SUMMARY: We attempt to assess the impact of different levels of dietary β-carotene on immune function in rainbow trout. Semi-purified diets containing 0, 40, 200, and 400 mg β-carotene/kg dry diet were fed for 12 weeks to fish with average weight of 45 g. In addition to the humoral and cellular immune parameters, growth and feed utilization were examined. There were no marked differences in growth and feed utilization showing that β-carotene was not particularly efficient in enhancing growth of rainbow trout. Of the immune parameters measured, total immunoglobulin was significantly highest for the 200 mg β-carotene fed group. Serum complement activity (alternate pathway) at 200 and 400 mg β-carotene supplementation was significantly higher than that of the unsupplemented group. An increasing trend in lysozyme activity was observed, however, the differences among the groups were not significant. Phagocytic activity was similar among diet groups except at the highest level of supplementation where it was the maximum. Oxygen radical production by peripheral blood leukocytes appeared to be lower at higher levels of carotenoid supplementation. Overall, dietary β-carotene clearly enhanced immune response parameters in rainbow trout such as serum complement activity and total plasma immunoglobulin but did not show a definite influence for the other factors examined in the present study. 相似文献

The supplemental effects of cholyltaurine and soybean lecithin to a defatted soybean meal (SBM)-based fish meal-free diet
on the hepatic and intestinal morphology of rainbow trout Oncorhynchus mykiss were investigated. A fish meal-based diet (FM), three fish meal-free diets supplemented either with soybean lecithin (SL),
bovine gall powder (BG) or cholyltaurine (C-tau) and an unsupplemented fish meal-free diet (NFM), were given to fish (−11
g) for 10 weeks. The growth was the lowest in fish fed diet NFM, intermediate in fish fed diet SL and the highest in fish
fed diet FM, BG and C-tau. Hepatocytes of fish fed diet NFM were atrophied, and the distal intestine of these fish showed
abnormal features: disintegrated microvilli and fatty degeneration in epithelial cells and increased amount of connective
tissue in the submucosa. Hepatic and intestinal histological features in fish fed diets SL, BG and C-tau were similar to those
in fish fed diet FM. These results indicate that soybean lecithin, bovine bile salts and cholyltaurine have similar effects
on normalizing the hepatic and intestinal morphologies of rainbow trout fed the SBM-based diet, although growth promotion

To clarify the effects of temperature on the recruitment of chub mackerel (Scomber japonicus) in the North Pacific, we investigated the influence of winter surface temperature (WST) on spawners at the time of maturity around the spawning grounds and the influence of ambient spring temperature on larvae using estimated temperature (ET) obtained from particle tracking experiments. We found a significant positive correlation between ET approximately 10 days following hatching and the recruitment per spawning stock biomass (RPS) after 2000. The closer (more meandering) the Kuroshio Current (KC) axis was in relation to the spawning ground, the higher (lower) the spring surface temperature and the higher (lower) RPS was in the spawning ground. In contrast, WST inside KC near the maturity/spawning ground was significantly negatively correlated with RPS. A significant negative correlation between the temperatures in winter and spring was detected in the area after 2000, when the conditions of the Pacific decadal oscillation index and the stability of the Kuroshio Extension were synchronous, indicating that KC shifted northward during this time. The reversed temperature pattern was consistent with the winter–spring movement of KC axis in the offshore direction and was correlated with the winter–spring difference in the intensity of the Aleutian low. These results suggest that the annual variation in chub mackerel recruitment after 2000 was strongly affected by the combined effects of ambient temperature because of the reversal of conditions that occurred between winter and spring around the maturity/spawning ground, which was related to the KC path. 相似文献

ABSTRACT: Seasonal changes in the infection state of marine birnavirus (MABV) in Japanese flounder Paralichthys olivaceus and in rearing sea water are described. Sea water and 10–11 healthy fish were sampled monthly from April 2002 to February 2003. The MABV genome was detected throughout the year in > 80% of the fish examined at each sampling. The virus was isolated from the liver, kidney, and spleen, but not from the brain. The detection rate in each organ increased from April to October, and then decreased. Detection of virus antigen by the indirect fluorescent antibody technique also showed that the virus was present from spring to autumn (June–September) in the liver, kidney, and spleen, but not the brain. Sequence analysis of the MABV genome at the VP2–NS region revealed two specific mutations compared to the standard yellowtail strain (Y-6). It is suggested that the infection state of MABV in Japanese flounder changes to a latent or persistent infection after autumn. MABV was detected in sea water between September and February, suggesting that virus particles in the environment are relatively higher during cool seasons. 相似文献

ABSTRACT: The effect of salt concentration on the thermal denaturation profile of myosin in walleye pollack and carp myofibrils was compared by studying the subfragment-1 (S-1) and rod denaturation rates upon heating. Species-specific denaturation mode observed at 0.1 M KCl was no longer detected when samples were heated above 0.5 M KCl, where S-1 and rod denaturation rates were identical to each other. As the heating of the chymotryptic digest of myofibril formed practically no rod aggregates, S-1 denaturation in a form of myosin was the rate limiting step for rod aggregate formation. As the aggregate formation by rod was remarkably suppressed by lowering the temperature, the free movement of myosin tail upon heating was suggested to play an important role in the rod aggregate formation in a high salt medium. 相似文献

ABSTRACT: A pleuronectiform fish, the barfin flounder Verasper moseri , expresses three gonadotropin-releasing hormone (GnRH) forms in the brain: salmon GnRH (sGnRH), chicken GnRH-II (cGnRH-II) and seabream GnRH (sbGnRH). To clarify the effects of photoperiod on GnRH systems, changes in brain and pituitary GnRH peptide levels were examined using time-resolved fluoroimmunoassays. In experiment 1, 5-month-old male barfin flounder (mean total length 9.0 cm, body weight 11.0 g) were divided into short (8:16 h light : dark [L:D] cycle; lights on 08.00–16.00 hours) and long photoperiod (16:8 h L:D cycle; lights on 04.00–20.00 hours) groups in mid September and maintained until November under natural water temperature (19.3–15.2°C). Brain sGnRH concentrations were significantly higher in the 16:8 h L:D group than in the 8:16 h L:D group, whereas no significant differences were observed in total length, body weight, plasma testosterone concentration, brain cGnRH-II concentration and pituitary sbGnRH content. In experiment 2, 7-month-old male barfin flounder (mean total length 16.5 cm, body weight 76.8 g) were divided into short and long photoperiod groups in mid December and maintained until February under natural water temperature (12.5–6.6°C). Total length, body weight and condition factor were significantly greater in the 16:8 h L:D group than in the 8:16 h L:D group, whereas no significant differences were observed in plasma testosterone concentration and GnRH levels in the brain and pituitary. ABSTRACT: Sporophyll formation in two alariaceous plants, Undaria pinnatifida and Alaria crassifolia , was studied in relation to the nutrient requirements. The sporophylls of U. pinnatifida formed zoosporangia when they had N and P contents greater than 1.4 kgN/m3 and 0.74 kgP/m3 . This indicates that these values are the critical nutrient levels for zoospore formation in U. pinnatifida . In the sporophytes of A. crassifolia , many sporophylls with zoosporangia showed nutrient contents higher than 6.25 kgN/m3 and 1.70 kgP/m3 . These results suggest that the U. pinnatifida can form zoospores at lower levels of N and P contents than A. crassifolia . Both species often formed zoosporangial sori on the blades in the late period of each reproductive season. The fertile parts of the blade showed N and P contents higher than the critical levels. This phenomenon indicates that the blades have the capability to form zoosporangial sori if there is a sufficient accumulation of nutrients for zoospore formation. The zoospore formation on the blade seems to be accomplished by an overflow of excess nutrients from sporophylls into the blade, or by accumulating sufficient nutrients to form sori even if the sporophylls are not formed. 相似文献

The major digestive enzymes in Pacific bluefin tuna Thunnus orientalis larvae were characterized, and the physiological characteristics of the enzymes during early ontogeny were clarified using biochemical and molecular approaches. The maximum activity of trypsin (Try), chymotrypsin (Ct) and amylase (Amy) was observed at pH 6–11, 8–11 and 6–9, respectively. Maximum activity of Try, Ct and Amy occurred at 50 °C, that of lipase (Lip) was at 60 °C and that of pepsin (Pep) was at 40–50 °C. These pH and thermal profiles were similar to those for other fish species but differed from those previously reported for adult bluefin tuna. Enzyme activity for all enzymes assayed was found to decrease at high temperatures (Try, Ct, Amy and Pep: 50 °C; Lip: 40 °C), which is similar to findings for other fish species with one marked exception—increased Try activity was observed at 40 °C. Lip activity appeared to be dependent on bile salts under our assay conditions, resulting in a significant increase in activity in the presence of bile salts. Ontogenetic changes in pancreatic digestive enzymes showed similar gene expression patterns to those of other fish species, whereas marked temporal increases in enzyme activities were observed at 10–12 days post hatching (dph), coinciding with previously reported timing of the development of the pyloric caeca in bluefin tuna larvae. However, complete development of digestive function was indicated by the high pep gene expression from 19 dph, which contradicts the profile of Pep activity and previously reported development timing of the gastric gland. We evaluated the nutrient contents of rotifers sampled from larval-rearing tanks (tank rotifers) without water exchange during
the seed production of amberjack Seriola dumerili at three facilities (Kamiura, Kagoshima, and Miyazaki) and compared them with the nutrient contents of freshly enriched rotifers.
Compared to the enriched rotifers, the lipid contents, especially neutral lipids and proportion of 22:6n-3, tended to decrease
in the tank rotifers. These trends were clearer at Miyazaki where the tank rotifers were sampled before daily supplementation
of microalgae (Nannochloropsis). Crude protein content of the tank rotifers did not decrease markedly although the proportion of lysine tended to decrease.
Vitamin C and E contents of the tank rotifers decreased significantly only at Miyazaki. Calcium content of the tank rotifers
increased at Kamiura and Miyazaki, and the increases in iron and manganese contents of the tank rotifers at Miyazaki and zinc
content at Kagoshima were pronounced. These results suggest that the nutritional value of rotifers in larval-rearing tanks
without water exchange can be maintained by appropriate supplementation of microalgae. The effect of certain minerals that
became high in tank rotifers on subsequent larval development requires further investigation. 相似文献
