The kinetics, depletion/repletion of zinc, and effects of dietary ligands/food matrices on (65)Zn uptake was studied in Caco-2 cells. The uptake of zinc showed a saturable and nonsaturable component, depending upon the media zinc concentrations. Intracellular depletion increased zinc uptake, whereas zinc loading did not. Phytic acid and histidine inhibited zinc uptake, while tannic acid, tartaric acid, arginine, and methionine increased zinc uptake. Tannic acid at a 1:50 molar ratio promoted zinc uptake from wheat- and rice-based food matrices. Further, Caco-2 cells responded similarly with zinc and iron uptake when fed Indian bread prepared from low- and high-extraction wheat flour, representing low and high phytate content. However, inclusion of tea extract or red grape juice as a source of polyphenols enhanced the uptake of zinc while decreasing that of iron. These results suggest that the Caco-2 cells predict the correct direction of response to dietary ligands even from complex foods. 相似文献

An investigation was initiated to examine the effects of nanoscale zinc oxide particles on plant growth and development. In view of the widespread cultivation of peanut in India and in other parts of the globe and in view of the potential influence of zinc on its growth, this plant was chosen as the model system. Peanut seeds were separately treated with different concentrations of nanoscale zinc oxide (ZnO) and chelated bulk zinc sulfate (ZnSO4) suspensions (a common zinc supplement), respectively and the effect this treatment had on seed germination, seedling vigor, plant growth, flowering, chlorophyll content, pod yield and root growth were studied. Treatment of nanoscale ZnO (25 nm mean particle size) at 1000 ppm concentration promoted both seed germination and seedling vigor and in turn showed early establishment in soil manifested by early flowering and higher leaf chlorophyll content. These particles proved effective in increasing stem and root growth. Pod yield per plant was 34% higher compared to chelated bulk ZnSO4. Consequently, a field experiment was conducted during Rabi seasons of 2008–2009 and 2009–2010 with the foliar application of nanoscale ZnO particles at 15 times lower dose compared to the chelated ZnSO4 recommended and we recorded 29.5% and 26.3% higher pod yield, respectively, compared to chelated ZnSO4. The inhibitory effect with higher nanoparticle concentration (2000 ppm) reveals the need for judicious usage of these particles in such applications. The 18s-5.8s-25s ribosomal gene (18s-25s rDNA) in higher plants is present in multiple copies, on different chromosomes, as
tandemly repeated units. Among the multiple BamHI sites that occur in the repeat unit, only the site in the middle of the 25s rRNA coding region is methylated in most cereals
and pulses. BamHI restriction enzyme analyses of the mungbean 18s-25s rDNA showed the presence of two populations. Nearly 50% of the 18s-25s
rDNA population had BamHI site situated in the internal transcribed spacer region (ITS-BamHI) resistant to cleavage by BamHI. The amplified ITS fragment was completely digestible by BamHI showing that partial cleavage by BamHI is not due to variation in the recognition sequence but most probably due to methylation. The complete cleavage of the ITS-BamHI site by BstYI that recognizes BamHI site but is insensitive to methylation confirms that the ITS-BamHI site is methylated. Methylation is probably due to the presence of a guanosine residue adjacent to the 3′ cytosine in the
recognition sequence.

reproductive events and influences other reproduction related aspects like spawning behavior and pheromonal action in fish.
Multiple forms of GnRH peptides have been reported across diverse vertebrate and invertebrate classes. Here we report the
partial seabream GnRH (sbGnRH) cDNA sequence cloned from the brain of Channa striatus (snake head murrel) a fresh water perciform with immense economic and medicinal value across Asiatic countries. sbGnRH mRNA
was found in brain, gill and ovary of mature murrel with possible implications to the effect of GnRH on pheromonal phenomena
and on reinitiation of oocyte meiosis. In keeping with the earlier reported role of GnRH in initiation of oocyte meiosis we
here present evidence from RT-PCR, ICC demonstrating an increase in the level of sbGnRH mRNA in ovary from pre-vitellogenic

Vitellogenin (Vtg) is a female specific glycophospholipoprotein which can be induced both in male and female with estradiol
and xeno-estrogens. The basic theme behind the purification of vitellogenin from the fish is to understand the evolutionary
relationship and for the purification and characterization of the Vtg receptor. The male catfish, Clarias gariepinus was administrated with estradiol over a period of time for the synthesis of Vtg and the serum was collected. The Vtg was
purified from the serum using a two step chromatographic technique. The serum was passed on to DEAE-ion exchange column and
the protein was eluted using a salt gradient. The fractions containing the Vtg were pooled and passed onto a gel permeation
chromatography column and the pure protein was obtained. The molecular weight is around 200 kDa on the SDS-PAGE and around
520 kDa on the native gel electrophoresis. 相似文献
