Sweet potato (Ipomoea batatas [L.] Lam) is a versatile crop globally serving as food, feed and raw material for industries. Designed selection for higher yields and related traits is crucial to identify complementary sweet potato clones for breeding. The objective of this study was to determine phenotypic variation among diverse sweet potato collections with regard to yield, dry matter content (DMC) and sweet potato virus disease (SPVD) resistance and to identify suitable clones for breeding. A total of 144 sweet potato genotypes were evaluated at 2 sites in Tanzania using a 12?×?12 simple lattice design. Data collected included 10 quantitative and 17 qualitative agro-morphological traits and virus reaction. Results indicated differences among genotypes for most traits studied. The mean DMC was 36% with clones Zapallo and Ukerewe exhibiting the lowest and highest values of 29% and 45%, respectively. The mean storage root yield of clones was 5.1?t/ha with genotype Jewel expressing the highest yield of 11.3?t/ha. Genotypes Resisto and Mataya were early flowering at 40 and 50 days, respectively, while Ex-Mwanza and Kandoro did not flower at all. Fifty-eight per cent of the genotypes showed resistant reaction to SPVD, while 31% and 11% were moderately susceptible and susceptible to the disease, respectively. A positive correlation was observed for the number of roots and fresh root yield. Seven clones including Simama, Ukerewe, Mataya, Resisto, 03–03, Ex-Msimbi-1 and Gairo were selected for high storage root yield and related traits or SPVD resistance. Background
The Andes-Amazon basin of Peru and Bolivia is one of the most data-poor, biologically rich, and rapidly changing areas of the world. Conservation scientists agree that this area hosts extremely high endemism, perhaps the highest in the world, yet we know little about the geographic distributions of these species and ecosystems within country boundaries. To address this need, we have developed conservation data on endemic biodiversity (~800 species of birds, mammals, amphibians, and plants) and terrestrial ecological systems (~90; groups of vegetation communities resulting from the action of ecological processes, substrates, and/or environmental gradients) with which we conduct a fine scale conservation prioritization across the Amazon watershed of Peru and Bolivia. We modelled the geographic distributions of 435 endemic plants and all 347 endemic vertebrate species, from existing museum and herbaria specimens at a regional conservation practitioner's scale (1:250,000-1:1,000,000), based on the best available tools and geographic data. We mapped ecological systems, endemic species concentrations, and irreplaceable areas with respect to national level protected areas.Results
We found that sizes of endemic species distributions ranged widely (< 20 km2 to > 200,000 km2) across the study area. Bird and mammal endemic species richness was greatest within a narrow 2500-3000 m elevation band along the length of the Andes Mountains. Endemic amphibian richness was highest at 1000-1500 m elevation and concentrated in the southern half of the study area. Geographical distribution of plant endemism was highly taxon-dependent. Irreplaceable areas, defined as locations with the highest number of species with narrow ranges, overlapped slightly with areas of high endemism, yet generally exhibited unique patterns across the study area by species group. We found that many endemic species and ecological systems are lacking national-level protection; a third of endemic species have distributions completely outside of national protected areas. Protected areas cover only 20% of areas of high endemism and 20% of irreplaceable areas. Almost 40% of the 91 ecological systems are in serious need of protection (= < 2% of their ranges protected).Conclusions
We identify for the first time, areas of high endemic species concentrations and high irreplaceability that have only been roughly indicated in the past at the continental scale. We conclude that new complementary protected areas are needed to safeguard these endemics and ecosystems. An expansion in protected areas will be challenged by geographically isolated micro-endemics, varied endemic patterns among taxa, increasing deforestation, resource extraction, and changes in climate. The potent aroma compound rose oxide was quantified in several white wines by a headspace solid-phase microextration stable isotope dilution assay (HS-SPME-SIDA) and the enantiomeric ratios of the cis diastereomers were determined by enantioselective capillary GC. The most odor-active stereoisomer (23)-cis-rose oxide was detectable in all investigated white wines ranging from 0.2 to 12 microg/L. However, its contribution to the overall aroma in some white wine varieties can be neglected as indicated by a low odor activity value (OAV). The highest concentrations were found in Gewürztraminer wines, confirming the importance of rose oxide as a varietal aroma compound in this variety. Surprisingly, the enantiomeric ratio of cis-rose oxide in all investigated wines was substantially lower than in nonfermented musts and in some wines almost racemic cis-rose oxide was detected. Fermentation studies with a model must that contained deuterated water revealed that yeast is capable of reducing the precursor 3,7-dimethyl octa-2,5-dien-1,7-diol (geranyl diol I) yielding 3,7-dimethyl-5-octen-1,7-diol (citronellyl diol I) that gives rise to cis- and trans-rose oxide after acid catalyzed cylization. The deuterium labeling pattern of the resulting rose oxide stereoisomers and a clearly detectable kinetic isotope effect indicate that at least two different reductive pathways in yeast exist that yield cis-rose oxide with different enantiomeric ratios altering the genuine enantiomeric ratio in grape musts. The presence of (+)-cis-rose oxides in wines can therefore be attributed to the reductive yeast metabolism during fermentation. This observation corroborates recent findings that the modification of terpene derived varietal aroma is an integral part of yeast metabolism and not only a simple hydrolytical process. 相似文献

Bacterial speck caused byPseudomonas syringae pv.tomato is an emerging disease of tomato in Tanzania. Following reports of outbreaks of the disease in many locations in Tanzania,
56 isolates ofP. syringae pv.tomato were collected from four tomato- producing areas and characterized using pathogenicity assays on tomato, carbon source utilization
by the Biolog Microplate system, polymerase chain reaction and restriction fragment length polymorphism (RFLP) analysis. All
theP. syringae pv.tomato isolates produced bacterial speck symptoms on susceptible tomato (cv. ‘Tanya’) seedlings. Metabolic fingerprinting profiles
revealed diversity among the isolates, forming several clusters. Some geographic differentiation was observed in principal
component analysis, with isolates from Arusha region being more diverse than those from Iringa and Morogoro regions. The Biolog
system was efficient in the identification of the isolates to the species level, as 53 of the 56 (94.6%) isolates ofP. syringae pv.tomato were identified asPseudomonas syringae. However, only 23 isolates out of the 56 (41.1%) were identified asPseudomonas syringae pv.tomato. The results of this work indicate the existence ofP. syringae pv.tomato isolates in Tanzania that differ significantly from those used to create the Biolog database. RFLP analysis showed that the
isolates were highly conserved in theirhrpZ gene. The low level of genomic diversity within the pathogen in Tanzania shows that there is a possibility to use resistant
tomato varieties as part of an effective integrated bacterial speck management plan.

Organic dormancy, the inability to germinate under favorable conditions, is a common problem in many crop species and their wild relatives, leading to more variable emergence, plant density, and growth rates, thus increasing costs, and lowering yield. To overcome these problems, several different methods have been developed for various crop and model plants. However, in the emerging crop being bred from wild and semi-domesticated Silphium, no such method has been established thus far. The objective of this study was to identify a dormancy-breaking assay to increase the seed germination rate. Seeds of three different Silphium integrifolium Michx. genotypes were treated with five chemical and one cold treatment, and dried before or after the treatment at 27 °C or 40 °C. Untreated, dried seeds were taken as control. Seeds soaked for 24 h in a ethephon/potassium nitrate solution followed by a 72 h drying step at 40 °C showed an increase of germination to up to 90?±?2% compared to control seeds (3?±?0% and 5?±?1%). We identified the minimum time frame of cold stratification needed to enhance seed germination in Silphium, and found that cold stratification was nearly as good as the chemical treatment. Our results provide two alternate ways to treat S. integrifolium seeds for breaking organic dormancy, which will help to facilitate future research in the Silphium domestication community. The fact that our optimal treatments were similar to protocols developed for sunflower suggests that these methods may also be applicable to many related economically important Asteraceae species. 相似文献

Setaria viridis (L.) P. Beauv. and its domesticated form, S. italica (L.) P. Beauv., have been developed over the past few years as model systems for C4 photosynthesis and for the analysis of bioenergy traits. S. viridis is native to Eurasia, but is now a ubiquitous weed. An analysis of the population structure of a set of 232 S. viridis lines, mostly from North America but also comprising some accessions from around the world, using 11 SSR markers, showed that S. viridis populations in the US largely separate by latitude and/or climatic zone. S. viridis populations from the Northern US and Canada (north of 44°N) group with accessions from Western Europe, while populations in the Mid and Southern US predominantly group with accessions from Turkey and Iran. We hypothesize that S. viridis in the US was most likely introduced from Europe, and that introductions were competitive only in regions that had climatic conditions that were similar to those in the regions of origins. This hypothesis is supported by the fact that Canadian S. viridis lines were fast cycling and undersized when grown in the Mid-Western and Southern US compared to their morphology in their native environment. A comparison of the population structure obtained with 11 SSR markers and ~40,000 single nucleotide polymorphisms (SNPs) in a common set of S. viridis germplasm showed that both methods essentially yielded the same groupings, although admixture was identified at a higher frequency in the SNP analysis. Small numbers of SSR markers can thus be used effectively to discern the population structure in this inbreeding species. 相似文献
