Soil water concentrations of cadmium and zinc were measured in plant pots with 15 contaminated soils which differed in origin, texture, pH (5.1 – 7.8) and concentrations of cadmium (0.2 – 17 mg Cd kg-1) and zinc (36 – 1300 mg Zn kg-1). The soil waters contained total concentrations of 0.5 to 17 µg Cd L-1 and 9 to 3600 µg Zn L-1, which were dominated by free metal ions as measured by an ion exchange-resin method. Annual leaching outflows were estimated from soil water concentrations to be 0.5 – 17 g Cd ha-1 y-1 and 9 – 3600 g Zn ha-1 y-1 per 100 mm of net percolation, corresponding to 0.1% per year of the total soil content of cadmium and zinc. The measured soil water concentrations of cadmium and zinc did not correlate linearly with the corresponding soil concentrations but correlated fairly well with concentrations measured in Ca(NO3)2 extracts of the soils and with soil water concentrations estimated from soil concentrations and pH. Such concentration estimates may be useful for estimating amounts of cadmium and zinc being leached from soils.  相似文献

The present study examined whether recombinant porcine leptin alters lipid synthesis in porcine adipocytes. The stromal-vascular cell fraction of neonatal pig subcutaneous adipose tissue was isolated by collagenase digestion, filtration, and subsequent centrifugation. These cells were seeded on 25-cm2 tissue culture flasks and proliferated to confluency in 10% (vol/vol) fetal bovine serum in Dulbecco's modified Eagle medium/F12 (DMEM/F12, 50:50). Cultures were differentiated using 2.5% pig serum (vol/vol), 10 nM insulin, 100 nM hydrocortisone. After 7 d of lipid filling, cultures were washed free of this medium, incubated overnight in DMEM/F12 containing 2% pig serum (vol/vol), and then used for experiments. Acute experiments assessed U-(14)C-glucose or 1-(14)C-palmitate metabolism in cultures exposed to porcine leptin (0 to 1,000 ng/mL medium) for 4 h. Chronic experiments used cultures incubated with 0 to 1,000 ng porcine leptin/mL medium for 44 h before measurements of U-(14)C-glucose and 1-(14)C-palmitate oxidation and incorporation into lipid. Another experiment examined whether chronic leptin treatment alters insulin responsiveness by including insulin (10 nM) with incubations containing leptin. Leptin had no acute effects on glucose oxidation or conversion to lipid (P > 0.05). Acute leptin treatment decreased palmitate incorporation into lipids up to 45% (P < 0.05). Chronic leptin exposure decreased glucose oxidation (21%), total lipid synthesis (18%), and fatty acid synthesis (23%) at 100 ng/mL medium (P < 0.05). Insulin increased rates of glucose oxidation, total lipid, and fatty acid synthesis (P < 0.05); however, chronic exposure to 10 ng leptin/mL medium decreased the effectiveness of 10 nM insulin to affect these measures of glucose metabolism by approximately 18 to 46% (P < 0.05). Higher concentrations of leptin inhibited all effects of insulin on glucose metabolism (P < 0.05). Chronic exposure to leptin increased palmitate oxidation by 36% (P < 0.05). Chronic leptin exposure decreased palmitate incorporation into total lipids by 40% at 100 ng/mL medium (P < 0.05). Lipoprotein lipase activity was not affected (P > 0.05) by leptin. These data indicate that leptin functions to promote partitioning of energy away from lipid accretion within porcine adipose tissue by inhibiting glucose oxidation and lipogenesis indirectly, by decreasing insulin-mediated stimulation of lipogenesis, and by stimulating fatty acid oxidation while inhibiting fatty acid esterification.  相似文献

The aim of the present study was to evaluate possible injury provoked by an intensive reproductive programme based on rectal palpation in the cow. Alterations to the mesothelial surface of the peritoneum and to the oviductal mucosa were explored by scanning electron microscopy (SEM). Nine multiparous Friesian cows were selected from each of two commercial dairy herds. Cows in herd 1 were maintained on a weekly reproductive health programme, while those in herd 2 were not subjected to gynaecological exploration. Surface features of the mesothelial cells of the reproductive tract and adjacent ligaments, and the oviductal mucosa were explored by SEM observation of tissue specimens from each animal. Six cows in the intensive reproductive programme showed microscopic alterations of the peritoneal surface. Strands were observed in the oviducts and adjacent ligaments in five of these cows. These strands, composed of filaments covered with mesothelial cells, were probably adhesions. Cells with abundant vacuoles on their surface were detected in the mesothelium corresponding to three animals, two of which also showed microscopic strands. No peritoneal surface alterations were observed in the cows from herd 2. No oviductal mucosal modifications were detected in either group. These findings suggest that intensive reproductive programmes based on rectal palpation may increase the risk of peritoneal surface alterations in the cow, but effects on reproductive physiology are unlikely.  相似文献

The influence of transplanting on height, root neck diameter, and grading was investigated for Carpinus betulus, Quercus robur, and Euonymus europaeus on three sites with different climate and soil properties. Additionally, plant growth was characterized by the shoot-to-root ratio, height-to-root neck diameter, and proportions of coarse and fine roots. The significance of the growth parameters as criteria for survival is discussed. Transplanting influenced shoot growth more than root growth. The growth of the plants was highly site-dependent. Transplanting increased growth most on the site with the best conditions for water and N supply. Transplanting had no effect on grading for the site with the worst water supply.  相似文献

Sunlight is known to reduce the incidence of blister blight disease of tea, caused by Exobasidium vexans . The effects of the ultraviolet-B (UV-B: 290–320 nm) component of solar radiation on the disease and the development of E. vexans were studied in the field in Sri Lanka using UV-screening filter materials held over a commercial crop. Exclusion of UV-B radiation by Polyester, which reduces fluxes in the UV-B region by approximately 75–85%, increased both the number of translucent spots (immature sites of infection) and number of sporulating blisters (mature sites of infection), but it had little or no effect on the sporulation of the pathogen. When basidiospores were artificially inoculated on leaves, and were exposed to full or filtered solar radiation, their survival and germination increased when UV-B wavelengths were removed; significant differences were found 62 h after inoculation. However, UV-B did not affect the extension of germ tubes. This study indicates that the UV-B component of solar radiation plays an important role in the natural regulation of blister blight disease in the field.  相似文献

A mathematical model was developed of the dynamics of a plant virus disease within a spatially-referenced lattice of fields of a host crop. The model can be applied to crops in continuous, contiguous cultivation such as tropical irrigated rice. Disease progress in each field of the host crop was assumed to be logistic and determined by incidence within the field itself as well as incidence in neighbouring fields, depending on the gradient of disease spread. The frequency distribution of planting dates (represented by the proportion of the total number of fields planted in successive months) was assumed to follow a normal distribution and the variance of planting date was used as a measure of cropping asynchrony. Analysis of the model revealed that disease incidence within the lattice (i.e. mean incidence over all fields) depended upon the infection efficiency, the slope of the dispersal gradient, and the variance in planting date. Disease endemicity depended mainly on planting date variance and disease persisted in the lattice if this variance exceeded a certain threshold. Above the threshold for persistence, the response of mean disease incidence to planting date variance was non-linear and the region of greatest sensitivity was closest to the threshold. Thus, disease systems that show moderate rather than high cropping asynchrony are more likely to be influenced by changes in the variance of planting date. Implications for the area-wide management of rice tungro virus disease are discussed.  相似文献
