Nonlethal exposure of honey bees to thiamethoxam (neonicotinoid systemic pesticide) causes high mortality due to homing failure at levels that could put a colony at risk of collapse. Simulated exposure events on free-ranging foragers labeled with a radio-frequency identification tag suggest that homing is impaired by thiamethoxam intoxication. These experiments offer new insights into the consequences of common neonicotinoid pesticides used worldwide.  相似文献

In puppies weaning is a high risk period. Fecal changes are frequent and can be signs of infection by digestive pathogens (bacteria, viruses, parasites) and indicators of nutritional and environmental stress. The aim of this study was to define a pathological fecal score for weaning puppies, and to study the impact on that score of two intestinal viruses (canine parvovirus type 2 and canine coronavirus). For this, the quality of stools was evaluated on 154 puppies between 4 and 8weeks of age (100 from small breeds and 54 from large breeds). The scoring was performed immediately after a spontaneous defecation based on a 13-point scale (from 1; liquid to 13; dry and hard feces). Fecal samples were frozen for further viral analysis. Each puppy was weighed once a week during the study period. The fecal score regarded as pathological was the highest score associated with a significant reduction in average daily gain (ADG). Fecal samples were checked by semi-quantitative PCR or RT-PCR for canine parvovirus type 2 and canine coronavirus identification, respectively. The quality of feces was affected by both age and breed size. In small breeds, the ADG was significantly reduced under a fecal score of 6 and 7 for puppies at 4-5 and 6-8weeks of age, respectively. In large breeds, the ADG was significantly reduced under a fecal score of 5 whatever the age of the puppy. Whereas a high viral load of canine parvovirus type 2 significantly impacted feces quality, no effect was recorded for canine coronavirus. This study provides an objective threshold for evaluation of fecal quality in weaning puppies. It also emphasizes the importance to be given to age and breed size in that evaluation.  相似文献

ABSTRACT: Foot-and-mouth disease virus (FMDV) is a highly infectious member of the Picornaviridae inducing an acute disease of cloven-hoofed species. Vaccine-induced immune protection correlates with the presence of high levels of neutralizing antibodies but also opsonising antibodies have been proposed as an important mechanism of the immune response contributing to virus clearance by macrophages and leading to the production of type-I interferon (IFN) by plasmacytoid dendritic cells (pDC). The present study demonstrates that the opsonising antibody titres mediating enhanced IFN-α responses in pDC were similar to neutralizing titres, when antigenically related viruses from the same serotype were employed. However, sera cross-reacted also with non-neutralized isolates of multiple serotypes, when tested in this assay. Both uncomplexed virus and immune complexed virus stimulated pDC via Toll-like receptor 7. An additional finding of potential importance for strain-specific differences in virulence and/or immunogenicity was that pDC activation by FMDV strongly differed between viral isolates. Altogether, our results indicate that opsonising antibodies can have a broader reactivity than neutralizing antibodies and may contribute to antiviral responses induced against antigenically distant viruses.  相似文献

Landscape dynamics increasingly challenge agronomists to explain how and why agricultural landscapes are designed and managed by farmers. Nevertheless, agronomy is rarely included in the wide range of disciplines involved in landscape research. In this paper, we describe how landscape agronomy can help explain the relationship between farming systems and agricultural landscape dynamics. For this, we propose a conceptual model of agricultural landscape dynamics that illustrates the specific contribution of agronomy to landscape research. This model describes the relationship between three elements: farming practices, landscape patterns and natural resources. It can stimulate agronomists to deal with research issues in agricultural landscape dynamics and enhance the interdisciplinary integration of farming systems in wider landscape research. On these premises, we discuss the main research issues that will benefit from an active involvement of agronomy, to understand, but also to assess landscape dynamics and to design relevant decision support systems.  相似文献

A better understanding of cell-mediated immune responses to classical swine fever virus (CSFV) is essential for the future development of improved vaccines. We analyzed the generation of cell-mediated and humoral immune responses in d/d histocompatible pigs following CSFV infection or vaccination. Viral infection induced high T cell responses with high primary and secondary CTL activity correlated with high IFN-gamma production, whereas vaccination with a live vaccine followed by infection mainly induced neutralizing antibody but low cell-mediated responses. Moreover, high IgG1 response was associated with high IFN-gamma response following infection whereas a weak IFN-gamma response was related to a good IgG2 response but a low IgG1 production. These data could reflect Th1/Th2-like balance of immune responses depending upon immunization protocols, which has not yet been described in the pig. T-cell responses to CSFV were evidenced by CSFV-specific CD25 upregulation on CD4-CD8+, but not on CD4+CD8- cells, which further illustrated the importance of CTL responses after infection. Our results indicated that generation of cell-mediated immune responses was much higher following intranasal/oral CSFV infection than after intramuscular vaccination, which implies that the capacity of new CSFV vaccines to induce higher T-cell responses should be considered.  相似文献

Graminaceous cyst nematodes form a group of eleven valid species including Heterodera avenae, Heterodera filipjevi and Heterodera latipons and constitute major pests to cereals. They are widely spread in circum-mediterranean areas where they are presumed to cause yield losses on bread and durum wheat. The objective was to document the diversity of these cereal cyst nematodes, in particular samples from Mediterranean regions, in comparison to species which develop on cultivated or wild grasses (H. arenaria, H. hordecalis, H. mani) and on rice or sugarcane (H. sacchari). Studies involved PCR-RFLP of ITS and morphometrics of the juvenile and cyst characters. UPGMA analysis of molecular data showed that the isolates segregated in two main clusters which seem to represent different evolutionary lineages. The H. avenae sensu lato cluster (I) contained H. arenaria, H. avenae, H. filipjevi and H. mani. The second cluster (II) contained isolates of H. hordecalis and H. latipons. Within H. avenae sensu lato, H. filipjevi was separated from the other related species with significant bootstrap value. The differentiation of H. arenaria, recognized for the first time based on molecular data, and H. mani with few restriction enzymes were the least significant. Intraspecific polymorphism allowed differentiation of isolates originating from Australia within H. avenae sensu stricto. The group H. hordecalis–H. latipons showed the greatest genetic variability between and within isolates. Two representatives of Heterodera sacchari, taxonomically included in the schachtiigrave group, were genetically as distant to this group as to the other graminaceous species belonging to either H. avenae sensu lato or H. hordecalisH. latipons group. Results inferred from multivariate analysis applied on morphometrics of the cysts and juveniles showed agreement between genetic and phenotypic classifications. This study demonstrates the utility of combined molecular and classical methods to enhance our knowledge about the diversity within the complex of graminaceous cyst nematodes and to establish robust techniques to identify a wider set of nematode species.  相似文献

The virulence of Viable But Non-Culturable (VBNC) cells of 4 strains of Listeria monocytogenes was investigated in both a human adenocarcinoma cell line (HT-29) and a mouse model. LO 28, ATCC 19115 and CNL 895807 strains of Listeria monocytogenes became VBNC when incubated in microcosm water at 20 degrees C and Scott A strain at 4 degrees C. No culturable bacteria were detected in the VBNC state, although 104 active cells/mL were found by the Direct Viable Count (DVC) and CTC-DAPI double staining methods. A comparison of virulence in both human adenocarcinoma cell line HT-29 and the mouse model showed that culturable controls were more virulent than VBNC cells, which appeared to be avirulent regardless of the virulence methods applied. Pathogenicity was tested in each model and was lost concomitantly with culturability, whereas some cells were still metabolically active (determined by CTC and DVC). Moreover, amplification of a 388 bp fragment with Immunocapture-PCR revealed the presence of Listeria monocytogenes DNA in all mixed spleen samples after intravenous injection of VBNC cells. These results demonstrate that VBNC cells were present in the mouse spleens. The results of the study suggest that Listeria monocytogenes strains might remain in the aquatic environment for prolonged periods in the VBNC state but these cells were not pathogenic in the conditions tested. These findings demonstrate the value of VBNC studies and show the need to investigate the role of VBNC cells in environmental transmission of Listeria monocytogenes. Further studies are needed in order to investigate the virulence of VBNC cells of Listeria monocytogenes after recovery of a culturable state.  相似文献

Information on earthworm burrowing behaviour is scarce and therefore the evolution of the macroporosity related to earthworm activities is still poorly known. We have designed a new apparatus, ‘Colonne Gamma’, to follow the three‐dimensional trajectories of radio‐labelled earthworms in artificial cores of soil. Earthworms are radio‐labelled by injecting into their coelomic cavity a small source of 60Co (volume 0.1 mm3, intensity 13.5 μCi). The emission of gamma rays is recorded by three detectors carried by a disc that oscillates vertically around the soil core where the earthworm is introduced. We have also developed a deterministic model to estimate the positions of the 60Co source from the number of gamma rays received by each detector during an oscillation. Numerical experiments showed that the uncertainties of estimates were less than 3 mm for each coordinate. To validate the results, we tracked the trajectory (one position every 4 minutes) of a radio‐labelled earthworm for 1 week and compared it with the skeleton of the macroporosity obtained by computer assisted tomography of the same soil core. There was a general qualitative agreement between the trajectory and the skeleton. Moreover, based on the precise study of the successive positions of the earthworm we could distinguish two different kinds of activities in the trajectory: displacement and digging events. The ‘Colonne Gamma’ apparatus therefore has great potential for studies of the ecology and the behaviour of earthworms.  相似文献

Vaccination is a useful option to control infection with porcine reproductive and respiratory syndrome virus (PRRSV), and several modified live-PRRSV vaccines have been developed. These vaccines have shown some efficacy in reducing the incidence and severity of clinical disease as well as the duration of viremia and virus shedding but have failed to provide sterilizing immunity. The efficacy of modified live-virus (MLV) vaccines is greater against a homologous strain compared with heterologous PRRSV strains. The objective of this study was to evaluate the efficacy of Fostera PRRS MLV vaccine in protecting against challenge with a heterologous field strain widely circulating in the swine herds of eastern Canada. Forty-six piglets were divided into 4 groups: nonvaccinated-nonchallenged; nonvaccinated-challenged; vaccinated-challenged; and vaccinated-nonchallenged. The animals were vaccinated at 23 d of age with Fostera PRRS and challenged 23 d later with a heterologous field strain of PRRSV (FMV12-1425619). Overall, the vaccine showed some beneficial effects in the challenged animals by reducing the severity of clinical signs and the viral load. A significant difference between nonvaccinated and vaccinated animals was detected for some parameters starting 11 to 13 d after challenge, which suggested that the cell-mediated immune response or other delayed responses could be more important than pre-existing PRRSV antibodies in vaccinated animals within the context of protection against heterologous strains.  相似文献
