements with Density-Functional Theory (DFT) conformational analysis and Time-Dependent Density-Functional Theory-Electronic Circular Dichroism (TDDFT-ECD) calculations. The spiro[furan-2,7''-pyrazino[1,2-b][1,2]oxazine] skeleton of 1 has not been reported yet among natural products and the biosynthetic pathway for 1 and 2 was discussed. Compounds 1 and 2 showed inhibitory activity against the pathogenic fungus Alternaria brassicae.  相似文献

Investigation of gap characteristics and tree regeneration patternsis central for our understanding of forest dynamics. By integratingaerial photograph analyses and ground surveys, we provide astudy of long-term canopy gap dynamics and tree regenerationpatterns in a Hungarian beech forest reserve. We found (1) thatin spite of the overall increase of gap area during the investigated30-years (from 2.5 to 7.7 per cent), total gap area and averagegap size (40–93 m2) were remarkably similar to those foundin other temperate and tropical forests, (2) if the fate ofindividual gaps was followed, two to three times more intensivecanopy dynamics (gap creation, closure and expansion) couldbe recognized than simple change of gap area indicated, and(3) average seedling density was considered to be sufficientfor natural regeneration. However, it was apparent that recentincreased deer browsing had prevented establishment of youngertrees of 1–2 m in height, as taller saplings were recordedonly in old gaps. Our results not only provide useful informationon forest dynamics but can also contribute to understandingthe potential roles that small forest reserves can play in providingessential reference data for nature-based forest managementof this forest type.  相似文献

Soil properties and agricultural practices take a joint effect on the communities of soil bacteria. The aim of the present study was to survey Alphaproteobacterial communities as possible indicators of soil quality considering clay, loamy and sandy soils under conventional and organic farming. Alphaproteobacteria community composition were analysed by 16S rRNA gene with nested-PCR (polymerase chain reaction) and denaturing gradient gelelectrophoresis (DGGE). Sequencing of partial 16S rRNA gene from the DGGE bands were performed. Conventional and organic farming resulted in significant differences in chemical properties of soils. According to the results community fingerprints were separated into groups depending on soil types and farming systems. This separation can be attributed mostly to soil pH, AL-P2O5,-K2O. The analysed sequences were identified as soil bacteria which could play the main role in nitrogen fixing, mineralisation and denitrification. The highest diversity index was revealed from the organic farming at sandy texture site, where mainly Mesorhizobium sp. and Rhizobium sp. were detected. The soil type and actual crop could have a stronger impact on the soil bacterial composition than the management.  相似文献

The main causative agents of Fusarium head blight in central Europe are Fusarium graminearum and F. culmorum. We examined the mycotoxin producing ability, aggressiveness and molecular variability of F. graminearum isolates. Altogether twenty-six Hungarian, three Austrian isolates and representatives of eight species identified in the F. graminearum species complex were involved in this study. Mycotoxin producing abilities of the isolates were tested by GC-MS and HPLC. The central European isolates were found to belong to chemotype I (producing deoxynivalenol). Most isolates produced more 15-acetyl-deoxynivalenol than 3-acetyl-deoxynivalenol suggesting that they belong to chemotype Ib. All F. graminearum isolates were found to be highly pathogenic in in vitro aggressiveness tests. Phylogenetic analysis of random amplified polymorphic DNA profiles, and restriction profiles of the intergenic spacer region of the ribosomal RNA gene cluster of the isolates allowed clustering of the central European isolates into 17 and 16 haplotypes, respectively. When RAPD and IGS-RFLP data were combined, almost every single central European F. graminearum isolate could be differentiated (27/29 haplotypes). Sequence analysis of a putative reductase gene of some isolates was also performed. Based on molecular data, the majority of the central European isolates belonged to F. graminearum sensu stricto characteristic to the northern hemisphere, with the exception of one Hungarian isolate, which was not related to any known species of the F. graminearum species complex based on sequence data. The taxonomic assignment of two other Hungarian isolates, previously suggested as belonging to F. boothii based on mitochondrial DNA restriction profiles, was supported by sequence analysis.  相似文献

Collaborative work was financed by the EU to develop and assess new diagnostic tools that can differentiate between bovine brucellosis and bovine infections due to Yersinia enterocolitica O:9 either in conjunction with, or as an alternative to, the classical serological, bacteriological or allergic skin tests. Sixteen heifers were experimentally infected with Brucella abortus biovar 1 (five heifers), Brucella suis biovar 2 (two heifers), Y. enterocolitica O:9 (six heifers) and Y. enterocolitica O:3 (three heifers). Four heifers, naturally infected with Y. enterocolitica O:9 that presented aspecific brucellosis serological reactions were also included in the experiment. A self-limited infection was induced in cattle by B. suis biovar 2. All the brucellosis serological tests used, i.e. the slow agglutination test (SAW), the Rose Bengal test (RB), the complement fixation test (CFT), indirect and competitive ELISA’s, lacked specificity when used to analyze sera from Y. enterocolitica O:9 infected animals. A Yersinia outer membrane proteins (YOPs)-ELISA was also used and although the test is able to detect a Yersinia group infection, it provided no evidence of whether or not there is a possible brucellosis infection when dual infections are present. The brucellergen IFN-γ test showed a lack of specificity also. The only test that was proven to be specific is the brucellergen skin test. All brucellosis serological tests, except the indirect ELISA, were limited in their ability to detect B. abortus persistently infected animals.

Based on these experimental studies, a strategy was implemented as part of the year 2001 Belgian Brucellosis Eradication Program to substantiate the eradication of bovine brucellosis. Epidemiological inquiries have identified risk factors associated with aspecific serological reactions, possible transmission and infection of cattle by B. suis biovar 2 from infected wild boars; and both legal and administrative measures taken by the veterinary services. No cases of bovine brucellosis have been confirmed in Belgium since March 2000.  相似文献

ObjectiveTo measure subarachnoid pressures, systemic circulatory and respiratory effects, and to calculate cerebral perfusion pressure during cisternal myelography.Study designProspective clinical study.AnimalsForty‐three client owned dogs with clinical signs of spinal disease, weighing 6–56 kg.MethodsDogs were premedicated with butorphanol and diazepam intravenously (IV) and anaesthesia was induced with propofol and maintained with isoflurane vaporized in oxygen. Ventilation was spontaneous. Heart and respiratory rates, invasive mean arterial blood pressure (MAP), end tidal carbon dioxide and isoflurane concentration were measured continuously. Initial subarachnoid pressure (SaP0) was measured in the cisterna magna with a needle pressure gauge. Iohexol 0.3 mL kg?1 was injected at a rate of 4.1 mL minute?1 into the cerebellomedullary cistern. The SaP was recorded during and at 120 seconds after contrast administration. The maximum SaP (SaPmax) and minimum calculated cerebral perfusion pressure (CPPmin) were recorded for each case.ResultsPrior to contrast injection, mean ± SD, MAP was 73 ± 20 mmHg and SaP0 was 10 ± 3 mmHg. The cerebral perfusion pressure (CPP) was 64 ± 20 mmHg. The contrast injection increased the SaP0 to 73 ± 33 mmHg (SaPmax). After injection, MAP increased to 97 ± 25 mmHg and the CPP decreased to 14 ± 34 mmHg. A negative correlation was found between the lowest CPP and body weight (ρ = ?0.77, p < 0.0001). Nine dogs had bradycardia, apnoea and hypertension, 21 dogs had at least one of these signs. The number of clinical signs showed significant correlation with body weight (ρ = ?0.68, p < 0.0001), SaPmax (ρ = ?0.66, p < 0.0001) and CPPmin (ρ = ?0.73, p < 0.0001).Conclusions and clinical relevanceCerebral perfusion can severely decrease during cisternal myelography using the standard dose of iohexol.
