Inflammatory colorectal polyps (ICRPs) in miniature dachshunds are recently recognized as a major cause of large bowel diarrhea in this dog breed in Japan. ICRPs are characterized by the formation of multiple small polyps and a space-occupying large polyp in the colorectal area, and are thought to be a novel form of inflammatory bowel disease (IBD). In humans, specific cytokine patterns attributed to T helper (Th)1, Th17 and regulatory T cells have important roles in the pathogenesis of IBD. Thus, the aim of the present study was to assess the gene expression of cytokines of T cell subsets in the colorectal mucosa from dogs with ICRPs. Colorectal mucosal specimens from 10 dogs with ICRPs and 14 control dogs were used in this study. Interferon (IFN)-γ, interleukin (IL)-4, IL-17A and IL-10 mRNA expression was assessed using quantitative real-time PCR. IL-17A mRNA expression was significantly increased in large polyps compared to small polyps and controls. IFN-γ and IL-10 mRNA expression in large polyps were significantly higher than in controls. There was no significant difference in IL-4 mRNA expression among the three groups. IL-17A is thought to play important roles in the pathogenesis of ICRPs. IL-10 up-regulation could oppose the proinflammatory function of IL-17A.  相似文献

Between September 2010 and November 2011, 350 EDTA blood samples were received from 73 Australian cattle herds, as cases suspected to be infected with Theileria orientalis. Beef cattle were predominantly affected, with Angus and Angus-crossbred cattle representing 48% of smear positive samples examined. DNA extracts were tested in conventional polymerase chain reaction (PCR) assays for genes encoding the p32, Ikeda, Chitose and Buffeli major piroplasm surface proteins (MPSP). PCR findings were compared with results of clinical pathology examinations of stained blood smears for parasitaemia and packed cell volume (PCV). PCR testing was much more sensitive than clinical pathology examinations in detecting T. orientalis infections, and concurrent testing of neat and diluted extracts gave significantly more PCR positive results than testing of neat extract alone. Significant associations and correlations were shown between PCR results of p32 and Ikeda assays with PCV levels indicative of anaemia, and with the level of parasitaemia estimated by smears. A high proportion of samples had concurrent Ikeda and Chitose infection, and significantly more clinical cases of theileriosis were associated with the Ikeda MPSP type as the sole infection, compared with sole infection with types Chitose or Buffeli. The findings indicate Ikeda type organisms were significantly associated with clinical parameters of theileriosis in cattle herds in eastern Australia, and that this type is most likely to be responsible for outbreaks of theileriosis experienced in affected Australian herds. In New South Wales, 11 of 14 regulatory districts yielded Ikeda positive samples, with five (Mid-Coast, Cumberland, Central North, Hume and Lachlan) containing 234/307 (76%) of the Ikeda positive samples.  相似文献

Declining population numbers coupled with the growing evidence of global change have focussed attention on the critically endangered riverine rabbit (Bunolagus monticularis) endemic to South Africa. The aim of this study is to develop a habitat model to aid in the identification of isolated populations, offer opportunities for re-introduction or introduction, and guide future conservation efforts by assessing the possible impacts of global change. We attempt a novel approach where plant species which afford the riverine rabbit cover from predation and its primary food sources are modelled utilising the same technique and are included as a predictor variable in the habitat model for both current and future projections of potential habitat. Inclusion of this proximal variable as well as riparian areas yields a more parsimonious habitat model than using climatic variables alone. Results suggest that unsurveyed suitable habitat east of Victoria West might harbour previously overlooked isolated populations or offer new opportunities for re-introductions. Future climatic conditions under the most severe general circulation model for the region (HADCM3) suggest that, on average, in excess of 96% of the current habitat could become unsuitable, mitigated only slightly by a possible 7% increase in range in adjacent upper catchment areas. Consideration of existing land transformation increases this range reduction by a further 1%. Given that ex situ captive breeding programmes have met with no success and that the bulk of future potential range lies well outside of the currently known and surveyed areas the current adaptation options of conservancy establishment and captive breeding need to be re-evaluated. Without positive human intervention the future of the critically endangered riverine rabbit under conditions of global change seems certain.  相似文献

Apples were sampled directly from orchard trees at 96, 45, and 21 days postapplication with one of three organophosphate insecticides (azinphos methyl, phosalone, or phosmet, respectively). Individual apples were prepared for analysis following one of three postharvest preparations: no preparation, rinsed with deionized water for 10-15 s, or rinsed and peeled. Azinphos methyl, phosalone, and phosmet concentrations ranged from below the level of detection to 5.26 ng/g, 94.7 to 5720 ng/g, and 0.011 to 663 ng/g in the apples that received no postharvest preparation, respectively. Although rinsed apples had lower maximum concentrations than observed in apples with no preparation, levels were not significantly lower. Concentrations of all three OP insecticides in apples that were rinsed followed by peeling, however, were much lower (below detection limits to 0.733 ng/g, azinphos methyl; 0.322-219 ng/g, phosalone; and below detection limits to 44.0 ng/g, phosmet) than observed in apples that had been rinsed alone. Rinsing and peeling of apples resulted in a 74.5-97.9% reduction in OP residues, while rinsing alone lowered mean concentrations by 13.5-28.7% relative to apples that received no postharvest preparation.  相似文献

Lactococcus lactis is a food-grade microorganism of major commercial importance. Antifreeze protein is a potent cryogenic protection agent for the cryopreservation of food and pharmaceutical materials. In this study, extracellular expression of a novel recombinant type I antifreeze protein analogue (rAFP) in L. lactis was optimized. An efficient SlpA promoter (P SlpA) was fused to various signal peptides (SPs) and propeptide sequences to examine the extracellular expression levels of rAFP. An efficient signal peptide, SP sacB, fused to prosequence AE, enabled higher extracellular rAFP production; use of the SlpA terminator (Ter SlpA) was a further improvement. The extracellularly expressed rAFP successfully inhibited ice recrystallization and is thus potentially applicable for cryogenic preservation.  相似文献

Despite exhaustive literature describing drought stress effects on photosynthesis in Gossypium hirsutum, the sensitivity of photosynthetic electron flow to water deficit is heavily debated. To address this, G. hirsutum plants were grown at a field site near Camilla, GA under contrasting irrigation regimes, and pre‐dawn water potential (ΨPD), stomatal conductance (gs), net photosynthesis (PN), actual quantum yield of photosystem II (ΦPSII) and electron transport rate (ETR) were measured at multiple times during the 2012 growing season. ΨPD values ranged from ?0.3 to ?1.1 MPa. Stomatal conductance exhibited a strong (r2 = 0.697), sigmoidal response to ΨPD, where gs was ≤0.1 mol m?2 s?1 at ΨPD values ≤ ?0.86 MPa. Neither ΦPSII (r2 = 0.015) nor ETR (r2 = 0.010) was affected by ΨPD, despite exceptionally low ΨPD values (?1.1 MPa) causing a 71.7 % decline in PN relative to values predicted for well‐watered G. hirsutum leaves at ΨPD = ?0.3 MPa. Further, PN was strongly influenced by gs, whereas ETR and ΦPSII were not. We conclude that photosynthetic electron flow through photosystem II is insensitive to water deficit in field‐grown G. hirsutum.  相似文献

Field studies were carried out to determine whether ticks are being imported into the British Isles on migratory birds. During spring and autumn migration 2004, ticks were collected from ringed birds at 11 bird observatories and 3 inland Riparia riparia colonies. A total of 38 ticks of 4 species (Ixodes ricinus, I. frontalis, I. lividus, I. arboricola) were collected from 12 species of bird. Ticks were tested for viruses in the Flavivirus and Nairovirus genera, with no positives found. This data demonstrates that ticks are being imported into the British Isles on migratory birds with future work recommended to determine the quantity of ticks imported and to detect low prevalence pathogens.  相似文献
