The effect of ovotransferrin (ovoTF), human lactoferrin (hLF) and bovine lactoferrin (bLF) on the obligate intracellular pathogen Chlamydophila (Cp.) psittaci was evaluated using a model of Buffalo Green Monkey kidney (BGM) cells and HD11 chicken macrophages as artificial hosts. Firstly, the effect of transferrins on the infectivity of the bacteria was evaluated. Pre-incubation of Cp. psittaci with 0.5 to 5 mg/mL ovoTF prior to infecting BGM cells significantly lowered the infection rate (P < 0.05). For both lactoferrins, the infection rate could only be reduced with 5 mg/mL, albeit not significantly as compared to the infection rate created by the untreated bacteria. Secondly, transferrins were tested for their ability to influence bacterial adhesion and entry in HD11 cells. Maximal non-cytotoxic and non-bactericidal concentrations of 0.05 mg/mL ovoTF and 0.5 mg/mL hLF and bLF were used. Overall, ovoTF was more effective than human and bovine LF in inhibiting bacterial irreversible attachment and cell entry and the latter was accompanied by a dose-dependent reduction of actin recruitment at the bacterial entry site. However, once bacteria had entered HD11 cells, transferrins had apparently no effect on intracellular replication. The present findings suggest a possible role for transferrins and especially ovoTF, in preventing avian Cp. psittaci infections.  相似文献

Recombinant human thyroid-stimulating hormone (rhTSH) was evaluated for the diagnosis of canine hypothyroidism, using TSH response tests. Phase I stimulation tests were performed in 6 healthy dogs weighing over 20 kg, using 50 and then 100 microg of freshly reconstituted rhTSH administered intravenously. In phase II, the same dogs were stimulated by using 100 microg of rhTSH frozen for 3 months at -20 degrees C. Phase III stimulation tests were performed by using 50 or 100 microg of freshly reconstituted or frozen rhTSH in healthy (n = 14), euthyroid sick (n = 11) and hypothyroid dogs (n = 9). A dose of 100 microg of rhTSH was judged more appropriate for dogs weighing more than 20 kg. Biological activity of rhTSH after freezing at -20 degrees C for up to 12 weeks was maintained. When stimulated, significant (P < 0.05) increases in total thyroxine concentration were observed only in healthy and euthyroid sick dogs. Results of this study show that the rhTSH stimulation test is able to differentiate euthyroidism from hypothyroidism in dogs.  相似文献

Tipburn is a calcium related and environmentally induced physiological disorder causing economic damage in all lettuce (Lactuca sativa L.) production regions. The objectives of this research were to determine (1) the genetic variation for tipburn incidence, (2) the genotype (G) × environment (E) interaction (GE) for tipburn incidence, and (3) the efficiency of field selection for tipburn resistance. Tipburn incidence was recorded over 2 years in Salinas, CA, and Yuma, AZ, for 55 romaine, crisphead, green leaf, and red leaf type cultivars, and over 3 years in Quebec for 15 romaine cultivars. Analysis revealed that G, E, and GE affected tipburn incidence, including crossover interactions that were not repeatable over years. This indicates that cultivar/breeding line evaluations should be based on mean performance and stability over multiple environments. Among lettuce types, only crisphead had significant genetic variability for tipburn resistance, reflecting the greater breeding effort applied to this type compared romaine, green and red leaf types. Analysis of a dataset with five romaine cultivars in eight environments in California, Arizona, and Quebec for 2 years revealed that Yuma in 2006 and Saint-Blaise in 2005 were highly correlated (r = 0.923, P < 0.05), and were the most discriminating and most representative environments for tipburn evaluation. Single plant selection for tipburn resistance in three F2 romaine populations was ineffective. Further, the degree of head closure was significantly associated with tipburn incidence. Identification and selection of morphological characters associated with resistance in conjunction with direct selection against tipburn may be an effective method for genetic improvement of tipburn resistance.  相似文献

Media-based bacteriological testing will fail to detect non-culturable organisms and the risk of consuming viable but non-culturable (VBNC) Listeria monocytogenes is unknown. We have here studied whether L. monocytogenes obtained from seafoods, processing environment and clinical cases enter the VBNC state and assessed the virulence of the non-culturable forms of the bacteria. A number of 16 L. monocytogenes strains were starved in microcosm water at 4 °C until loss of culturability. Metabolic activity in the VBNC form was measured as ATP generation using a luciferase assay and membrane integrity was examined using the LIVE/DEAD BacLight assay. All tested L. monocytogenes strains entered the VBNC state after starvation in microcosm water. Ongoing mRNA synthesis of hly in VBNC L. monocytogenes cells re-incubated in culture medium indicated a potential virulence of these forms. Sodium pyruvate and replenishment of nutrient were used in attempts to resuscitate VBNC cells. However, VBNC L. monocytogenes were not resuscitated under these conditions. VBNC L. monocytogenes were tested for virulence in a cell plaque assay and by intraperitoneally inoculation in immunodeficient RAG1−/− mice. Inoculation of VBNC L. monocytogenes in immunodeficient mice did not cause morbidity, and plaque assay on HT-29 cells in culture indicated that the VBNC cells were avirulent. The results indicate that the risk of non-culturable L. monocytogenes in foods, when the VBNC state is induced by starvation, is negligible.  相似文献

Quantitative scenarios are coming of age as a tool for evaluating the impact of future socioeconomic development pathways on biodiversity and ecosystem services. We analyze global terrestrial, freshwater, and marine biodiversity scenarios using a range of measures including extinctions, changes in species abundance, habitat loss, and distribution shifts, as well as comparing model projections to observations. Scenarios consistently indicate that biodiversity will continue to decline over the 21st century. However, the range of projected changes is much broader than most studies suggest, partly because there are major opportunities to intervene through better policies, but also because of large uncertainties in projections.  相似文献

Although it is well accepted that the present Asian H5N1 panzootic is predominantly an animal health problem, the human health implications and the risk of human pandemic have highlighted the need for more information and collaboration in the field of veterinary and human health. H5 and H7 avian influenza (AI) viruses have the unique property of becoming highly pathogenic (HPAI) during circulation in poultry. Therefore, the final objective of poultry vaccination against AI must be eradication of the virus and the disease. Actually, important differences exist in the control of avian and human influenza viruses. Firstly, unlike human vaccines that must be adapted to the circulating strain to provide adequate protection, avian influenza vaccination provides broader protection against HPAI viruses. Secondly, although clinical protection is the primary goal of human vaccines, poultry vaccination must also stop transmission to achieve efficient control of the disease. This paper addresses these differences by reviewing the current and future influenza vaccines and vaccination strategies in birds.  相似文献

The behavior of different exogenous enzymes (soybean lipoxygenase [SLOX], horseradish peroxidase [HPOD], catalase from bovine liver [BCAT], and glucose oxidase [GOX] from Aspergillus niger) added to dough was studied during mixing. The effect of adding these exogenous oxidoreductases on the activity of three oxidative enzymes present in wheat flour (lipoxygenase [WLOX], peroxidase [WPOD], and catalase [WCAT]) was examined. Proper assay conditions were established to differentiate between added WLOX, WPOD, and WCAT and the corresponding activities present in wheat flour. For doughs with added SLOX, an immediate loss of extractable SLOX (≈40%) was observed which remained constant during further mixing. When compared with the control dough, addition of SLOX decreased the losses in WLOX and WCAT activities, whereas WPOD activity was unaffected. With doughs supplemented by HPOD, an immediate loss of 20% in the HPOD activity was observed which did not change after 20 min of mixing. Compared with control dough, addition of HPOD did not affect the behavior of WLOX and WPOD, whereas a slight decrease in the WCAT losses was observed. Addition of BCAT to the dough did not change the behavior of WLOX and WPOD, whereas the losses in WCAT were less rapid. Half of the extractable activity of BCAT was lost at the beginning of mixing with no change during further mixing. For doughs supplemented with GOX, 25% of the GOX activity was lost in the first 5 min of mixing and an additional loss of 20% was observed after 20 min of mixing. Compared with dough without GOX, addition of GOX decreased the losses in WLOX, whereas losses in WCAT and WPOD increased. Glucose and ferulic acid were also added to doughs supplemented with GOX. Added glucose decreased the losses in GOX and WLOX and did not change the behavior of WPOD and WCAT during mixing. Addition of ferulic acid promoted a slight increase of the losses in WLOX and WCAT and almost no change for GOX and WPOD.  相似文献

Abnormal compressions and fusions of vertebral bodies are frequently observed in reared rainbow trout and could result from chronic and unbearable muscle pressures acting on the axial skeleton during intensive growth. Sustained swimming at moderate speeds was shown to induce many positive effects on growth and swimming performances in salmonids, but yet little is known about its effects on vertebral remodeling processes and related hormonal regulation. Rainbow trout were subjected to three different swimming speeds (0, 1.0 and 1.5 body length (BL) s− 1), starting one month after they were first fed (65 mm) and ending when they reached 260 mm in size (market-size of 275 g). At the end of the experiment, 20 trout were sampled in each lot (N = 60) and blood samples were taken. Vertebrae abnormalities were assessed by radiological examinations. Vertebrae from the middle axial region (V32–38) were selected to evaluate bone mineralization (BM) and total bone area (Tt-B.Ar.) on radiographed transverse sections (125 ± 10 μm). Assays were performed to evaluate mineral homeostasis (calcemia and phosphatemia), bone cell activities (alkaline phosphatase, ALP, and tartrate-resistant acid phosphatase, TRAP) and bone regulating hormones (calcitonin, CT and thyroid hormones, THs). Sustained exercise reduced the appearance of fused vertebrae, enhanced vertebral BM and decreased vertebral Tt-B.Ar., while it increased circulating CT and TH levels. No variations were observed on mineral homeostasis and bone cell activities. Increasing the swimming speed up to 1.5 BL s− 1 had positive effects on the vertebral skeleton, and therefore, seems to be a suitable approach to prevent aggravation of vertebral abnormalities in juvenile trout. The changes observed in vertebral features are interpreted as a compromise between the necessity to mobilize vertebral mineral ions in response to various physiological demands and to maintain vertebral strength against mechanical constraints.  相似文献
