ABSTRACT Differences in the penetration process by Rhynchosporium secalis were compared in resistant and susceptible barley cultivars at the seedling stage. Percent penetration and percent host cell wall alteration (HCWA) differed significantly among cultivars and isolates as revealed by light microscopy. Based on these two variables, the cultivars were statistically separated into two groups that corresponded to their disease reactions. The resistant cultivars, Johnston and CDC Guardian, showed 81.2 to 99.4% HCWA and 0.1 to 20.1% penetration at encounter sites, whereas the susceptible cultivars, Harrington, Argyle, and Manley, had 30.1 to 78.3% HCWA and 31.8 to 81.8% penetration. In the current study, cv. Leduc, which is susceptible at the seedling stage and resistant at the adult stage, showed the same percent HCWA and penetration as did susceptible cultivars. A significant negative correlation (P < 0.01) was found between percent penetration and percent HCWA for cultivars inoculated with two isolates of the pathogen. Isolate 1 was less virulent than isolate 2 with respect to percent penetration and induced significantly fewer HCWA. Scanning electron microscopy showed various shapes of fungal appressoria but no apparent difference in host reaction between resistant and susceptible cultivars. Transmission electron microscopy revealed interactions between the host and pathogen at various stages of penetration. The resistant cv. Johnston responded by producing appositions, as evidenced by a layer of compact osmiophilic material deposited on the inner side of the cell wall. Infection pegs produced by conidia were unable to penetrate the cuticle where an apposition had formed inside. When penetration occurred in the susceptible cv. Argyle, cytoplasmic aggregates and separation of the plasmalemma were visible from the host cell wall, but the layer of compact osmiophilic material was not always present. Data based on light microscopic observations suggested that HCWA may be one of the mechanisms responsible for resistance that is characterized as penetration prevention rather than as a slow rate of mycelial growth after successful penetration. HCWA occurred in response to attempted cuticle penetration, suggesting that HCWA may produce chemical barriers that help to prevent penetration.  相似文献

In a laboratory incubation study, the periodic changes in different chemical fractions of copper (Cu) in three sludge-amended soil types (acidic sandy clay loam, neutral clay loam and alkaline clay loam) of varying soil reaction were monitored under field capacity and flooding moisture regime over 16 months. The water soluble and exchangeable fraction of Cu was very low (≤1% of total Cu) in all three soil types. At the end of incubation (16 months), the sodium acetate extractable (carbonate sorbed)- and residual-Cu fractions transformed into Fe-Mn oxide fraction, irrespective of moisture regime in all three soil types. The resistance status of Rhipicephalus (Boophilus) microplus to "Diazinon" was evaluated in 20 locations situated at various agro-climatic regions of India. Adult immersion test (AIT) was optimized using laboratory reared acaricide susceptible IVRI-I strain of R. (B.) microplus and minimum effective concentration of Diazinon was determined as 635.2 ppm. The discriminating dose (DD) was worked out as 1270.4 ppm and was tested on female ticks collected from organized and unorganized farms located at different agro-climatic regions of India. On the basis of the data generated on three variables viz., mortality, egg masses and reproductive index, the resistance level was categorized as I, II, III and IV. The average resistance factor (RF) of 6.1 (level II) was recorded in the ticks collected from the northern sub-temperate trans-gangetic plains while high average RF values of 26.65 (level III) was recorded in the ticks collected from tropical middle-gangetic plains. The tropical middle gangetic plain has a very high density of animal populations where farmers use Diazinon for tick control, for agricultural practices and for mosquito control. Due to the continuous use of OP compounds the environmental load of Diazinon has become high in the area. DNA from tobacco leaf chloroplasts was isolated as a single component with a buoyant density in CsCl of 1.702 compared to 1.697 for nuclear DNA. 5-Methylcytosine is present in nuclear DNA but absent in chloroplast DNA. Chloroplast DNA, with a guanine-cytosine content of 43 percent, has a melting temperature of 86 degrees C and renatures completely on slow cooling, whereas nuclear DNA ( melting temperature, 84 degrees C; guanine-cytosine content, 40 percent) does not renature. About 9 percent of the total DNA in tobacco leaves is chloroplast DNA representing about 4.7 xX 10-(15) gram of DNA per chloroplast with a molecular weight of approximately 4 xX 10(7).  相似文献

ABSTRACT

Growth modelling is an important and effective tool for evaluating the effects of a particular management action on the future development of a forest ecosystem. However, such necessary growth models are not available for many indigenous tree species in India. Acacia nilotica is an important multipurpose tree species found in India and growth models are required for proper management of the species in the region. This paper presents equations for estimating potential stand density and predicting basal area in pure even-aged stands of A. nilotica in Gujarat State of India. Although no thinning was suggested, decrease in the number of trees in the stands was observed because of mortality due to overcrowding and some biotic factors. Relationships between quadratic mean diameter and stems per hectare were developed, which was used to establish the limiting density line. Eight different stand level models, belonging to the path invariant algebraic difference form of a non-linear growth function, were compared for projecting basal area. They can be used to predict future basal area as a function of stand variables like dominant height and stem number per hectare and are crucial for evaluating different silvicultural treatment options. The performance of the models was evaluated using different statistical criteria to recommend the suitable model for projecting the basal area in A. nilotica stands.  相似文献
