This phase II, randomized, open-label field trial was designed to evaluate and compare the safety and efficacy of four treatment durations (10, 20, 28 or 40 days) with marbofloxacin administered orally at the dosage of 2mg/kg once a day for canine visceral leishmaniosis. Twenty-four dogs naturally infected with visceral leishmaniosis and without biochemical disorder evidences of renal insufficiency, were recruited by two Greek veterinarian clinics. They were also randomly assigned to one of the four treatment duration groups, and have been clinically, haematologically, biochemically and parasitologically followed-up regularly for 9 months. Efficacy was achieved for 5/6 dogs treated for 28 days, 4/6 dogs treated for 10 or 20 days and for 3/6 dogs treated for 40 days. Moreover, efficacy was reached more quickly (58.4 days) in dogs treated for 28 days. Improvement of clinical signs tended to be better and faster in the 28 days treatment group too. After 9 months of follow-up, a total of three cases could be considered as relapsing (two dogs treated for 40 days and one dog treated for 28 days). There was a significant reduction in amastigotes density in macrophages after 3 months in the four groups when compared with the parasite density at inclusion. No adverse effects were noticed during this 9 months study. Results obtained with marbofloxacin at the dosage of 2mg/kg once a day for 28 days seem encouraging and may offer a safe alternative for treating canine visceral leishmaniosis.  相似文献

Recent publications indicate that single‐step models are suitable to estimate breeding values, dominance deviations and total genetic values with acceptable quality. Additive single‐step methods implicitly extend known number of allele information from genotyped to non‐genotyped animals. This theory is well derived in an additive setting. It was recently shown, at least empirically, that this basic strategy can be extended to dominance with reasonable prediction quality. Our study addressed two additional issues. It illustrated the theoretical basis for extension and validated genomic predictions to dominance based on single‐step genomic best linear unbiased prediction theory. This development was then extended to include inbreeding into dominance relationships, which is a currently not yet solved issue. Different parametrizations of dominance relationship matrices were proposed. Five dominance single‐step inverse matrices were tested and described as C1 , C2 , C3 , C4 and C5 . Genotypes were simulated for a real pig population (n = 11,943 animals). In order to avoid any confounding issues with additive effects, pseudo‐records including only dominance deviations and residuals were simulated. SNP effects of heterozygous genotypes were summed up to generate true dominance deviations. We added random noise to those values and used them as phenotypes. Accuracy was defined as correlation between true and predicted dominance deviations. We conducted five replicates and estimated accuracies in three sets: between all ( S1 ), non‐genotyped ( S2 ) and inbred non‐genotyped ( S3 ) animals. Potential bias was assessed by regressing true dominance deviations on predicted values. Matrices accounting for inbreeding ( C3 , C4 and C5 ) best fit. Accuracies were on average 0.77, 0.40 and 0.46 in S1 , S2 and S3 , respectively. In addition, C3 , C4 and C5 scenarios have shown better accuracies than C1 and C2 , and dominance deviations were less biased. Better matrix compatibility (accuracy and bias) was observed by re‐scaling diagonal elements to 1 minus the inbreeding coefficient ( C5 ).  相似文献

An in situ determination of biological soil‐crust stability was carried out in two study sites along a sharp rainfall gradient. Penetration resistance (PR) of the crusts was measured using a newly developed light‐weight needle‐type electronic micropenetrometer. The depth‐related PR data revealed two sections possessing different structures and stability. The topcrust (0–2 mm) had significantly higher amounts of N, organic C, carbonates, and salts as compared to the underlain subcrust. The mean PR of the topcrust was 0.68 MPa for the southern study site, Nizzana‐South (≈ 100 mm annual rainfall), and it increased significantly for the northern study site, Nizzana‐69 (≈ 170 mm annual rainfall), with a mean of 1.11 MPa. A subcrust (2–30 mm) was identified that was characterized by a high amount of carbonates and a PR > 2 MPa. The electronic micropenetrometer system is a promising device for identifying areas of changing crust stability in relation to biological soil‐crust properties. The overall stability of biological soil crusts depends on the topcrust and subcrust structure. This structure is linked to abiotic and biotic factors and likely in relation to the amount of annual precipitation.  相似文献

Our understanding of how mineral nutrition affects productivity and composition of bioenergy crops grown on marginal lands remains fragmented and incomplete despite world‐wide interest in using herbaceous biomass as an energy feedstock. Our aim was to determine switchgrass (Panicum virgatum L.) biomass production and maize (Zea mays L.) grain yield on marginal soils used previously to evaluate the effect of soil phosphorus (P) and potassium (K) fertility on alfalfa (Medicago sativa L.) forage production. Grain yield of maize was reduced on P‐ and/or K‐limited plots that also impaired alfalfa forage yield, whereas switchgrass biomass yields were high even in plots possessing very low available P (4 mg kg–1) and K (< 70 mg kg–1) levels. Linear‐plateau regression models effectively described the relationship of soil test P and K to tissue P and K concentrations, and tissue P and K concentrations accurately predicted removal of P and K in harvest biomass. However, neither soil‐test P and K, nor tissue P and K concentrations were effective as diagnostics for predicting switchgrass biomass yield nor could soil tests and their change with cropping predict nutrient removal. Concentrations of cellulose, hemicellulose, lignin, and ash were not influenced by P and K nutrition. Predicted bio‐ethanol production was closely associated with biomass yield whereas high biomass K concentrations reduced estimated bio‐oil production per hectare by as much as 50%. Additional research is needed to identify diagnostics and managements to meet the bioenergy production co‐objectives of having high yield of biomass with very low mineral nutrient concentrations (especially K) while sustaining and improving the fertility of marginal soils.  相似文献

A comparative serologic and virologic study was performed in pigs from 5 herds with postweaning multisystemic wasting syndrome (PMWS) and 2 herds without PMWS in Quebec. In each herd, 60 blood samples were collected at 4-wk intervals from pigs from 3 to 23 wk of age. The serum was evaluated for the presence of antibodies to porcine circovirus type 2 (PCV2) and porcine reproductive and respiratory syndrome virus (PRRSV), as well as for the presence of nucleic acid of PCV2, PRRSV, and porcine parvovirus (PPV), by means of the polymerase chain reaction (PCR). Serologic profiles for PCV2 were very similar in 6 of the 7 herds, including the 2 without PMWS, and were characterized by a gradual decrease in antibody titres from 3 until 11 wk of age, followed by seroconversion at 15 wk, and high PCV2 antibody titres thereafter in all pigs. Only starting at 11 to 15 wk of age could PCV2 viremia be detected, except in 1 herd, in which clinical signs were observed at 6 to 7 wk of age. A PCV2 viremia could be detected within the same pigs for a minimum of 8 wk, and the virus could still be detected in 41% of the serum samples obtained at 23 wk of age. The antibody level did not appear to influence the occurrence of disease, since titres were similar in pigs in the herds with or without PMWS. Infection with PRRSV, as demonstrated by PCR and seroconversion, preceded that of PCV2 by at least 1 mo in both types of herd. Both PRRSV and PCV2 were detected in some pigs in 5 of the 7 herds, including 1 herd without PMWS. Porcine parvovirus could be detected in serum by PCR in 2 herds with PMWS after the onset of clinical signs and also in 1 herd without PMWS. Genomic analysis of PCV2 strains identified in the herds without PMWS indicated complete or very high homology (99.4% to 100%) with the PCV2 strains identified in 4 herds with PMWS. In our field study, the triggering of PMWS in the herds could not be linked to coinfection with either PRRSV or PPV or to the use of a specific immunostimulant, such as vaccines, or to particular genomic differences between the PCV2 strains identified.  相似文献

We compared N fluxes in a 150-year-old Fagus sylvatica coppice and five adjacent 25-year-old plantations of Fagus sylvatica, Picea abies, Quercus petraea, Pinus laricio and Pseudotsuga menziesii. We measured net N mineralization fluxes in the upper mineral horizon (A1, 0–5 cm) for 4 weeks and gross N mineralization fluxes for two days. Gross rates were measured during the 48-h period after addition of 15NH4 and 15NO3. Mineralization was measured by the 15NH4 dilution technique and gross nitrification by 15NO3 production from the addition of 15NH4, and by 15NO3 dilution. Net and gross N mineralization was lower in the soil of the old coppice, than in the plantations, both on a soil weight and organic nitrogen basis. Gross nitrification was also very low. Gross nitrification measured by NO3 dilution was slightly higher than measured by 15NO3 production from the addition of 15NH4. In the plantations, gross and net mineralization and nitrification from pool dilution were lowest in the spruce stand and highest in the beech and Corsican pine stands. We concluded that: (1) the low net mineralization in the soil of the old coppice was related to low gross rate of mineralization rather than to the concurrent effect of microbial immobilisation of mineral N; (2) the absence of nitrate in the old coppice was not related to the low rate of mineralization nor to the absence of nitrifyers, but most probably to the inhibition of nitrifyers in the moder humus; (3) substituting the old coppice by young stands favours nitrifyer communities; and (4) heterotrophic nitrifyers may bypass the ammonification step in these acid soils, but further research is needed to check this process and to characterize the microbial communities.  相似文献
