To clarify the biosynthetic pathway for syringyl lignans, especially syringyl tetrahydronaphthalene lignans and formation of the C2–C7′ linkage, production of (+)-lyoniresinol (LYR) and its predicted intermediates [syringaresinol (SYR), 5,5′-dimethoxylariciresinol (DMLR), and 5,5′-dimethoxysecoisolariciresinol (DMSLR)] in Lyonia ovalifolia var. elliptica was investigated by means of feeding experiments with radiolabeled precursors. Following individual administration of l-[U-14C]phenylalanine (Phe), [8-14C]sinapyl alcohol (SA), and [8,8′-14C]SYR to excised young shoots of L. ovalifolia and their subsequent metabolism, free [14C]lignans and [14C]lignan glycosides were extracted with methanol from stems and leaves and were divided into ethyl acetate-soluble fractions (lignans) and aqueous fractions (lignan glycosides), respectively. Using a combination of xylanase, cellulase, and β-glucosidase, the glycosides were hydrolyzed to liberate [14C]lignans as aglycones. l-[U-14C]Phe was incorporated into (+)-[14C]SYR [stem 0.38%, 8% enantiomeric excess (e.e.)], (−)-[14C]SYR (leaves 2.75%, 72% e.e.), (+)-[14C]DMLR (stem 0.07%, 18% e.e. and leaves 0.009%, 58% e.e.), (−)-[14C]DMSLR (stem 0.03%, 46% e.e. and leaves 0.05%, 20% e.e.), (+)-[14C]LYR (leaves 0.013%, 22% e.e.) and glycosides of (+)-[14C]LYR (stem 0.036%, 50% e.e.) in 24h. Based on the percent incorporation and enantiomeric composition of the lignans, the biosynthetic pathway of (8R,8′R)-(+)-LYR was proposed as follows: a nonselective dehydrogenative dimerization of sinapyl alcohol yields (±)-SYR, which is reduced with low specificity to give (8R,8′R)-(+)-DMLR. This is cyclized to directly give (+)-LYR as well as reduced again to (8R,8′R)-(−)-DMSLR. Although further transformation of (−)-DMSLR also leads to the formation of (+)-LYR, cyclization could be a main pathway for (+)-LYR biosynthesis. We investigated the energy metabolism in roots of flooded Melaleuca cajuputi Powell, a tropical flood-tolerant tree species, by measuring adenylate concentrations and activities of glycolytic and fermentative enzymes under flooded conditions. Adenylate energy charge (AEC) decreased slightly to 0.72 on the second day of flooding and recovered to around 0.8 by the fourth day of flooding. Activities of pyruvate decarboxylase (EC 4.1.1.1) and alcohol dehydrogenase (EC 1.1.1.1) increased initially and then decreased to the control level after 14 days of flooding. On the other hand, activities of pyruvate kinase (EC 2.7.1.40), phosphoenolpyruvate phosphatase (EC 3.1.3.2), and a series of phosphoenolpyruvate carboxylase (EC 4.1.1.31), malate dehydrogenase (EC 1.1.1.37), and NADP dependent malic enzyme (EC 1.1.1.40), which can convert PEP into pyruvate, were not induced in flooded roots throughout the experiment. These results suggest that neither the downstream reactions of glycolysis nor ATP production via glycolysis was enhanced by flooding, whereas alcohol fermentation was enhanced. With the low ATP yield of the glycolysis–alcohol fermentation pathway and no induction of glycolytic enzymes, the glycolysis–alcohol fermentation pathway itself contributes little to ATP production in flooded roots of M. cajuputi. These physiological responses of M. cajuputi to flooding may have the advantages of surviving flooded conditions because they can avoid exhaustion of sugar and accumulation of ethanol, a toxic end product of alcohol fermentation.  相似文献

We investigated optimum self-bonding conditions of kenaf core composites manufactured by steam treatment, and discussed on the roles of cinnamic acids in the self-bonding mechanism. The presence of cinnamic acids in the kenaf core and its composites were analyzed by pyrolysis gas chromatography-mass spectrophotometry in the presence of tetramethyl ammonium hydroxide (TMAH/Py-GC-MS). The results showed that the optimum bonding properties of kenaf core composites were achieved under these conditions: steam pressure of 0.8–1.0 MPa and pressing time of 10–15 min were able to provide shear strength of 0.40–0.42 MPa while having 2–5% of weight loss. Lignin analysis showed that steam-treated kenaf core composites had a lower proportion of syringyl- to guaiacyl-derived moieties and also cinnamic acids to guaiacyl-derived moieties than its native counterpart. The results indicated that some parts of the ester-linked cinnamic acids were also cleaved due to the degradation of hemicelluloses and lignin during steam treatment. Based on these results, it was concluded that in addition to three main components, the cinnamic acid was also suggested to participate in the self-bonding mechanism of non-wood lignocellulosic binderless boards.  相似文献

We conducted creep tests to evaluate creep behaviors of conventional Japanese framing (jikugumi) structures as reported in a previous article. We measured beam deflections of two structures: one of them was composed of only green timbers (G) and the other with only kiln-dried timbers (D). Besides the two structures, we prepared green and kiln-dried beams to measure moisture content (MC), weight, and dynamic Young’s modulus (E f) by the longitudinal vibration method. We attempted to predict deflections of beams in the structures by using experimental data for single beam specimens. The proposed simple predictive model was derived from two equations: a relation between MC and equilibrium moisture content calculated with temperature and relative humidity, and a relation between MC change and relative deflection change. Beam deflections were traced for 2.5 years, while the predictions were based on experimental data from loading to the 11th day of the test. It was assumed that sensitivity of deflection change to MC should differ during desorption or adsorption. Although annual cyclic changes were observed in E f, there was no obvious relationship between E f and beam deflection. Aging is a major risk factor for spermatogenesis deterioration. However, the influence of age on spermatogenic stem cells and their progenitors in bulls is largely unknown. Here, we report age-related changes in undifferentiated and differentiating spermatogonia in Japanese Black cattle with nearly constant sperm output, by using spermatogonial markers. The numbers of differentiating spermatogonia and more differentiated spermatogenic cells were significantly decreased in aged bovine testes compared with those in young testes. In contrast, the number of undifferentiated spermatogonia was maintained, and their proliferative activity did not differ significantly between young and aged bovine testes. Although severe calcification was only observed to a small extent in aged testes, fewer Sertoli cells and interstitial fibrosis were observed in noncalcified testicular regions. These results suggest that, even in old bulls with nearly constant sperm output, testicular spermatogenic activity declined whereas undifferentiated spermatogonia numbers were maintained. Thus, we propose that undifferentiated spermatogonia may be resistant to age-related changes in bovine testes. Because undifferentiated spermatogonia may contain stem cell activity, our findings highlight the potential utility of undifferentiated spermatogonia as an agricultural resource to produce spermatozoa beyond the natural bovine lifetime through transplantation and in vitro spermatogenesis in future animal production.  相似文献

Although gpt delta rats, as reporter gene-transgenic rats, were originally developed for in vivo mutation assays, they have also been used to evaluate chemical carcinogenesis and comprehensive toxicity. Therefore, it is necessary to accumulate background data on carcinogenicity and general toxicity in gpt delta rats. Here, we investigated the background data of 110-week-old male and female F344 gpt delta rats and wild-type rats. There was no effect of reporter gene transfection on animal survival rates and body weights during the experiment. The relative weight of male gpt delta rat adrenals was significantly higher than that of wild-type rats, possibly due to the higher incidence of pheochromocytoma. There were no intergenotype differences in the incidence of nonneoplastic lesions in both sexes, including chronic progressive nephropathy and focus of cellular alteration in the liver, which had a higher incidence in both genotypes. Additionally, the significantly higher incidence of adrenal pheochromocytoma in male gpt delta rats than that in wild-type rats was likely incidental because of the lack of differences in the incidences of preneoplastic (male and female) and neoplastic (female) adrenal lesions in both genotypes. Other neoplastic lesions in both sexes showed no intergenotype differences in incidence rates, although large granular lymphocytic leukemia in the spleen and Leydig cell tumors in the testes of males showed higher incidence rates. Overall, there were no effects of reporter gene transfection on the spectrum of spontaneous lesions in F344 gpt delta rats, thus supporting their applicability in evaluating chemical toxicity and carcinogenicity.  相似文献

The blood luteinizing hormone (LH) surge in cows is well studied. However, little is known about urinary LH in cows. This study examined urinary LH concentrations after administration of gonadotropin-releasing hormone (GnRH) in six Japanese black cows to induce LH secretion from the pituitary gland into the bloodstream. Abrupt rises in plasma and urinary LH were observed after GnRH administration. Plasma and urinary LH peaked at 2 and 5 hr, respectively. A positive correlation was observed between plasma LH concentrations and urinary LH amounts. Ovulation was confirmed in the cows after 48 hr of GnRH administration. These data strongly suggest that urinary LH is derived from plasma LH, which triggers ovulation in cows.  相似文献

Preimplantation genomic selection combined with an in vitro embryo production system is expected as a means of accelerating genetic improvement in cattle. While micromanipulation-based biopsy approaches are often used to collect embryonic cells for genetic testing, they require expensive equipment and sophisticated skills, hindering the adoption of this system. In the present study, to develop a simple method for preimplantation genomic selection using the blastomere separation (BS) technique in bovine in vitro fertilized embryos, we examined the accuracy of single nucleotide polymorphism (SNP) genotyping and optimal cryopreservation method in demi-blastocysts produced by the BS technique. We demonstrated reliable SNP genotyping using DNA derived from demi-blastocysts. We indicated a suitable equilibrium time in vitrification solution for demi-blastocysts and succeeded obtaining pregnancies by the transfer of vitrified demi-blastocysts. In conclusion, our findings suggest that the BS technique provides a simple method for preimplantation genomic selection in bovine in vitro fertilized embryos.  相似文献

Several successful in vitro culture experiments have used oocyte-cumulus cell-mural granulosa cell complexes (OCGCs) from early antral follicles (0.5–0.7 mm) for the growth of bovine oocytes. However, in studies related to in vitro oocyte maturation and in vitro embryo production, oocyte-cumulus cell complexes (OCCs) that have no mural granulosa cells have been widely used instead of OCGCs. The purpose of this study was to determine whether cumulus cells alone support oocyte growth. First, OCCs and OCGCs were cultured in vitro for 14 days to compare the integrity of the complexes as well as antrum formation. After 14 days, the diameter and meiotic competence of oocytes in OCCs and OCGCs were examined. Oocytes in OCCs grew fully and acquired meiotic competence similar to OCGCs, whereas antrum formation occurred later in OCCs as compared to OCGCs. Subsequently, the effects of follicle stimulating hormone (FSH) on in vitro growth of OCCs were examined for 14 days. When FSH was added to the culture medium, OCCs formed antrum-like structures one day earlier than those cultured without FSH. Oocytes cultured with 1 mIU/ml FSH grew fully and acquired meiotic competence. In contrast, when oocytes were cultured in media containing high concentrations of FSH, some of the OCCs collapsed and the number of degenerated oocytes increased. In conclusion, bovine oocytes in OCCs grow and acquire meiotic competence similar to OCGCs and, 1 mIU/ml FSH supports the development of OCCs and oocyte growth as observed in our culture system.  相似文献
